Abstract:Background/Aims: Bufalin can induce apoptosis in certain human cancer cell lines, but bufalin has not yet been thoroughly evaluated in colorectal cancer cells. Cleavage and polyadenylation specific factor 4 (CPSF4) and human telomerase reverse transcriptase (hTERT) play important roles in colorectal cancer growth. The aim of this study was to investigate the roles and interactions of bufalin, CPSF4 and hTERT and the effects of bufalin in human colorectal cancer. Methods: We treated LoVo and SW620 cells with bufalin to investigate the effect of bufalin on proliferation, apoptosis and migration. We verified the relationship between CPSF4 and hTERT using pulldown assays, luciferase reporter assays and chromatin immunoprecipitation (ChIP) assays. Results: Bufalin inhibited the proliferation and migration of and induced apoptosis in LoVo and SW620 cells. We identified CPSF4 as an hTERT promoter-binding protein in colorectal cancer cells. Conclusion: Our study identified bufalin as a potential small molecule inhibitor for cancer therapy.

Abstract:Bufalin is a natural toxin with anti-leukemic properties. It induces cell differentiation and apoptosis, as well as increasing the sensitivity of leukemia cells to other chemotherapeutic agents. We investigated the biological effects and molecular mechanisms of bufalin triggered apoptosis in HL-60 cells by gene expression profiling. The broad transcriptional response to bufalin was consistent with bufalin’s action of regulating HL-60 cell proliferation and apoptosis, as well as its synergistic effect with other drugs. Further transcription factor ELISA experiments suggested that the transcription factors NFκB and AP-1 were activated to promote bufalin-induced HL-60 cell apoptosis. Our study provides new insights into the molecular mechanisms of bufalin, might prove to be beneficial in leukemia therapy.

Abstract:Introduction: Bufalin is a component of Chinese traditional medicine, Chansu, which is reported to induce cell death among various kinds of tumors. Apoptosis evasion is a common problem of cancer treatment. Materials and methods: The proliferation of U-87 and U-373 treated by bufalin combined with or without apoptosis inhibitor was detected by MTT assay. The protein levels related to apoptosis and necroptosis were measured by Western blotting. Immunoprecipitation (IP) was applied for monitoring the formation of necrosome. The gene knockdown by CRISPR/Cas9 was applied to determine the roles of the proteins in apoptosis and necroptosis. Results: In this study, we found that bufalin could induce apoptosis or necroptosis when U-87 and U-373 escaped from apoptosis. Bufalin triggered cell death by upregulating tumor necrosis factor (TNF) -α, TNF receptor 1 (TNFR1) and receptor-interacting protein 1 (RIPK1). Antagonizing cellular inhibitor of apoptosis 1 (cIAP1) and cIAP2 were also contributory. Caspase-8 activation led to apoptosis. When caspase-8 was functionally lost, necrosome consisted of RIPK1, receptor-interacting protein 3 (RIPK3) and mixed lineage kinase domain-like protein (MLKL) formed and necroptosis happened. The knockdown of above genes or the drug treatment confirmed the mechanism of bufalin-induced cell death. Cytotoxicity of bufalin to caspase-8 knockdown cell lines made control cell lines more sensitive to bufalin in their mixture. Discussion: The cytotoxicity of bufalin to U-87 and U-373 was by inducing apoptosis or necroptosis when they were sensitive to apoptosis or not. The results indicated that seeking for treatments that could induce apoptosis and necroptosis was a good solution for the tumor evasion of apoptosis.

Abstract:AIM:To investigate whether bufalin induces apoptosis of human pancreatic cancer cells via the JNK signaling pathway.METHODS:The cytostatic effect of bufalin on human pancreatic cancer cell line BxPC-3 was evaluated by MTT assay.The impact of bufalin on apoptosis and p-JNK expression was determined by flow cytometry (FCM) and Western blot,respectively.The mRNA expression level of survivin was detected by fluorescent quantitative PCR in BxPC-3 cells treated with bufalin in the absence or presence of a JNK inhibitor.RESULTS:Bufalin inhibited cell growth in a concentrationand time-dependent manner.BxPC-3 cells treated with 0.16,0.32,or 0.64mg/L of bufalin for 48 h had cell apoptotic rates of 19.36%±0.39%,40.69%±0.44% and 59.63%±1.14%,respectively,showing a significant difference among the three groups (P<0.01).After the JNK signaling pathway was blocked,cell apoptotic rate decreased significantly (P<0.01).After bufalin treatment,p-JNK expression was upregulated at 1 h and reached the peak at 2 h.The mRNA expression of survivin decreased obviously after treatment with 0.32mg/L of bufalin for 48 h,but increased significantly when the NK signaling pathway was blocked.CONCLUSION:Bufalin can induce apoptosis of human pancreatic cancer cells.The anti-pancreatic cancer activity of bufalin might involve down-regulation of survivin mRNA via the SAPK/JNK signal transduction pathway.

