Abstract:A method for rapid optical-electrochemical assessment of the pro- and antibiotic properties of pharmaceutical products is described, which provides for periodic (in our case, every 3 h) instrumented recording of changes in the efficiency of elastic light scattering, pH, and electrical conductivity of liquid nutrient media (sterile aqueous solutions pH 7.2 ± 0.2, containing 20 g/liter protein hydrolysate, 5 g/liter glucose, 10 g/liter sucrose, 1 g/liter lecithin, and 2 g/liter NaCl) incubated in the presence and absence of viable test microorganisms and test samples. We report here the results of a comparative analysis, performed using this method, of the effects of different concentrations (from 3% to 0.1 %, v/v) of essential oils produced industrially by steam distillation from six different types of plant raw materials used in the pharmaceutical industry on the dynamics of the vital activity of Escherichia coli , selected as a typical representative of the human microbiota. These studies confirmed that as compared with methods previously used for similar purposes, the method proposed here is faster, more objective, more informative, and less labor- and material-intensive in performing assessments of the effects of different samples of pharmaceutical products on the dynamics of the vital activity of microorganisms. Among the samples studied here, the most active antimicrobial properties were found with by essential oils obtained from celandine and angelica roots and sage leaves. The nature of the biological activity of the study materials was determined not only by the composition of the substances present within them, but also by their concentrations, whereby decreases in concentration lead to decreases in antimicrobial activity; the duration of the interaction of test samples with test organisms was also a factor, increases in duration leading to decreases in the biological activity of the samples. The results obtained here can be used in the quality control of existing and the development of new pharmaceutical products containing various plant substances.
