The Effect of Hyperbaric Oxygen on HIF-1α of Brain in Rats with Intracerebral Hemorrhage

Abstract:Objective:To observe the effects of hyperbaric oxygen(HBO) on the behavioral changes and the mechanism of angiogenesis in the intracerebral hemorrhage rats in order to further explain the pathological and physiological bases of HBO on cerebral hemorrhage.Methods: Eighty SD rat intracerebral hemorrhage models were randomly assigned to model group(n=40) and HBO group(n=40).The rats in HBO group were subjected to HBO treatment with the pressure of 0.20 MPa,once every day,60 min.On the postoperative day 4,7,14,21 and 28 day,8 rats in each group were randomly chosen respectively for observation and scoring of the behavioral changes.Three animals were randomly selected and killed by decapitation.The expression of HIF-1α around the hematoma region after intracerebral hemorrhage was detected by immunohistochemistry.Results: The intracerebral hemorrhage rats showed abnormal behavioral changes to varying degrees in both groups after operation.On the postoperative day 4,7 and 14,there was no significant difference in three assessments(the score of Langa,the score of Berderson and the balance beam walking test) between two groups.On the postoperative day 21 and 28,scores of three assessments in HBO group were significantly lower than in model group(P0.05).In HBO group the expression of HIF-1α around the hematoma region after intracerebral hemorrhage was higher than in model group at the same time point.The expression of HIF-1α in model group reached the peak at the 4th day,then showed a decreased tendency,and that in HBO group showed an increased tendency at the 4th day,then reduced at the 28th day.Conclusion: The expression of HIF-1α around the hematoma region after intracerebral hemorrhage was increased.HBO could increase the expression of HIF-1α,subsequently promoting the expression of VEGF,advancing the absorption of hematoma and angiogenesis,and improving the recovery of neurologica function of intracerebral hemorrhage rats.
