Abstract:Hyaluronic acid (HA), which is a highly versatile glycosaminoglycan, is widely applied across the fields of food, cosmetics, and pharmaceuticals. It is primary produced through Streptococcus fermentation, but the product presents inherent challenges concerning consistency and potential pathogenicity. However, recent strides in molecular biology have paved the way for genetic engineering, which facilitates the creation of high-yield, nonpathogenic strains adept at synthesizing HA with specific molecular weights. This comprehensive review extensively explores the molecular biology underpinning pivotal HA synthase genes, which elucidates the intricate mechanisms governing HA synthesis. Moreover, it delineates various strategies employed in engineering HA-producing strains.

Study on Screening and Mutation of High Hyaluronic Acid-Producing Strain

Abstract:15 Strains were isolated from fresh buffalo nasal mucosa through screening.Five of the obtained strains were confirmed to be hyaluronic acid(HA)-producing strains by biochemical reaction.The strain B with the highest HA yield was cultured and its fermentation extract was characterized by IR,13CNMR and UV spectra.The results indicated that the strain B could produce HA.When strain B was treated by physical mutation of ultrasonic wave combined with UV-irradiation,the HA yield increased from 0.793 g·L-1 to 1.46 g·L-1.

Abstract:A hyaluronic acid (HA) pure culture screened from 140 cattle nasal mucosa was mutagenized with combination of UV and NTG. A mutant without haemolysis was screened from blood agar plate. The white purified HA was increased from 0.69 g/L to 0.94 g/L. The research laid a foundation for the next research of producer improvement.

Abstract:It was very critical for improving the quality and quantity of HA to select a high hyaluronic acid-producing stain.A selected mutant,designated as Streptococcus equi FJ-23 was selected from wild-type Streptococcus equi SH-0 by UV and NTG inducing mutagenesis,which did not produce streptolysin or hyaluronidase.The mutant could produce HA seven times as much as the original strain.Hyaluronic acid productivity and its relative molecular weight of the mutant kept stable after 6 generations of reproduction,which displayed perfect genetic stability.The research laid a good foundation for the further study on producing HA by fermentation.

Abstract:A strain of mutant Streptococcus equi JC-63, which can produce high molecular weight hyaluronic acid, was obtained from compound mutation breeding of Streptococcus equi JC-0 using 5-Bromouracil, UV rays and nitrosoguanidine as mutagenic agents. After inoculation for 25 generations, Streptococcus equi JC-63 showed inheritance stability, but not hemolysis. The culture conditions for producing HMW-HA were optimized in a 1-L shake flask through multi-factor orthogonal tests. Results showed the optimum ratio of fermentation medium comprised 50 g/L sucrose, 80 g/L corn steep liquor, 10 g/L glucose, 2 g/L MgSO4, 2 g/L NaHCO3, 2 g/L Na2HPO4, and 0.1 g/L UTP. Under these fermentation conditions, the molecular weight of the produced HA was 2.76 × 106 Da, and HA yield reached 4.12 g/L.

Abstract:Hyaluronic acid is a linear high molecular weight mucopolysaccharide with its molecular weight determining its rheological properties,physiological role and applications.Traditional studies have made significant efforts to improve the yield of hyaluronic acid by optimizing the fermentation.In recent years,the research focus has gradually shifted to how to improve the molecular weight of hyaluronic acid products.At present,the high molecular weight hyaluronic acid has good viscoelasticity,moisture retention and adhesion,and the application of the medicine is irreplaceable by the middle or low molecular weight hyaluronic acid.The mechanism of hyaluronic acid molecular weight regulation and the research progress of the production of high molecular weight hyaluronic acid by microbial fermentation were reviewed,and its development direction was prospected.

Abstract:After wild streptococcus zooepidemicus C55151 was mutated byN-methyl-N-nitro-N-nitrosogua-nidin(NTG), a mutanted strain(J18) capable of producing HA in a high yield was acquired, which can produce HA twice as much as the original strain. The optimum fermentation culture medium was studied by an orthogonal method, which contained glucose 50 g/L, yeast extract paste 30 g/L, (MgSO_4) (0.5) g/L. It was found that SDS can improve the yield of HA obviously. The kinetics of the HA fermentation process of the original one was compared with that of the mutanted strain.

