Abstract:The peroxisome proliferator-activated receptor-γ2 (PPAR-γ2) gene has been reported in the pathogeny of obesity. However, the results have been inconsistent. The purpose of this meta-analysis was to acquire a more accurate assessment of the association between PPAR-γ2 Pro12Ala polymorphism and obesity. PubMed, Wan Fang (Chinese) databases, Chinese Biomedical Medical databases, and Chinese National Knowledge Infrastructure were searched to identify eligible studies. Finally, 25 studies (6491 cases and 8242 controls) were enrolled in this meta-analysis. The effect summary odds ratio (OR) with 95 % confidence interval (CI) was applied. Random-effects or fixed-effects model was performed based on the heterogeneity. STATA 12.0 was applied for this meta-analysis. The combined results showed that PPAR-γ Pro12Ala polymorphism was associated with the obesity risk (Ala vs. Pro: OR = 1.55, 95 % CI 1.34–1.80; Pro/Ala vs. Pro/Pro: OR = 1.54, 95 % CI 1.31–1.82; Ala/Ala + Pro/Ala vs. Pro/Pro: OR = 1.61, 95 % CI 1.36–1.90). Subgroup analysis by ethnicity showed that there were significant associations between PPAR-γ Pro12Ala polymorphism and obesity risk in Caucasians, Asians, and Mixed population. Subgroup analysis by obesity’s cutoff points showed that the associations were found among the patients with the cutoff point of BMI ≥24 and BMI ≥30 but not among the patients with the cutoff point of BMI ≥95th percentile. These results suggested that PPAR-γ Pro12Ala polymorphism might be a risk factor for obesity susceptibility.

Abstract:Objective To study the association of peroxisome proliferator activated receptorγ2 (PPARγ2) gene polymorphism with type 2 diabetes mellitus.Methods PPARγ2 gene Pro12Ala polymorphism was examined using polymerase chain reaction and restriction fragment length polymorphism (PCR RFLP) in 101 couples of type 2 diabetes mellitus normal control.Results In control group, heterozygosity carriers had lower serum apolipoprotein B (P=0 006).The Pro12Ala polymorphism in PPARγ2 gene is not associated with type 2 diabetes mellitus(OR=1.48,95%CI=0.52～4.18,P=0.461).Conclusion PPARγ2 gene Pro12Ala polymorphism is associated with abnormal lipid metabolism; PPARγ2 gene may not play a critical role in the development of type 2 diabetes mellitus in Han of Beijing Chinese.
Uncoupling Protein 2 and Peroxisome Proliferator-Activated Receptor γ Gene Polymorphisms in Association with Diabetes Susceptibility in Chinese Han Population with Variant Glucose Tolerance

Abstract:Objective . To investigate the association of polymorphisms in uncoupling protein 2 (UCP2) and peroxisome proliferator-activated receptor (PPAR γ ) with glucolipid metabolism in Chinese Han population. Methods . Five hundred eighty-nine subjects were divided into normal glucose tolerance (NGT) group ( n = 198 ) and abnormal glucose tolerance group ( n = 358 ). HbA1c, blood lipid profile, plasma glucose, and insulin were determined. Insulin sensitivity (HOMA-IR and Matsuda index (ISI M )) and insulin secretion indexes (HOMA- β , early and total phase disposition index) were evaluated. Eight potential functional SNPs in UCP2 and 7 in PPAR γ were selected. SNPs were genotyped on Sequenom MassARRAY platform. Results . The GG genotype of rs2920502 in PPAR γ was associated with decreased risk of impaired glucose tolerance (G allele: OR: 0.818, 95%CI: 0.526–0.969, P = 0.042 ; GG: OR: 0.715, 95%CI: 0.527–0.97, P = 0.031 ). The TT genotype of rs3856806 in PPAR γ was associated with increased risk of impaired glucose tolerance (T allele: OR: 1.46, 95%CI: 1.055–2.017, P = 0.022 ; TT: OR: 1.58, 95%CI: 1.104–2.761, P = 0.032 ). The GG genotype of rs2920502 in PPAR γ had better blood glucose and increased insulin secretion and had lower HOMA-IR than GC/CC genotypes. Conclusion . It probably could prevent insulin resistance in early stage by classifying the genotype of rs649446 and rs7109266 in UCP2. The GG genotype of rs2920502 in PPAR γ had a decreased risk for diabetes. The TT genotype of rs3856806 in PPAR γ had an increased risk for diabetes.

