Abstract:Tetrodotoxin (TTX) is a highly potent and widely distributed ion-channel marine neurotoxin; it has no specific antidote and poses a great risk to human health. Therefore, detecting and quantifying TTX to effectively implement prevention strategies is important for food safety. The development of novel and highly sensitive, highly specific, rapid, and simple techniques for trace TTX detection has attracted widespread attention. This review summarizes the latest advances in the detection and quantitative analysis of TTX, covering detection methods based on biological and cellular sensors, immunoassays and immunosensors, aptamers, and liquid chromatography-mass spectrometry. It further discusses the advantages and applications of various detection technologies developed for TTX and focuses on the frontier areas and development directions of TTX detection, providing relevant information for further investigations.

Abstract:Tetrodotoxin (TTX), which is found in various marine organisms, including pufferfish, shellfish, shrimp, crab, marine gastropods, and gobies, is an effective marine toxin and the cause of many seafood poisoning incidents. Owing to its toxicity and threat to public health, the development of simple, rapid, and efficient analytical methods to detect TTX in various food matrices has garnered increasing interest worldwide. Herein, we reviewed the structure and properties, origin and sources, toxicity and poisoning, and relevant legislative measures of TTX. Additionally, we have mainly reviewed the state-of-the-art progress of analytical methods for TTX detection in the past five years, such as bioassays, immunoassays, instrumental analysis, and biosensors, and summarized their advantages and limitations. Furthermore, this review provides an in-depth discussion of the most advanced biosensors, including cell-based biosensors, immunosensors, and aptasensors. Overall, this study provides useful insights into the future development and wide application of biosensors for TTX detection.

Abstract:Tetrodotoxin (TTX) is a neurotoxin that binds to sodium channels and blocks sodium conduction. Importantly, TTX has been increasingly detected in edible aquatic organisms. Because of this and the lack of specific antidotes, TTX poisoning is now a major threat to public health. However, it is of note that ultra-low dose TTX is an excellent analgesic with great medicinal value. These contradictory effects highlight the need for further research to elucidate the impacts and functional mechanisms of TTX. This review summarizes the latest research progress in relation to TTX sources, analogs, mechanisms of action, detection methods, poisoning symptoms, therapeutic options, biosynthesis pathways, and mechanisms of transport and accumulation in pufferfish. This review also provides a theoretical basis for reducing the poisoning risks associated with TTX and for establishing an effective system for its use and management to ensure the safety of fisheries and human health.

Abstract:Saxitoxin (STX) and brevetoxin (PbTX-2), which are produced by marine dinoflagellates, are highly-toxic marine toxins targeting separate sites of the α subunit of voltage-dependent sodium channels (VDSCs). In this work, a portable cardiomyocyte-based potential biosensor is designed for rapid detection of STX and PbTX-2. This potential biosensor is constructed by cardiomyocyte and microelectrode array (MEA) with a label-free and real-time wireless 8-channel recording system which can dynamically monitor the multisite electrical activity of cardiomyocyte network. The recording signal parameters, spike amplitude, firing rate and 50% of spike potential duration (SPD50) extracted from extracelluar field potential (EFP) signals of the potential biosensor is analyzed to quantitatively evaluate toxicological risk of STX and PbTX-2. Firing rate of biosensor signals presents high sensitivity to STX with the detection limit of 0.35ng/ml within 5min. SPD50 shows high sensitivity to PbTX-2 with the detection limit of 1.55ng/ml within 5min. Based on the multi-parameter analysis, cardiomyocyte-based potential biosensor will be a promising tool for rapid detection of these two toxins.

Abstract:With the rapid development of modern industry,the threat from marine toxins to human health has be?come increasingly serious,especially the paralytic biological poison. This paper presents a new method of cell-based potential sensor to detect shellfish toxicity. The cardiomyocyte combined with microelectrode array(MEA)by microelectronic processing technique which construct a novel cardiomyocyte potential sensor. The sensor can not on?ly real-time monitoring,but also could output good extracellular field potential signal with stability and consistency, which cardiomyocytes seeded on the MEA high-closely. By testing saxitoxin(STX)and analyzing the characteristics of signal parameters,the experimental results illustrate the STX have obvious inhibitory effect for electrical activity of cardiomyocyte. And the peak potential amplitude and frequency have obvious inhibition of concentration depen?dence,and presented the different concentration sensitivity. The LOD(limit of detection)of conventional standard laboratory toxicity detection on mouse bioassay method is 40μg/100 g,while the LOD and detection range of cardio?myocyte potential sensor is 1.004 ng/mL and 25 nmol/L~1 600 nmol/L,respectively. By comparative experiments, this method has great LOD and the detection is relatively simple and easy to fit standardization. With further im?provement,the method has good prospects for development on detection rapidly toxicity of marine toxins.

Abstract:Tetrodotoxin (TTX), found in various organisms including pufferfish, is an extremely potent marine toxin responsible for numerous food poisoning accidents. Due to its serious toxicity and public health threat, detecting TTX and its analogues in diverse food matrices with a simple, fast, efficient method has become a worldwide concern. This review summarizes the advances in sample preparation and analytical methods for the determination of TTX and its analogues, focusing on the latest development over the past five years. Current state-of-the-art technologies, such as solid-phase microextraction, online technology, novel injection technology, two-dimensional liquid chromatography, high-resolution mass spectrometry, newly developed lateral flow immunochromatographic strips, immunosensors, dual-mode aptasensors, and nanomaterials-based approaches, are thoroughly discussed. The advantages and limitations of different techniques, critical comments, and future perspectives are also proposed. This review is expected to provide rewarding insights to the future development and broad application of pretreatment and detection methods for TTX and its analogues.