Abstract:<p>Owing to its outstanding water-retention ability, viscoelasticity, biocompatibility and non-immunogenicity, Hyaluronic acid (HA), a natural linear polymer alternating linked by D-glucuronic acid and N-acetylglucosamine, has been widely employed in cosmetic, medical and clinical applications. With the development of synthetic biology and bioprocessing optimization, HA production via microbial fermentation is an economical and sustainable alternative over traditional animal extraction methods. Indeed, recently <em>Streptococci</em> and other recombinant systems for HA synthesis has received increasing interests due to its technical advantages. This review summarizes the production of HA by microorganisms and demonstrates its synthesis mechanism, focusing on the current status in various production systems, as well as common synthetic biology strategies include driving more carbon flux into HA biosynthesis and regulating the molecular weight (MW), and finally discusses the major challenges and prospects.</p>

Abstract:Abstract Hyaluronic acid (HA) is a natural and linear polymer composed of repeating disaccharide units of β-1, 3- N -acetyl glucosamine and β-1, 4-glucuronic acid with a molecular weight up to 6 million Daltons. With excellent viscoelasticity, high moisture retention capacity, and high biocompatibility, HA finds a wide-range of applications in medicine, cosmetics, and nutraceuticals. Traditionally HA was extracted from rooster combs, and now it is mainly produced via streptococcal fermentation. Recently the production of HA via recombinant systems has received increasing interest due to the avoidance of potential toxins. This work summarizes the research history and current commercial market of HA, and then deeply analyzes the current state of microbial production of HA by Streptococcus zooepidemicus and recombinant systems, and finally discusses the challenges facing microbial HA production and proposes several research outlines to meet the challenges.

Abstract:Background: Background: Hyaluronic acid (HA), a natural polymer with wide applications in biomedicine and cosmetics, is mainly produced by Streptococcal fermentation at industrial scale. In the present study, chemical random mutagenesis was used for development of Streptococcus equisimilis group G mutant strains with high HA productivity.Methods: Methods: The optimum of the pH of culture condition and cultivation time for HA production by wild strain group G were assessed. At first, two rounds of mutation at different concentrations of NTG was used for mutagenesis. Then, the nonhemolytic and hyaluronidase-negative mutants were screened on the blood and HA agar. HA productivity and molecular weight were determined by carbazole assay, agarose gel electrophoresis and specific staining. Moreover, stability of the high producer mutants was evaluated within 10 generations.Results: Results: The results showed that the wild-type strain produced 1241 ± 2.1 µg/ml of HA at pH 5.5 and 4 hours of cultivation, while the screened mutants showed a 16.1-45.5% increase in HA production. Two mutant strains, named Gm2-120-21-3 (2470 ± 8.1 µg/ml) and Gm2-120-21-4 (2856 ± 4.2 µg/ml), indicated the highest titer and a consistent production. The molecular weight (Mw) of HA for the mutants was less than 160 kDa, considering as a low Mw HA.Conclusion: Conclusion: The mutant strains producing a low polydisperse, as well as low Mw of HA with high titer might be regarded as potential industrial strains for HA production after further safety investigations.

Abstract:Using Streptococcus equi SH-0 as original strain,a mutant strain named SH-109,which was stable in heredity and could produce hyaluronic acid(HA)with high molecular weight,was achieved by 5-bromouracil(5-BU),ultraviolet and N-mEthyl-N-nitro-nitrosoguanidinc(NTG)treatments. The production conditions of high molecular weight HA were optimized by the fermentation in 500 mL flask and 5 L fermentor.According to the orthogonal test results of multi-factors,and the optimal medium for HA production was composed of 50g/L sucrose,100g/L mixed nitrogcn sources, 10g/L giucosc,2.0g/L MgSO_4·7H_2O,2.0g/L Na_2HPO_4·12H_2O,1.0g/L NaHCO_3 and 0.1g/L UTP.R was also indicated that the conditions of pH 7.6 and agitation spcod 550 r/min favored the production of HA.Under the optimized culture conditions,HA with a molecular weight of 4.36×10~6 Da was accumulated in the culture broth of mutant SH-109.
Comprehensive review on biotechnological production of hyaluronic acid: status, innovation, market and applications