PGC-1αGene Polymorphisms Are Associated with the Lipid Levels in Northern Han Chinese Population

Abstract:Objectives Peroxisome proliferator-activated receptor(PPAR)-γcoactivator 1α(PGC-1α)was the first member identified as a cofactor for the nuclear hormone receptor PPAR_γ.It plays a critical role in the control of energy thermogenesis,including glucose uptake in muscle,and hepatic gluconeogenesis in liver.In view of the close relationship between type 2 diabetes and hypertension in clinical elements and etiology,the present study focused on the association of polymorphisms in PGC-1αgene with lipid levels in diabetic patients with hypertension in northern Chinese population.Method We selected 427 cases with type 2 diabetes and hypertension and 549 age-and sex-matched normal controls.All participants were unrelated Han population from the northern area of China.All subjects have not taken medication to regulate lipids.The polymerase chain reaction-restriction fragment length polymorphism was used to genotype the polymorphisms of 1302G/A(rs2970847)and G482S(rs8192678)in the PGC-1αgene.Results In case group,the 1302G/A polymorphism was associated with the triglycerides(TG)level adjusted by age, gender and body mass index(P=0.0296 and 0.0147 in the additive and recessive model,respectively). Patients carried with 1302A allele have a lower low-density lipoprotein cholesterol(LDLc)level than homozygous GG in the dominant model(P=0.0412).For the G482S polymorphism,patients with GG genotype have a lower LDLc level than patients with 482S allele(P=0.0467).Compared with patients with 482S allele, patients with 482GG genotype had a lower level of total cholesterol,although not significantly(P= 0.0699).Among patients with 6 common diplotypes composed by these two polymorphisms there were borderline difference in the level of TG(P=0.0502).Patients with 1302AA-482GG had the lowest TG level which confirmed the results of single SNP analysis.No similar findings were in control group.Conclusion The 1302G/A and G482S polymorphisms were associated with the lipid phenotypes among patients with type 2 diabetes and hypertension.These results suggested that 1302A and 482G alleles might be protective factors of abnormal level of lipids in Northern Han Chinese population.

Abstract:AimsTo explore associations between polymorphisms of IGF2-related genes including H19, IGF2, IGF2BP2 and IGF2R and Metabolic syndrome (MetS) susceptibility in the Chinese Han population.Methods66 subjects with MetS and 257 control subjects were collected for inclusion in a case-control study. PCR-RFLP was used to investigate polymorphisms in the H19, IGF2, IGF2BP2 and IGF2R genes. Elisa was used to detect the serum IGF2 concentrations.ResultsFemales carrying the GG and AG genotypes of rs680 (IGF2) exhibited a lower risk of MetS, compared with those harboring AA (adjusted OR = 0.388, p = 0.027), while GG and AG genotypes were associated with lower fasting glucose and HbA1c. In males, the Waist-to-Hip Ratio (WHR) and the level of TG were significantly higher in GG and AG genotypes than in the AA genotype of rs680 in IGF2. Levels of HDL-c were lower in men with GG and AG genotypes compared with those carrying the AA genotype. Serum IGF2 concentrations did not change among different genotypes. Finally, multifactor dimensionality reduction (MDR) analysis identified interactions between four polymorphisms: rs3741279 (H19), rs680 (IGF2), rs1470579 (IGF2BP2) and rs629849 (IGF2R).ConclusionsOur study suggests that IGF2-related genes including H19, IGF2, IGF2BP2 and IGF2R genes may play pivotal roles in the development of MetS.
Association of Mitochondrial DNA Polymerase Γ Gene POLG1 Polymorphisms with Parkinsonism in Chinese Populations.

