Abstract:A new method for synthesis of synthesis 6-hydroxy-3, 4-dihydro-2(lH) quinolinone was investigated with higher yield and simpler operation. The title compound was prepared starting from 3-chloropropionyl chloride, via cyclizing, nitration, reduction, diazotization and hydrolysis. The total yield is 67.7%, and the yield of each step is between 86.5% -99.6%. 6-Hydroxy-3, 4-dihydro-2(1H)quinolinone was characterized by MS and 1H NMR.

Abstract:The Winterfeldt oxidation (NaOH, DMF, air, rt) of substituted 1,2,3,4-tetrahydro-gamma-carbolines has been developed, which provides a convenient and efficient method for the synthesis of the corresponding dihydropyrrolo[3,2-b]quinolones in moderate to excellent yields (38-94%). The generality and substrate scope of this reaction are explored and a possible mechanism is proposed. The results imply that electron-withdrawing groups on N-2 of tetrahydro-gamma-carbolines and N-5-H are necessary. The synthesis of 5 or 7-substituted pyrrolo[3,2-b]quinolones in near quantitative yields was also achieved through deprotection and aromatization of N-1-Boc-dihydropyrrolo[3,2-b]quinolones.

Abstract:A direct and efficient synthesis of N-methyl-1,2,3,4-tetrahydroquinolines (MTHQs) is achieved through the one-pot reductive N-methylation of quinolines. The SnOx-decorated Pt/Al2O3 (Pt-SnOx/Al2O3) catalyst proves to be highly effective in the transformation from quinolines to MTHQs, using methanol as the exclusive source for hydrogen (H) and methyl (CH3), without the addition of additional bases or additives. Mechanistic and kinetic studies reveal a tandem reaction pathway: (1) the dehydrogenation of methanol, (2) reduction of quinolines to 1,2,3,4-tetrahydroquinolines (THQs), and (3) N-methylation of THQs to MTHQs, with the dehydrogenation of methanol identified as the rate-determining step. Characterization and catalytic performance results demonstrate that SnOx modifies the morphology and electronic properties of Pt-based metal active sites, which dramatically facilitates the cleavage of O-H and less reactive C-H bonds in methanol, and maintains a delicate balance between methanol dehydrogenation and reductive N-methylation of quinolines, thus obtaining a high reactivity for the total transformation.

Abstract:An iron-catalyzed redox condensation of 2-nitrobenzyl alcohols, formic acid, and alcohols has been developed, which affords substituted quinolines with carbon dioxide and water as the only side products. With the use of formic acid as a redox moderator to fill the electron gap of the global redox condensation process, the reaction goes smoothly with a smaller amount of alcohol in comparison to previous reports (i.e. 1.2 equiv versus 3.3-4 equiv). The reaction goes equally well when 2-nitrobenzyl methyl ether was used instead of 2-nitrobenzyl alcohol under otherwise identical conditions, shedding a new light on the study of this quinoline synthetic method.

Abstract:A novel, practical, highly efficient, and transition metal free nitrogen insertion reaction for the synthesis of 2,3-disubstituted quinazolinone derivatives was developed. Diverse functionalized 3-indolinone-2-carboxylates and nitrosoarenes with a wide range of substituted nitrosobenzenes, nitrosopyridines, dibenzofuranyl, or dibenzothienyl nitroso compounds worked smoothly to give 2,3-disubstituted quinazolinone derivatives in good to excellent yields (69-98%). A gram-scale reaction was achieved, and an afloqualone analogue was synthesized under the mild reaction conditions.

Abstract:3,4-Dihydroquinolones are fundamental motifs widely found in variety of natural products and drug molecules. Conventional methods suffer from disadvantages of harsh reaction conditions, inefficiency, as well as selectivity issues. Herein, we develop a practical and efficient platform for the synthesis of 3,4-dihydroquinolones through successive hydrolysis and transfer hydrogenation process. A wide range of 2-chloroquinolines and others were compatible with this catalytic system and converted into the corresponding products in moderate to excellent yields. The scale-up experiment and late-stage functionalization performance verified the practical use of this protocol.

Abstract:A mild, efficient and practical protocol for the preparation of 2-sulfonylquinolines through CS2/Et2NH-induced deoxygenative C2-H sulfonylation of quinoline N-oxides with readily available RSO2Cl was developed. The reaction proceeded well under transition-metal-free conditions and exhibited a wide substrate scope and functional group tolerance. The preliminary studies suggested that the nucleophilic sulfonyl sources were generated in situ via the reaction of CS2, Et2NH and sulfonyl chlorides.

Abstract:A mild, rapid and efficient method for copper-catalyzed nitration of quinolines at the C5 or C7 position was reported firstly by using sodium nitrite as the nitro source. A series of nitrated quinoline derivatives were achieved in moderate to good yields via remote C-H activation. The method overcomes some shortcomings reported in previous nitration strategies, such as employing expensive metal catalysts or using toxic and unstable nitrating agents. Furthermore, some of the products can be obtained through filtration which displays the operational simplicity of this methodology. Importantly, this protocol may provide a practical synthetic tool for the introduction of nitro groups into drug molecules.