Abstract:The growing, existing demand for low-cost and high-quality hyaluronic acid (HA) needs an outlook of different possible production strategies from renewable resources with the reduced possibility of cross-infections. Recently, the possibility of producing HA from harmless microorganisms appeared, which offers the opportunity to make HA more economical, without raw material limitations, and environmentally friendly. HA production is mainly reported with Lancefield Streptococci A and C, particularly from S. equi and S. zooepidemicus. Various modes of fermentation such as batch, repeated batch, fed-batch, and continuous culture have been investigated to optimize HA production, particularly from S. zooepidemicus, obtaining a HA yield of 2.5 g L-1 - 7.0 g L-1. Among the different utilized DSP approaches of HA production, recovery with cold ethanol (4°C) and cetylpyridinium chloride is the ideal strategy for lab-scale HA production. On the industrial scale, besides using isopropanol, filtration (0.22 um), ultrafiltration (100 kDa), and activated carbon absorption are employed to obtain HA of low molecular weight and additional ultrafiltration to purify HA of higher MW. Even though mature technologies have already been developed for the industrial production of HA, the projections of increased sales volume and the expansion of application possibilities require new processes to obtain HA with higher productivity, purity, and specific molecular weights. In this review, we have put forth the progress of HA technological research by discussing the microbial biosynthetic aspects, fermentation and downstream strategies, industrial-scale scenarios of HA, and the prospects of HA production to meet the current and ongoing market demands.

Abstract:The unique biological and rheological properties make hyaluronic acid a sought-after material for medicine and cosmetology. Due to very high purity requirements for hyaluronic acid in medical applications, the profitability of streptococcal fermentation is reduced. Production of hyaluronic acid by recombinant systems is considered a promising alternative. Variations in combinations of expressed genes and fermentation conditions alter the yield and molecular weight of produced hyaluronic acid. This review is devoted to the current state of hyaluronic acid production by recombinant bacterial and fungal organisms.

Abstract:A high molecular weight hyaluronic acid-producing mutant, designated as Streptococcus equi SH-109 that did not produce streptolysin or hyaluronidase was selected from wild-type Streptococcus equi SH-0 by NTG treatment. The selected mutant could produce high molecular weight HA and had perfect genetic stability after reproduction. Several culture conditions were optimized in a 5-l fermentor for the production of hyaluronic acid. It was found that the optimal pH and agitation speed were 7.6 and 550rpm, respectively. Under the optimized culture condition, hyaluronic acid with a molecular weight of 4.0 x 106 was accumulated in the culture broth.

Abstract:Atmospheric and room temperature plasma (ARTP) is a new and efficient mutation breeding technique. In this study, we discuss a strategy combining ARTP mutagenesis and high-throughput screening to engineer Corynebacterium glutamicum towards high yield production of heterologous proteins. First, three target strains, MC2, MA8, and MA6, were screened from the mutant library with enhanced green fluorescent protein (EGFP) as the reporter protein, and their growth stability and the influence of heterologous protein production were verified. Second, genes encoding three high-value medicinal proteins (glycoprotein D, gD; endoxylanase, XynA; and variable domain of heavy chain of heavy-chain antibody, VHH) were expressed in the mutagenized strain, which confirmed its applicability for an increased biosynthesis of other heterologous proteins. During the large-scale fermentation of C. glutamicum for VHH production, the fermentation characteristics of the best mutant MA6 were verified. Compared to the original strain, the yield of VHH obtained with strain MA6 was increased by nearly 91 % to approximately 862 mg/L. Finally, through systematic genome analysis mutations in five genes were obtained. These genes code for putative proteases or are potentially related to the bacterial restriction repair systems. These findings will help to obtain optimized chassis cells and provide a direction for in-depth research on genetic targets that can increase protein production.
