Neuron apoptosis increases in morphine-dependent rats

Abstract:Objective To determine whether morphine has an effect on apoptosis of rat brain cells in vivo.Methods Twenty-four adult male Sprague-Dawley rats were randomly divided into morphine-dependent group,morphine-abstinent group and control group(n=8).Morphine intake was increased progressively in the rats of morphine-dependent and abstinent groups for 13 d to create morphine dependent models.Then intraperitoneal injection of 5 mg/kg naloxone was performed in the rats of abstinent group for 30 min after addiction.Normal saline was injected in the rats of control group at the same time.Apoptosis was calculated by the terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL) method.Immunohistochemistry was used to detect the expression of Caspase-3 protein.Results The score of abstinent symptom in morphine-dependent group is not statistically different from control group(P>0.05),while it was significantly lower than that in morphine-abstinent group(P<0.01),indicating that morphine-dependent rat models were successfully created.Neuron apoptosis and Caspase-3 expression in hippocampus in both morphine-dependent group and morphine-abstinent group were significantly higher than those in control group(P<0.01).But there were no significant differences in neuron apoptosis and Caspase-3 expression between morphine-dependent group and morphine-abstinent group(P>0.05).Conclusion Chronic morphine can promote abnormal neuronal apoptosis in rat brain,which might be related to opiate-induced nerve damage.(Shanghai Med J,2012,35:864-866)
