Botulinum toxin type A for modulating calcitonin gene-related peptide contents in jugular plasma and brain stem of migraine model rats triggered by nitroglycerin

Abstract:AIM: To observe the change of calcitonin gene-related peptide (CGRP) content in jugular plasma,medulla oblongata and pons,following botulinum toxin type A (BTX-A) is injected subcutaneously into migraine model rats triggered by nitroglycerin,and analyze the mechanism of relieving pains. METHODS: Twenty adult female Sprague-Dawley rats of (180±40) g weight were divided randomly into control group (n=5) and migraine model group (n=15),which was subdivided into migraine model treated with 0.9 % NaCl group (M+saline),migraine model treated 5 U/kg BTX-A group (M +BTX-A 5 U) and migraine model treated 10 U/kg BTX-A group (M+BTX-A 10 U),with 5 rats in each. There was no treatment in control group The subcutaneous injection of 10 mg/kg nitroglycerin was administrated into frontal and temporal regions of rats in migraine model group for establishing the models. Then the treatments of BTX-A and saline were injected into frontal and temporal subcuticle of corresponding groups after 2 hours of migraine episode,once a day continuously for 6 days. The contents of CGRP in jugular plasma,medulla oblongata and pons were detected after migraine treatment by radioimmunoassay (RIA). RESULTS: 1The behavioral changes were more in nitroglycerin model group than in control group (1.48±0.26,35.77±2.30,P 1 0.05).2The content of CGRP in jugular plasma was increased significantly in M+saline group and control group than in M+BTX-A 5 U group and M+BTX-A 10 U group (55.31±14.02,57.15±14.58,51.81±13.90,93.84±13.62,P 1 0.05).3The content of CGRP in medulla oblongata and pons was increased significantly in M+saline group and control group than in M+BTX-A 5 U group and M+BTX-A 10 U group (6.91±2.21,7.28±2.24,6.43±2.27,11.54±0.41,P 1 0.05). CONCLUSION: BTX-A can relieve migraine symptoms by inhibiting CGRP releasing from trigeminovascular system and preventing meningeal sterile inflammatory response.
