Analysis on Multiple SNPs of MPO to Screen the Genetic Marker Associated with Acute Leukemia in Chinese Han Population

Abstract:Myeloperoxidase (MPO) is an important metabolizing enzyme involved in oxidative stress response with some environmental carcinogens including benzene which is associated with bone marrow toxicity and leukemia after chronic exposure. A novel multiplex PCR technique based on adapter -ligation mediated allele specific amplification (ALM-ASA) is simple, inexpensive and of speciality for multiplex snp genotyping simultaneously. The purpose of this study was to explore the genetic markers associated with acute leukemia from multiple single nucleotide polymorphisms (SNPs) of MPO in Chinese Han population. A case-control study was conducted, 135 patients with adult acute leukemia as a case group and 187 cancer-free subjects as a control group. Nine SNPs in promoter and exons were selected,which were probable markers related to the gene function and the risk of leukemia. The multiplex PCR technique based on ALM-ASA was used to detect multiple SNPs genotypes .The accuracy of ALA-ALM was tested by sequence and PCR-RFLP genotyping analysis. No polymorphisms were found at -1468T>C, -1053T>C, codon90C>T, condon93C>A, condon115 T>C. The mutant alleles frequency of -1816 T>C, -586 T>C, -463G>A, condon53 G>T were 16.6%, 16.6%, 22.5%, 5.1%, respectively. A strong linkage disequilibrium was found among -1816, -586 and -463. The three haplotypes were constructed. SNPs and haplotypes analysis showed the frequency of the MPO -463G>A mutant genotypes (G/A+A/A)was significantly different between cases and controls (P=0.021). The results of sequence and PCR-RFLP were correspondent with that of ALM-ASA. It suggests that MPO G-463A mutant genotypes may be associated with a reduced risk of acute leukemia with an OR of 0.5756(95%CI: 0.3599-0.9208)due to its decreasing the activation of carcinogens. MPO G-463A variant may be a dependent protective genetic marker of acute leukemia in Chinese Han population. The ALM-ASA was a suitable technique for multiplex PCR analysis.