Abstract:Background: Mitochondrial DNA polymerase gamma (POLG1) mutations were associated with levodopa-responsive Parkinsonism. POLG1 gene contains a number of common nonsynonymous SNPs and intronic regulatory SNPs which may have functional consequences. It is of great interest to discover polymorphisms variants associated with Parkinson's disease (PD), both in isolation and in combination with specific SNPs.Materials and Methods: We conducted a case-control study and genotyped twenty SNPs and poly-Q polymorphisms of POLG1 gene including in 344 Chinese sporadic PD patients and 154 healthy controls. All the polymorphisms of POLG1 we found in this study were sequenced by PCR products with dye terminator methods using an ABI-3100 sequencer. Hardy-Weinberg equilibrium and linkage disequilibrium (LD) for association between twenty POLG1 SNPs and PD were calculated using the program Haploview.Principal Results: We provided evidence for strong association of four intronic SNPs of the POLG1 gene (new report: c. 2070-12T>A and rs2307439: c.2070-64G>A in intron 11, P = 0.00011, OR = 1.727; rs2302084: c. 3105-11T>C and rs2246900: c.3105-36A>G in intron 19, P = 0.00031, OR = 1.648) with PD. However, we did not identify any significant association between ten exonic SNPs of POLG1 and PD. Linkage disequilibrium analysis indicated that c.2070-12T>A and c.2070-64G>A could be parsed into one block as Haplotype 1 as well as c.3105-11T>C and c.3105-36A>G in Haplotype 2. In addition, case and control study on association of POLG1 CAG repeat (poly-Q) alleles with PD showed a significant association (P = 0.03, OR = 2.16) of the non-10/11Q variants with PD. Although intronic SNPs associated with PD didn't influence POLG1 mRNA alternative splicing, there was a strong association of c.2070-12T>A and c.2070-64G>A with decreased POLG1 mRNA level and protein levels.Conclusions: Our findings indicate that POLG1 may play a role in the pathogenesis of PD in Chinese populations.
Association between GLP-1R gene polymorphism and dyslipidemia in Chinese patients with type 2 diabetes mellitus: A case-control study

Abstract:Objective To evaluate the relationship between GLP-1R gene polymorphisms and type 2 diabetes mellitus with dyslipidemia and without dyslipidemia in China. Methods A total of 200 patients with Type 2 Diabetes Mellitus (T2DM) were included in this study, including 115 with dyslipidemia and 85 without dyslipidemia. We used Sanger double deoxygenation terminal assay and PCR-RFLP to detect genotype of the GLP-1R rs10305420 and rs3765467 loci. T-test was used to analyze the association between gene polymorphisms and lipid indicators. SHEsis online analysis software was used to analyze the linkage balance effect of loci, and SPSS 26 was used to calculate the gene interaction by dominant model. Results The genotype distribution of the two loci in the sample of this study was in accordance with Hardy-weinberg equilibrium. There were significant differences in the genotype distribution and allele frequency of rs3765467 between T2DM patients with and without dyslipidemia (GG 52.9%, GA + AA 47.1% vs . GG 69.6%, GA + AA 30.4%; P = 0.017). Under the dominant model, the effects of rs3765467 A allele and rs10305420 T allele on dyslipidemia had multiplicative interactions ( P = 0.016) and additive interactions (RERI = 0.403, 95% CI [-2.708 to 3.514]; AP = 0.376, 95% CI [-2.041, 2.793]). Meanwhile, HbA 1c levels in rs3765467 A allele carriers (GA + AA) were found to be significantly lower than those in patients with GG genotype ( P = 0.006). Conclusion The rs3765467 (G/A) variant is associated with the incidence of dyslipidemia, and G allele may be a risk factor for dyslipidemia.

Abstract:Aims: Genetic variations in the PI3K/AKT/mTOR signaling pathway may be associated with an increasing risk of obesity and diabetes. In this study, we aimed to test whether polymorphisms in the PIK3CA (catalytic subunit of PI3K), AKT1, AKT2, and FRAP1 (mTOR) genes were associated with the risk of type 2 diabetes mellitus (T2DM) among Chinese population.Methods: A case-control study was conducted and included 248 cases with T2DM and 101 controls. A total of 28 tagSNPs from the 4 genes were chosen based on HapMap datasets and these were genotyped using a MassARRAY Compact Analyzer.Results: Individuals carrying the rs2494746 CG/GG or rs2494738GA/GG genotype in AKT1 had a higher risk of T2DM, compared with those carrying homozygous variants (adjusted OR = 1.79, 95% CI: 1.05-3.05, P = 0.03 for rs2494746; adjusted OR = 1.58, 95% CI: 1.19-2.10, P = 0.02 for rs2494738). Furthermore, we found that haplotype GC in the AKT1 gene comprised rs2494738 and rs3803304, indicating a significant association with T2DM (OR = 1.08, 95% CI: 1.01-1.15, P = 0.03). Finally, generalized multifactor dimensionality reduction (GMDR) analysis indicated that the best interactive model included 3 polymorphisms: rs2494746 (AKT1), rs4802071 (AKT2), and rs4845856 (FRAP1).Conclusions: Our study suggests that PI3K/AKT/mTOR pathway genes may participate in the development of T2DM. Peroxisome Proliferator-Activated Receptor-γ Coactivator-1α Polymorphism is Not Associated with Essential Hypertension and Type 2 Diabetes Mellitus in Chinese Population

