Cell Line Cross-Contamination: KB is Not an Oral Squamous Cell Carcinoma Cell Line.

Abstract:Dear Editor, Established cell lines are important research tools in the field of biomedical research, including cancer biology studies. Currently, a large number of cell lines have been established and are utilized in various studies. Unfortunately, because of the relatively high frequency of crosscontamination among cell lines, as well as a lack of cell identification and verification prior to each study, the credibility and the reproducibility of some research results are severely threatened (1, 2). According to recent studies, about 18–36% of cell lines used in various studies are incorrectly designated, and most of the problems encountered regarding cell line cross-contamination are related to HeLa cells (3, 4). HeLa, derived from a uterine cervical carcinoma, was the first established human cancer cell line. Because of the robustness and rapid growth rate of HeLa cells, other cell lines are easily overgrown and replaced by HeLa cells through the inadvertent introduction of HeLa cells into the cultures. So far, more than 10 cell lines have been confirmed to be contaminated with HeLa cells, including the liver cell line Chang liver, the atrial myoblast cell line Girardi heart, the larynx carcinoma cell line Hep-2, the embryonic intestine cell line INT407, the monocytic leukemia cell line J111, the embryonic lung epithelium cell line L132, the breast cancer cell line MT-1, the skin epithelium cell line NCTC2544, the amnion cell line WISH, and the conjunctiva-derived cell line Wong– Kilbourne (3, 5, 6). In the field of oral squamous cell carcinoma (OSCC) studies, the KB cell line was originally designed to be derived from an epidermal carcinoma of the mouth and has been regarded as an OSCC cell line. However, KB was found to be contaminated with HeLa cells at the time when it was established. Through the use of isoenzyme analysis, HeLa marker chromosomes, and DNA fingerprinting, it has been clearly demonstrated that the KB cell line is in fact a subline of HeLa and is not derived from an epidermal carcinoma of the mouth (7, 8). Recently, a simple, reliable, and reproducible test based on short tandem repeat (STR) analysis has been introduced as an international reference standard for identifying human cell lines (9, 10). With this improved test, several cell-culture repositories/cell banks (including ATCC, the American Type Culture Collection; DSMZ, the Deutsche Sammlung von Mikroorganismen und Zellkulturen; and JCRB, the Japanese Collection of Research Bioresources) have documented that the KB cell line is contaminated with HeLa cells and strongly suggest not using KB for studying OSCC.