Abstract:Objective:To investigate the effect of Bufalin in inducing apoptosis and Survivin expression of human pancreatic cancer cell in nude mice.Methods:Human pancreatic cancer models were established in mice and 48 models were randomly divided into 4 groups:control(NS)group,5-fluorouracil(5-Fu)group,Bufalin(Bu)group and Bufalin/5-fluorouracil(Bu/5-Fu)group.The mice were injected with normal saline(NS)20 ml/kg,5-Fu 24 mg/mg,Bu 1 mg/kg and Bu 1 mg/kg plus 5-Fu 24 mg/mg respectively.Seven days after injection,the mice of each group were sacrificed and the volume of the tumor were compared among all groups.The apoptosis index was detected by terminal deoxynucleotidyl transferase-mediated dUTP nick-end-labeling(TUNEL);the expression of Survivin protein was analyzed by immunohistochemistry(S-P method);the expression of Survivin gene was assessed by reverse transcription-polymerase chain reaction(RT-PCR).Results:Compared with NS group,the volume of tumor was significantly smaller in other groups(P < 0.05),the apoptosis index was obviously higher(P < 0.01),the expression of Survivin protein was down-regulated significantly(P < 0.01).While the tumor volume of Bu/5-Fu group was smaller than that of Bu group and 5-Fu group(P < 0.05),the apoptosis index of Bu/5-Fu group was obviously higher than that of Bu group and 5-Fu group(P < 0.01),the expression of Survivin protein of Bu/5-Fu group were down-regulated significantly than that of Bu group and 5-Fu group(P < 0.01).Conclusion:Bufalin can result obvious growth inhibitory effects of human pancreatic cancer cell and induce apoptosis;the suppressed expression of Survivin may be one of the mechanisms of apoptosis induction,Bufalin/5-fluorouracil can augment the curative effects.

Abstract:Objective To investigate the effect of the bioconversion product of bufotalin from bufadienolide on the apoptosis of BGC-823 cells and to explore its mechanism.Methods BGC-823 cells were exposed to the compound and the cytotoxicity was measured with MTT assay.Hoechst 33258 staining and Annexin Ⅴstaining were adopted to detect apoptosis,and western blotting method was used for the assay of cracked caspase-3.Results Bufotalin showed a cytotoxicity against BGC-823 cells in a concentration-dependent manner,the cytotoxicity of 1 μmol was up to 50 %and above.Bufotalin promoted BGC-823 cell apoptosis and increased the level of activated caspase-3.Conclusion Bufotalin can induce the apoptosis of BGC-823 cells and its mechanism may be through caspase-3 pathway.

Abstract:Background: Glioma is a common primary central nervous system tumor, and there is still a lack of effective therapeutic drugs that can effectively improve the survival rate of patients in clinical. Bufalin has a good effect in the treatment of various tumors, but the mechanism of how it promotes the apoptosis of glioma cells is still unclear. The aim of this study was to investigate the drug target of bufalin acting on glioma cells, and to clarify the mechanism of apoptosis. Methods: Cell viability and proliferation were evaluated by cck-8 and colony formation assay. Then, the cell cycle and apoptosis, intracellular ion homeostasis, oxidative stress levels and mitochondrial damage were detected after bufalin treatment. DARTS-PAGE technology combined with LC-MS/MS were employed to explore the drug targets of bufalin on U251 cells. Molecular docking and western blot were used to further identify the potential targets. siRNA of Annexin A2 and a DRP1 protein inhibitor Mdivi-1 were also used to verify the target of bufalin.Results: Bufalin upregulated the expression of cytochrome C, cleaved caspase 3, p-Chk1 and p-p53 proteins to cause U251 cell apoptosis and cycle arrest in S phase. Bufalin can also induce oxidative stress in U251 cells, destroy intracellular ion homeostasis, and cause mitochondrial damage. The expression of mitochondrial separation/fusion-related proteins in U251 cells was abnormal, Annexin A2 and DRP1 protein were translocated from cytoplasm to mitochondria, and MFN2 protein was released from mitochondria into cytoplasm after bufalin treatment, which disrupted the mitochondrial division/fusion balance of U251 cells. Conclusions: Our research indicated that bufalin can cause Annexin A2 and DRP1 oligomerization to be located on the surface of mitochondria, and disrupt the mitochondrial division/fusion balance to induce U251 cell apoptosis.