Abstract:To investigate whether variations in the peroxisome proliferator-activated receptor-gamma coactivator-1 alpha (PGC-1 alpha) are associated with essential hypertension and type 2 diabetes in a Chinese population. A case-control study design was applied in a Chinese population. Two single nucleotide polymorphisms (SNPs), +1302G > A and G482S, in the PGC-1 alpha gene were genotyped and compared between 494 unrelated Chinese subjects with essential hypertension and type 2 diabetes and 555 normal control subjects with the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. These two polymorphisms were in highly significant linkage disequilibrium with each other (p < 0.0001). The frequency of the 482S allele was 42.9% in the Chinese population, which was similar to the frequency in the Japanese population (43.7%), but much higher than those of Caucasian populations (30.8% to 38.1%). There were no associations of the G482S and +1302G > A polymorphisms and haplotype combinations with essential hypertension and type 2 diabetes. In addition, no associations were found between these two polymorphisms and blood pressure. In conclusion, these results indicated that these two variations in the PGC-1a gene might not contribute to the risk of hypertension and type 2 diabetes in the Chinese population studied here.
Lepr Gene Gln223arg Polymorphism And Type 2 Diabetes Mellitus: A Meta-Analysis Of 3,367 Subjects

Abstract:Background: The Leptin receptor (LEPR) Gln223Arg gene polymorphism has been associated with an increased susceptibility to Type 2 diabetes mellitus (T2DM). Results from various studies, however, are inconsistent.Objective and methods: To better elucidate the association of LEPR Gln223Arg gene polymorphism with T2DM in the Chinese population, a meta-analysis of 3,367 subjects from seven independent studies was performed. The pooled odds ratios (ORs) and corresponding 95% confidence intervals (95% CI) were evaluated by the fixed-effects model.Results: A significant relationship between LEPR Gln223Arg gene polymorphism and T2DM in the Chinese population was found under allele (OR: 1.432, 95% CI: 1.211-1.694, P=2.67x10(-5)), dominant (OR: 1.466, 95% CI: 1.215-1.769, P=6.33x10(-5)), recessive (OR: 0.539, 95% CI: 0.303-0.960, P=0.036), heterozygous (OR: 0.700, 95% CI: 0.577-0.849, P=3.06x10(-4)), and homozygous (OR: 0.472, 95% CI: 0.265-0.839, P=0.011) genetic models.Conclusions: LEPR Gln223Arg gene polymorphism was associated with an increased risk of T2DM in the Chinese population. Therefore, Chinese carriers of the G allele of LEPR Gln223Arg gene polymorphism may be more susceptible to T2DM than the general population.
An Association Study of Paraoxonase Gene G/A148 Polymorphism with Type 2 Diabtets Mellitus and Blood Lipids Level in Chinese Population

Abstract:Objective To test the role of paraoxonase (PON) 2 gene in the pathogenesis of type 2 diabetes mellitus in Chinese population. Methods The PON2 gene G/A148 polymorphism was detected by polymerase chain reaction (PCR) and appropriate restriction enzyme (PCR RFLP) in 132 couples of type 2 diabetes spouse based controls. Results The genotype frequency and allele frequency were not different between the case and the control group. Compared with non obesity subjects without G allele, the obesity subjects without G allele and the obesity subjects with G allele had an OR for type 2 diabetes of 2.04, 3.18, respectively. In control group, the levels of total and low density lipoprotein cholesterol and apolipoprotein B (ApoB) were higher in the GA/GG genotype than that in the AA genotype and the OR of G allele for hyperlipidemia was 2.48, P =0.05. Conclusion There were synergistic effects between the PON2 gene G allele and obesity on type 2 diabetes mellitus. PON2 gene G/A148 polymorphism was significantly associated with total cholesterol level in non diabetic Chinese population.
Association of Rs10830962 Polymorphism with Gestational Diabetes Mellitus Risk in a Chinese Population