Abstract:An efficient silver‐catalyzed δ‐regioselective unactivated C(sp3 )–H of free alcohols dehydrogen‐coupling with quinazolines for the synthesis of δ‐hydroxylalkyl substituted quinazolines in aqueous media has been developed. Various alcohols including 1o and 2o alcohols, reacted with quinazolines through 1,5‐hydrogen‐atom transfer strategies to give the corresponding 2‐ or 4‐ δ‐hydroxylalkyl substituted quinazolines with good to excellent yields under mild reaction conditions. Notably, several substituted quinazolines and quinazolinones could be selectively dehydrogenative‐coupled with divergent free alcohols. This protocol provides a platform to access divergent functionalizations of quinazolines and quinazolinones to meet the growing needs for screening.

Abstract:A novel and efficient method for the synthesis of 2‐alkylamino and 2‐dialkylamino derivatives of N‐oxides of 5‐, 6‐, 7‐ and 8‐nitroquinolines, which can potentially exhibit biological activity, as containing a pharmacophoric quinoline fragment, is described herein. The reactions of 5(6,7,8)‐nitroquinoline N‐oxides with various primary or secondary amines in the presence of potassium ferricyanide at in aqueous dioxane medium afforded the corresponding secondary and tertiary amines in moderate to excellent yields. This reaction is the first example of a Chichibabin reaction leading to the formation of 2‐alkylaminoquinoline derivatives at position 2 under mild conditions. A study of the antibacterial activity of some products was carried out.

Abstract:The exploration of green C–H oxidation for the rapid construction of functional molecules, remains a permanent goal in synthetic chemistry. Herein, a traceless N -nitroso enabled oxidative Heck and amidation cascade was realized, leading to quinolinones that feature green oxidation and great functional group tolerance. The expedient construction of quinolinone containing pharmaceuticals including Flucarbril, Clostamide and Aripiprazole was achieved in a concise and environmentally friendly manner.

Abstract:The switchable synthesis of 3-non, 3-mono, 3,3′-disubstituted 3,4-dihydroquinolin-2(1H)-ones was developed through a redox-neutral hydride-transfer/N-dealkylation/N-acylation strategy from o-aminobenzaldehyde with 4-hydroxycoumarin, and Meldrum's acid, respectively. The unprecedented strategy for the synthesis of 3,3′-highly functionalized 3,4-dihydroquinolin-2(1H)-one has been realized with the in situ utilization of the released HCHO via the o-QM involved Michael addition. Abstract:A new and expedient photocatalytic protocol for the construction of quinolin-2(1H)-ones via Markovnikov-type sulfonylation/6-endo-trig cyclization/selective C(O)-CF3 bond cleavage starting from N-alkyl-N-(2-ethynylphenyl)-2,2,2-trifluoroacetamides and sulfinic acids has been developed. It is as an unprecedented protocol for the preparation of 4-sulfonylquinoline-2(1H)-ones with high efficiency, mild reaction conditions, acceptable yields and a wide range of substrates.

Abstract:Quinolin-2(1H)-ones and their derivatives have attracted considerable attention because of their extensive applications in the field of pharmaceuticals, agricultural and dye chemistry. In contrast to the existed synthetic methods, herein we present the one-pot cascade reaction of 1,1-dimeoxyth-2-nitroethene and 2-aminobenzaldehydes which is catalyst-free and via eco-friendly H2O/AcOH solvent, achieving 3-nitro-quinolin-2(1H)-ones in moderate to excellent yields. A series of quinolines are obtained by the coupling reaction of substituted aminobenzyl alcohols with various secondary alcohols in the presence of bidentate terpyridine Mn(I) complex in good to excellent yields. Importantly, this investigation also provides a green and environmentally friendly protocol for constructing heterocyclic compounds. Abstract:An unexpected annulation among 2-aminobenzyl alcohols, benzaldehydes, and DMSO to quinolines has been disclosed. For the reported annulation between 2-aminobenzyl alcohols and benzaldehydes, the change of the solvent from toluene to DMSO led to the change of the product from the diheteroatomic cyclic benzoxazines to monoheteroatomic cyclic quinolines. This annulation can be used to synthesize regioselectively different substituted quinolines by the choice of different 2-amino alcohols, aldehydes, and sulfoxides as substrates. Interestingly, introducing substituent groups to the α-position of sulfoxides resulted in the interchange of the positions between benzaldehydes and sulfoxides in the product quinolines. On the basis of the control experiments and literatures, a plausible mechanism for this annulation was proposed.

Abstract:A Ni-catalyzed asymmetric hydrocyanation that enables the formation of 4-cyanotetrahydroquinolines in good yields with excellent enantioselectivities is presented herein. A variety of functional groups are well-tolerated, and a gram-scale reaction supports the synthetic potential of the transformation. Additionally, several crucial intermediates for pharmaceutically active agents, including a PGD2 receptor antagonist, are now accessible through asymmetric synthesis using this new protocol.

Abstract:A reduction of quinolines to synthetically versatile tetrahydroquinoline molecules with I2 and HBpin is described. In the presence of iodine (20 mol%) as a catalyst, reduction of quinolines and other N-heteroarenes proceeded readily with hydroboranes as the reducing reagents. The broad functional-group tolerance, good yields and mild reaction conditions imply high practical utility.