Study on Proteomics of HeLa Cell Apoptosis in Bufalin-Induced Human Cervical Carcinoma

Abstract:OBJECTIVE:To seek possible effect targets of bufalin in HeLa cells by studying the impact of bufalin on cell protein expression profile after treatment on human cervical carcinoma cell lines HeLa.METHOD:Bufalin's ICs0was measured by MTr assay. The apoptosis of cells was observed by FCM (flow cytometry) and Hoechst 33342 staining assay. Differentiated expression protein spots were founded and identified using proteomic techniques, which could induce HeLa cell apoptosis.RESULT:Bufalin showed remarkable cytotoxic effect on HeLa cells. IC50 (154 +/- 21.5) nmol X L(-1) indicated the possibility of inducing cell apoptosis. The protein expression profile showed 11 differentiated expression protein spots. Among the 11 proteins, nudix-type motif 5, vimentin, hnRNP C1/hnRNP C2 variant, HNRPK, HNRPK isoform a variant (two spots are the same protein), heat shock protein 27, macrophage-capping protein, SELENBP1 protein were down-regulated, while ribosomal protein, large, P0 and S-adenosylmethionine synthetase 2 were up-regulated by bufalin treatment. They may be effect targets of bufalin in HeLa cells. Western blotting showed consistent results in heat shock protein 27, vimentin and HNRPK between expression after treatment with bufalin and two-dimensional electrophoresis.CONCLUSION:Bufa-Lin can induce apoptosis in human cervical carcinoma cells HeLa and the effect of bufalin may be related to the joint intervention with multiple protein targets.

Abstract:Bufalin has been shown to induce apoptosis in osteosarcoma cells; however, the underlying mechanism has not been elucidated. The purpose of the present study was to investigate whether mitochondria‑mediated signaling pathways trigger the process of apoptosis in the U‑2OS osteosarcoma cell line. Bufalin inhibited the proliferation and induced apoptosis in U‑2OS cells in a time- and dose‑dependent manner. Bufalin‑induced apoptosis was accompanied with a significant reduction of the mitochondrial membrane potential, release of mitochondrial cytochrome c into the cytosol, activation of caspase‑3, caspase‑9 and poly(adenosine diphosphate ribose) polymerase, as well as downregulation of B-cell lymphoma 2 (Bcl-2)/Bcl-2-associated X protein. Cyclosporin A, a specific inhibitor of the mitochondrial permeability transition pore, attenuated bufalin-induced apoptosis. In conclusion, the present study revealed that bufalin induced apoptosis in the U‑2OS human osteosarcoma cell line via triggering of the mitochondrial pathway.

Abstract:Bufalin is the active ingredient of the Chinese medicine Chan Su, and it has been reported that bufalin induces apoptosis in some human leukemia and solid cancer cell lines. The exact mechanism of bufalin-induced apoptosis is, however, still not clear. In this study, we demonstrated that bufalin inhibited the proliferation of gastric cancer MGC803 cells in a dose-dependent and time-dependent manner. At a low concentration (20 nmol/l), bufalin induced M-phase cell cycle arrest, whereas at a high concentration (80 nmol/l) it induced apoptosis in MGC803 cells. Bufalin increased the Bax/Bcl-2 ratio and activated caspase-3 during the apoptotic process of MGC803 cells. It should be noted that bufalin transiently activated the phosphatidylinositol 3-kinase (PI3K)/Akt signaling pathway and then inhibited it completely, and upregulated the Casitas B-lineage lymphoma (Cbl) family of ubiquitin ligases, upstream modulators of PI3K. A combination of bufalin and LY294002, a PI3K-specific inhibitor, enhanced apoptosis, but PD98059, an extracellular-regulated protein kinase-specific inhibitor, had no significant effect on bufalin-induced apoptosis. These results suggested that the PI3K/Akt pathway might play a key role in bufalin-induced apoptosis in gastric cancer MGC803 cells.

