GANODERMA ATRUM POLYSACCHARIDE PROTECTS HUMAN UMBILICAL VEIN CELLS AGAINST INJURY INDUCED BY HIGH GULCOSE

Abstract:Objective The aim of this study was to examine the effect of Ganoderma atrum polysaccharide (PSG-1) on injury induced by high glucose and possible mechanisms involved in human umbilical vein cells (HUVECs).Methods Cultured HUVECs were randomly divided into three groups:Control group,High glucose (HG) group and High glucose+ PSG-1 (PSG-1+HG) group.Cell viability was messured by using MTT assay.The activity of superoxide dismutase (SOD) and malondialdehyde (MDA) contents in cells were measured by SOD and MDA assay kits.Intracellular reactive oxygen species (ROS) generation was detected by flow cytometry.Results Compare to Control group,cell viability and SOD activity were significantly decreased and ROS generation and MDA contents were significantly increased in HG group.Compare to HG group,PSG-1 increased cell viability and SOD activity and very significantly decreased ROS generation and MDA contents.Conclusion PSG-1 could protect HUVECs against high glucose-induced damage by attenuating ROS generation and increasing activity of SOD.