Abstract:To date, only three polymorphisms (rs10830962, rs7754840 and rs1470579) are included in the genome-wide association study Catalog ( www.ebi.ac.uk/gwas ). However, the available evidence is limited in pregnant Chinese women. We aimed to explore the associations of three polymorphisms (rs10830962, rs7754840 and rs1470579) with GDM risk in a Chinese population. We conducted a case-control study (964 GDM cases and 1,021 controls) to evaluate the associations of these polymorphisms with GDM risk. A logistic regression model was used to calculate odds ratios (ORs) and their confidence intervals (CIs). After adjustment for age, prepregnancy BMI, parity, abnormal pregnancy history and family history of diabetes, the minor allele of rs10830962 (C > G) demonstrated a significant association with an increased risk of GDM (OR = 1.16, 95% CI = 1.02–1.31, P = 0.029 in the additive model). However, no significant association was observed between the other two polymorphisms and GDM. Subsequent functional annotation shows that rs10830962 is located in the regulatory elements of pancreatic islets, alters the binding affinity of motifs and regulates SNORA8 expression. Our findings demonstrate that rs10830962 is associated with an increased risk of GDM in the Chinese population. Further functional characterization is warranted to uncover the mechanism of the genotype-phenotype association.

Abstract:Objective To explore the relationship between two functional polymorphisms in exons of the PPARγ gene and the susceptibility to T2DM. MethodsPCRRFLP was performed to test the genotypes in 296 newlydiagnosed T2DM cases and 477 controls matched for age and sex. Results The rs3856806 polymorphism of PPARγ gene showed a significant difference between T2DM cases and controls. Compared with CC genotype, those carrying CT/TT genotype had a lower T2DM risk. In the stratified analysis, compared with CC genotype, those carrying CT/TT genotype had a significantly reduced risk in female，the elders (aged above 50 years)，hypertensive subjects and normal weight individuals with OR value of 0.33(95%CI: 0.14, 0.76), 0.45(95%CI: 0.23, 0.88), 059(95%CI: 0.36, 0.95) and 0.34(95%CI: 0.13, 0.94), respectively. There was no significant difference in genotype frequency of rs1801282 polymorphism between T2DM patients and controls. However, the obese individuals carrying CG(Pro/Ala) genotypes had a lower T2DM risk. Conclusions The polymorphisms of PPARγ gene might play a role in the susceptibility of T2DM in Han population of Jiangsu.
Relationship between type 2 diabetes mellitus and a novel polymorphism C698T in C5L2 in the Chinese Han population

Abstract:In a previous study, we reported a novel single nucleotide polymorphism (SNP) 698C>T (P233L) in the gene, C5L2. This gene has been demonstrated to encode a functional receptor of acylation-stimulating protein (ASP), a G-protein-coupled receptor (GPCR), that has been shown to influence insulin secretion in cultured pancreatic islet cells in vitro and is a stimulator of triglyceride synthesis and glucose transport in vivo. In this study, we evaluated the relationship between this novel C5L2 SNP and development of type 2 diabetes mellitus (T2DM) in the Chinese Han population. A case-control study examining Chinese Han T2DM patients (n = 554) and healthy controls (n = 648) was performed to investigate the role of the 698C>T (P233L) C5L2 polymorphism. Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis was used to determine expression of this SNP. Heterozygote carriers of the 698CT C5L2 genotype were more frequent among T2DM patients (13.5%) than controls (3.2%; P < 0.001). The frequency of 698CT heterozygote carriers was significantly higher in women (12.8%) than in male subjects (5.7%, P < 0.001). The odds ratio (OR) of T2DM for 698CT carriers was 4.675 [95% confidence interval (CI) 2.840-7.694]. After adjustment of confounding factors such as age, sex, smoking, drinking, hypertension, and triglyceride (TG), total cholesterol, high-density lipoprotein, and low-density lipoprotein levels, the difference remained significant (P < 0.001, OR 5.556, 95% CI 2.444-12.630). Furthermore, the diabetic 698CT carriers displayed an increase in their serum TG level. However, there were no significant differences observed in any of the parameters measured in the control group. We conclude that the 698CT genotype of C5L2 may be an influencing genetic factor for T2DM in the Chinese Han population. These findings also indicate that heterozygous expression of 698CT C5L2 may contribute to metabolic abnormalities.
GLP1R rs3765467 Polymorphism Is Associated with the Risk of Early Onset Type 2 Diabetes