Abstract:Cinobufacini has been widely used at oncology clinics in China to treat many kinds of cancers and bufalin is one active compound of it, but the anti-cancer effect and the underlying mechanisms of bufalin on breast cancer cells are still unclear. Our study demonstrated that bufalin could effectively decrease the viability of MDA-MB-231 cells with IC50 values of 152.2 ± 8.0 nM for 24 h and 22.4 ± 2.3 nM for 48 h, respectively. Further study showed that bufalin could trigger MDA-MB-231 cells to undergo apoptosis with up-regulating protein expression of p-JNK/JNK, p53, p-ERK/ERK, Puma and down-regulating protein expression of Bcl-2, supporting the possible application of bufalin to breast cancer treatment.
Bufalin exerts antitumor effects in neuroblastoma via the induction of reactive oxygen species‑mediated apoptosis by targeting the electron transport chain

Abstract:The prognosis of high‑risk neuroblastoma remains poor. Clinical first‑line drugs for treating neuroblastoma have been developed over the previous half‑century; however, progress in the identification of new drugs with high efficiency is required. Bufalin, one of the major components of extracts obtained from the venom of the Chinese toad Bufo gargarizans, which is used to treat heart failure in Asian Pacific countries, has been reported to be a potential drug against multiple types of tumor; however, the detailed mechanisms underlying its antitumor activities remain unclear, largely due to lack of knowledge regarding its targets. In the present study, bufalin was revealed to exhibit potent antitumor effects against neuroblastoma, both in vitro and in vivo, using cell proliferation, colony formation, Transwell migration and flow cytometry assays, as well as a nude mouse subcutaneous xenograft model. Moreover, a chemically modified bufalin probe was designed to identify the potential targets of bufalin in neuroblastoma via chemical proteomics. With this strategy, it was revealed that the electron transport chain (ETC) on the inner membrane of mitochondria may contain potential targets for bufalin, and that bufalin‑induced mitochondrial‑dependent apoptosis may be caused by disruption of the ETC. Collectively, the present study suggests that bufalin may a promising drug for chemotherapy against neuroblastoma, and provides a foundation for further studies into the antitumor mechanisms of bufalin.

Abstract:Bufalin, a major cardiotonic compound of the traditional Chinese medicine Chanshu has been used for cancer treatment for several years. However, the molecular mechanisms of Bufalin‐induced autophagy in osteosarcoma (OS) is not fully understood. In the present study, it was shown that Bufalin induced crosstalk between apoptosis and autophagy, which resulted in OS cell death. Mechanistically, Bufalin induced autophagy by increased the ratio of microtubule‐associated protein 1A/1B‐light chain 3 (LC3)‐II/LC3‐I, and inducing apoptosis via the caspase‐dependent pathway. Inhibition of autophagy promoted Bufalin‐induced cell death. In contrast, suppression of apoptosis enhanced Bufalin‐induced autophagy. In addition, it was found that Bufalin activated the Ca2+/calmodulin‐dependent protein kinase β/AMPK/Beclin1 pathway, which resulted in induction of autophagy. These findings provide a mechanistic understanding of the means by which Bufalin mediates autophagy and apoptosis in OS cells.

Abstract:The aim of this study was to investigate the effects of bufalin on human nasopharyngeal carcinoma NPC-TW 076 cells in vitro. Bufalin is a cardiotonic steroid and a key active ingredient of the Chinese medicine ChanSu. The extracts of Chansu are used for various cancer treatments in China. In the present study, bufalin induced cell morphological changes, decreased total cell viability and induced G2/M phase arrest of cell cycle in NPC-TW 076 cells. Results also indicated that bufalin induced chromatin condensation (cell apoptosis) and DNA damage by DAPI staining and comet assay, respectively. The induced apoptotic cell death was further confirmed by annexin-V/PI staining assay. In addition, bufalin also increased ROS and Ca 2+ production and decreased the levels of ΔΨm . Furthermore, the alterations of ROS, ER stress and apoptosis associated protein expressions were investigated by Western blotting. Results demonstrated that bufalin increased the expressions of ROS associated proteins, including SOD (Cu/Zn), SOD2 (Mn) and GST but decreased that of catalase. Bufalin increased ER stress associated proteins (GRP78, IRE-1 α , IRE-1 β , caspase-4, ATF-6 α , Calpain 1, and GADD153). Bufalin increased the pro-apoptotic proteins Bax, and apoptotic associated proteins (cytochrome c, caspase-3, -8 and -9, AIF and Endo G) but reduced anti-apoptotic protein Bcl-2 in NPC-TW 076 cells. Furthermore, bufalin elevated the expressions of TRAIL-pathway associated proteins (TRAIL, DR4, DR5, and FADD). Based on these findings, we suggest bufalin induced apoptotic cell death via caspase-dependent, mitochondria-dependent and TRAIL pathways in human nasopharyngeal carcinoma NPC-TW 076 cells.

