Participation of NO-cGMP in Skeletal Muscle Mitochondrial Biogenesis:The Role of Endurance Training and L-Arginine Supplementation

Abstract:Objective This study was to determine whether endurance exercise and L-Arginine supplementation could promote the production of NO-cGMP in skeletal muscle and the signalization of NO in exercise-induced mitochondrial biogenesis in skeletal muscle. Methods Male Sprague-Dawley rats were divided into 4 groups: normal control group(NC);6-week training group(Ex);6-week L-Arg supplementing group(L-Arg);6-week L-Arg supplementing and training group(L-Arg+Ex).Rats were forced to run on the treadmill for 6 weeks,five days per week,90min per day.Drinking water for L-Arg group contained L-arginine and the average dose of L-arginine was 500 mg per kg body weight per day.Gastrocnemius and soleus muscles were collected and NO and cGMP concentration,mRNA transcription of NRF-1 and Tfam,mRNA and protein expressions of PGC-1α and COX IV were observed. Results As compared with the NC group,cGMP concentration in muscle increased,the protein contents of PGC-1α enhanced,and the levels of NRF-1 and Tfam mRNA,mitochondrial COX IV mRNA and protein expression were up-regulated in Ex group.As compared with the NC group,the levels of cGMP and NO in L-Arg+Ex group increased,and also the PGC-1α,NRF-1,Tfam and COX IV mRNA and protein contents of PGC-1α and COX IV were up-regulated significantly.Conclusion Our data suggested that NO-cGMP signaling pathway involved in endurance training-induced mitochondrial biogenesis in skeletal muscle.