Abstract:Objective: To investigate the relationship between glucagon-like peptide-1 receptor gene polymorphisms and susceptibility to early onset type 2 diabetes. Methods: Samples from 316 type 2 diabetes patients with early onset type 2 diabetes (n = 137) and late-onset type 2 diabetes (n = 179) and 145 nondiabetic individuals were analyzed. Multiplex PCR combined with resequencing Hi-Reseq technology was used to detect single nucleotide polymorphisms of the glucagon-like peptide-1 receptor gene, and the allele frequency, genotype distribution, and clinical parameters were analyzed between each diabetes subgroup and the control group. Results: Sixteen single nucleotide polymorphisms were identified in the exonic region of the glucagon-like peptide-1 receptor gene according to the minor allele frequency (MAF > 0.05) in the participants. Among these, the glucagon-like peptide-1 receptor rs3765467 (G⟶A) mutation was statistically associated with early onset type 2 diabetes. Compared with that of the GG carriers, carriers of genotype AA at rs3765467 had a decreased risk of early onset type 2 diabetes after adjusting for sex and body mass index. In the dominant model, the frequencies of the rs3765467 AA + GA genotype were significantly decreased in the early onset type 2 diabetes group, and carriers of genotype AA + GA at rs3765467 had a decreased risk of early onset type 2 diabetes after adjusting for sex and body mass index. Moreover, fasting C peptide levels were significantly higher in GA + AA genotype carriers than those in GG genotype carriers. Conclusion: The glucagon-like peptide 1 receptor rs3765467 polymorphism was significantly associated with age at type 2 diabetes diagnosis and thus may be used as a marker to screen and detect individuals at risk of developing early onset type 2 diabetes.
Association Between IGF2BP2 Polymorphisms and Type 2 Diabetes Mellitus: A Case–Control Study and Meta-Analysis

Abstract:Background: Genome-wide association studies (GWAS) found that IGF2BP2 rs4402960 and rs1470579 polymorphisms were associated with type 2 diabetes mellitus (T2DM) risk. Many studies have replicated this association, but yielded inconsistent results. Materials and Methods: A case-control study consisting of 461 T2DM patients and 434 health controls was conducted to detect the genetic susceptibility of IGF2BP2 in a northern Han Chinese population. A meta-analysis was to evaluate the association more precisely in Asians. Results: In the case-control study, the carriers of TT genotype at rs4402960 had a higher T2DM risk than the G carriers (TG + GG) (adjusted odd ratio (AOR) = 1.962, 95% confidence interval (95% CI) = 1.065–3.612, p = 0.031]; CC carriers at rs1470579 were more susceptible to T2DM than A carriers (CA + AA) (AOR = 2.014, 95% CI = 1.114–3.642, p = 0.021). The meta-analysis containing 36 studies demonstrated that the two polymorphisms were associated with T2DM under the allele comparison, genetic models of dominant and recessive in Asians (p < 0.05). The rs4402960 polymorphisms were significantly associated with the T2DM risk after stratification by diagnostic criterion, size of sample and average age and BMI of cases, while there’re no consistent results for rs1470579. Conclusions: Our data suggests that IGF2BP2 polymorphisms are associated with T2DM in Asian populations.

Abstract:Objective To investigate the relationship between glucose-dependent insulinotropic polypeptide receptor (GIPR) gene polymorphisms and the risk of cardiovascular disease in Chinese Han type 2 diabetic patients (T2DM ). Methods Of total 666 T2DM patients ,390 with coronary artery disease (CAD) as cases and 276 without CAD as controls were recruited. We selected and genotyped four haplotype-tagging single nucleotide polymorphisms of GIPR gene (rs1800437 ,rs11671664 ,rs4803846 and rs12709891). The four tag-SNPs were selected from CHB data in HapMap Phase Ⅱ database (r2 <0.8 and MAF≥0.05 ,R#27). The PCR-RFLP assay was adopted to genotype haplotype-tagging single nucleotide polymorphisms (SNPs) in GIPR gene. Allele and genotype frequencies were compared between the CAD-positive and controls in all SNPs. Results (1) There was no statistical difference in the distribution of allele frequencies between the cases and controls in GIPR gene (rs1800437 ,79.8% vs 82.5% ;rs11671664 , 62.6% vs 59.3% ;rs4803846 ,89.0% vs 89.3% ;rs12709891 ,48.8% vs 50.4% ;respectively ,P>0.05) . (2) There were no significant differences in the distribution of GIPR rs1800437+ rs4803846 haplotype between two groups (GG , 69.2% vs 72.0% ;GC , 19.8% vs 17.3% ;AG , 10.6% vs 10.5% ;respectively ,P>0.05) . Conclusion GIPR gene mutation may not be a decisive factor of coronary heart disease in type 2 diabetic population in China .
Association of Single Nucleotide Polymorphism in ATP-Binding Cassette Transporter 1 Gene with Type 2 Diabetes Mellitus and Obesity