Abstract:Background and purpose:Cervical cancer remains the second leading cause of death in gynecologic malignancies partially because of resistance to chemotherapy. Bufalin, a component of the traditional Chinese medicine Chansu, has been widely used in cancer treatment in China. This study aimed to investigate the effects of bufalin on inhibiting the proliferation of ME180 and C33A and explore its possible mechanism.Methods:The cytostatic effects of bufalin on ME180 and C33A cells were evaluated by CCK8 assay (cell counting kit-8). Glucose levels in ME180 and C33A cells were measured using glucose assay kit. Then the alterations of GLUT1 (glucose transporter 1) and HK2 (hexokinase 2) gene expression were detected by quantitative real-time polymerase chain reaction (qRT-PCR). The expressions of proto-oncogene C-MYC and HIF1α (hypoxia-inducible factor 1α) were determined by Western blot.Results:According to the results of CCK-8, bufalin can significantly inhibit the proliferation of carcinoma cells ME180 and C33A (P=0.027,P=0.018). Test on glycometabolism indicated that glucose uptake in cells treated with bufalin decreased (P=0.034,P=0.036). Results from real-time PCR showed that the expression of glycometabolism related indicators GLUT1 (P=0.019) and HK2 (P=0.016) levels were signiifcantly down-regulated in bufalin treated group. Western blot showed that the expression of C-MYC and HIF1αin cells with bufalin treatment was down-regulated markedly.Conclusion:Bufalin can inhibit the proliferation of the cervical carcinoma cells ME180 and C33A through inhibition of their glucose metabolism.
Bufalin Induces Reactive Oxygen Species Dependent Bax Translocation and Apoptosis in ASTC-a-1 Cells

Abstract:Bufalin has been shown to induce cancer cell death through apoptotic pathways. However, the molecular mechanisms are not well understood. In this study, we used the confocal fluorescence microscopy (CFM) to monitor the spatio-temporal dynamics of reactive oxygen species (ROS) production, Bax translocation and caspase-3 activation during bufalin-induced apoptosis in living human lung adenocarcinoma (ASTC-a-1) cells. Bufalin induced ROS production and apoptotic cell death, demonstrated by Hoechst 33258 staining as well as flow cytometry analysis. Bax redistributed from cytosol to mitochondria from 12 to 48 h after bufalin treatment in living cells expressed with green fluorescent protein Bax. Treatment with the antioxidant N -acetyl-cysteine (NAC), a ROS scavenger, inhibited ROS generation and Bax translocation and led to a significant protection against bufalin-induced apoptosis. Our results also revealed that bufalin induced a prominent increase of caspase-3 activation blocked potently by NAC. Taken together, bufalin induced ROS-mediated Bax translocation, mitochondrial permeability transition and caspase-3 activation, implying that bufalin induced apoptosis via ROS-dependent mitochondrial death pathway in ASTC-a-1 cells.

Abstract:Objective: To explore the effect of extracellular signal-regulated kinase(ERK) on the apoptosis induced by bufalin in K562 cells.Methods:Apoptosis was determined by morphological method and flow cytometry.(Results):Bufalin of 0.5 μmol/L induced the apoptosis of K562 cells at 24 hours,and the action of bufalin was enhanced by ERK inhibitor PD98059 and reduced by 12-O-tetradecanoylphorbol-13-acetate.Conclusion:ERK negatively regulated the apoptosis induced by bufalin in K562 cells.

Abstract:It has been reported that there are multiple mechanisms by which bufalin could exert its antimetastatic effect. HIF-1 alpha has been reported to be involved in tumor migration and invasion by regulating EMT. However, it is not known whether bufalin could exert the antimetastatic effect by modulating HIF-1 alpha expression in hepatocellular carcinoma. In the present study, we aimed to evaluate the antimetastatic potential of bufalin in vivo and in vitro. Our results demonstrated that the liver/lung metastases were significantly reduced in bufalin-treated mice, as tested in the orthotopic transplanted and tail vein injection tumor models. Furthermore, the epithelial-tomesenchymal transition (EMT) was inhibited in bufalin-treated tumors, as reflected the upregulation of E-cadherin, and downregulation of N-cadherin, vimentin, Snail. Similar results were observed in SMMC7721 cells treated with bufalin. Moreover, the transforming growth factor-beta 1 (TGF-beta 1)-induced EMT was also abrogated by bufalin. Mechanistically, our study demonstrated that hypoxia-inducible factor-1 alpha (HIF-1 alpha) played an important role in the antimetastatic effect of bufalin in hepatocellular carcinoma. Importantly, HIF-1 alpha expression may be regulated through the inhibition of the PI3K/AKT/mTOR pathway. Taken together, our results suggest that bufalin suppresses hepatic tumor invasion and metastasis and that this process may be related to the PI3K/AKT/mTOR/HIF-1 alpha axis.