Abstract:Objective To study the association of ATP-binding cassette transporter 1(ABCA1) gene polymorphism with type 2 diabetes and obesity in Chinese Han population living in Shanghai. Methods Four hundred and forty patients with type 2 diabetes mellitus and 315 normal control subjects were enrolled in the study.Diabetic patients were further divided into normal weight patients(n=188,BMI25 kg/m~(2)) and obese patients(n=252,BMI25 kg/m~(2)).A single nucleotide polymorphism(SNP) rs2020927 was chosen and allele-specific real-time PCR method was employed to identify the loci genotype in all the subjects. Results The distribution of genotype frequency of the loci rs2020927 of ABAC1 was significantly different between the groups of type 2 diabetes and normal controls(P=0.001).The frequency of AA,AG,GG genotype was 43.8%,35.9% and 20.3%,respectively in the control group,and 33%,53.2% and 13.8%,respectively in the normal-weight diabetic group,and 40.9%,47.2% and 11.9%, respectively in the obese diabetic group.The frequency of genotype GG was significantly diffe-(rent) between the control group and obese diabetic group(P=0.009),while no significant difference was found between the control group and normal-weight diabetic group(P0.05). Conclusion The SNP rs2020927 of ABCA1 gene may be associated with type 2 diabetes mellitus and obesity in Chinese Han population(living in Shanghai.)

Abstract:Aim: To investigate the polymorphism of rs2304796 and rs894160 loci of PLIN gene in patients with type 2 diabetes from Henan Han population. Methods: A total of 1 274 adults( 161 type 2 diabeties and 1 113 controls) were recruited. PLIN rs2304796 polymorphism was genotyped by PCR-RFLP and PLIN rs894160 was genotyped by Real-time PCR. The software Unphased 3. 0. 7 was performed to construct the haplotypes of every individual. Association between PLIN polymorphism and type 2 diabetes was estimated by logistic regression model adjusted for covariates. Results: Genotype and allele frequency of rs2304796 between the 2 populations had statistical difference( χ2= 6. 018,P = 0. 049,χ2= 5. 916,P = 0. 015). There were no statistical differences in rs894160 genotype and allele frequency between type 2 diabeties and controls( χ2= 0. 680,0. 295,P > 0. 05). Data from logistic analysis revealed that( CT + TT) of PLIN gene rs2304796 polymorphism decreased the risk of type 2 diabetes after adjusting obesity and blood liqid indexes[OR( 95% CI) = 0. 670( 0. 475- 0. 944) ]. Conclusion: The SNP( CT + TT) in rs2304796 of PLIN gene might be a protective factor for type 2 diabetes.

Abstract:BACKGROUND:This study was to investigate the relationship between SLC22A1 and SLC22A4 gene polymorphisms and genetic susceptibility to type 2 diabetes in Chinese Han population. METHODS:The research group comprised 110 Chinese Han patients with type 2 diabetes, and the control group included 110 healthy volunteers. The polymorphisms of SLC22A1 gene rs628031 and rs2282143 loci and SLC22A4 gene rs2073838 and rs272893 loci were detected in the two groups. RESULTS:Statistically significant differences were identified in the genotype distributions of SLC22A1 gene rs628031 and rs2282143 loci between the two groups (p < 0.05). The A allele frequency of SLC22A1 gene rs628031 locus and the T allele frequency of rs2282143 locus were higher in the research group than in the control group (p < 0.05). The genotype distributions of rs272893 locus showed a significant difference between the two groups (p < 0.05), but not SLC22A4 gene rs2073838 locus (p > 0.05). CONCLUSIONS:The polymorphisms of SLC22A1 gene rs628031 and rs2282143 loci and SLC22A4 gene rs272893 locus of patients with type 2 diabetes indicated a significant difference between the two groups, suggesting that these genetic locus mutations increase the risk of type 2 diabetes in Chinese Han patients.