Abstract:Outbreaks of paralytic snail poisoning have recently occurred in Asia, especially in China. The epidemiological characteristics of this disease from an outbreak in Zhoushan City, China, were recorded. Forty-two outbreaks of paralytic snail poisoning, involving 309 cases of illness, occurred from 1977 to 2001. Sixteen people (5.2%) died, 48 people (15.5%) required intubations, and 140 people (45.3%) required emergency hospital treatment as a result of these outbreaks. Outbreaks involved multiple marine snail species and occurred primarily during the summer (from June to August) on 11 islands with high population densities. Peak numbers of outbreaks and amounts of snail toxicity occurred from 1978 to 1979, from 1985 to 1987, and from 1992 to 1994. Toxicity varied depending on specimen, region, and season. The toxin involved was identified as tetrodotoxin. The data obtained in this study suggest that snails should not be eaten unless they are certified to be nontoxic.

Abstract:Tetrodotoxin (TTX) is a naturally occurring toxin that has been responsible for human intoxications and fatalities. Its usual route of toxicity is via the ingestion of contaminated puffer fish which are a culinary delicacy, especially in Japan. TTX was believed to be confined to regions of South East Asia, but recent studies have demonstrated that the toxin has spread to regions in the Pacific and the Mediterranean. There is no known antidote to TTX which is a powerful sodium channel inhibitor. This review aims to collect pertinent information available to date on TTX and its analogues with a special emphasis on the structure, aetiology, distribution, effects and the analytical methods employed for its detection.

Abstract:A method was established for determining tetrodotoxin (TTX) in bivalve mollusks by hydrophilic interaction liquid chromatography-triple quadrupole mass spectrometry (HILIC-MS/MS) using kasugamycin as the internal standard for quantification. Samples were subjected to ultrasonic extraction with methanol-water (1:1, v/v) containing 0.5% acetic acid, protein precipitation with acetonitrile, clean-up using a cation exchange solid phase extraction cartridge, elution with acetonitrile:water (1:1, v/v) containing 0.3% hydrochloric acid, neutralization with ammonia before HILIC-MS/MS analysis. The average recovery of the samples spiked at 3 levels ranged in 84.6%-98.1% with the relative standard deviation less than 7.2%. Using this method, the contamination of TTX in 429 bivalve mollusk samples collected in the local markets during 2018 and 2020 was investigated. The detection rates were 12.0-18.8%, following the order of oyster > mussel > clam > scallop. High contaminated oysters and mussels with TTX were found in July to August. Moreover, TTX analogs found in bivalve mollusks included 4-epiTTX, 5,6,11-trideoxyTTX, 4,9-anhydroTTX, and 5-deoxyTTX/11-deoxyTTX.

This drug has been widely used in the medical field, such as treating cancer related pain and severe arrhythmias. It is widely distributed and has been found in various high and low organisms such as aquatic and terrestrial organisms. But the source of the TTX and detoxification methods still need to be studied, so the study of the TTX is very significant. At the same time, TTX can block sodium ion channels, has obvious analgesic effect, and also has antiarrhythmia, abandon drug addiction and other effects. As a result, TTX has high medical and commercial value. This research will mainly summarize the pharmacological action, side effects of TTX and its underlying principle. In addition, it is expected that this study can provide new ideas for the study of new functions of TTX and the treatment of new diseases.

Abstract:An amperometric immunosensor based on a composite electrode of single-walled carbon nanotubes and ionic liquid n-octylpyridinum afluorophosphate (SWCNT-ILE) was developed for the determination of tetrodotoxin (TTX). Compared with the glassy carbon electrode (GCE), the electrode combined advantages of carbon nanotubes and ionic liquid, which exhibited the excellent antifouling ability of p-nitrophenol (PNP) so that it remarkably improved the stability of the p-nitrophenyl phosphate-based sensor. Combining the enzyme-linked immune sorbent assay (ELISA) by alkaline phosphatase (AP) and magnetic particles immobilized with antigens, a real-time assay of tetrodotoxin was developed by amperometric immunosensors. Under the optimium condition, the developed sensor demonstrated a linear range of tetrodotoxin from 2 to 45 ng/mL with a low detection limit of 5 ng/mL. Furthermore, the amperometric immunosensor was applied to determine TTX in real samples and could be used as an effective and sensitive sensor for direct detection of tetrodotoxin within 20 min.

Abstract:Abstract Tetrodotoxin (TTX) is a toxin mainly occurring naturally in contaminated puffer fish, which are a culinary delicacy in Japan. It is also detected in various marine organisms like globefish, starfish, sunfish, stars, frogs, crabs, snails, Australian blue-ringed octopuses, and bivalve molluscs. TTX is produced by marine bacteria that are consumed mainly by fish of the Tetraodontidae family and other aquatic animals. TTX poisoning through consuming marine snails has recently begun to occur over a wider geographical extent through Taiwan, China, and Europe. This neurotoxin causes food intoxication and poses an acute risk to public health. The aim of this review is to present the most recent information about TTX and its analogues with particular regard to toxicity, methods of analysis, and risk to humans of exposure.

Abstract:Tetrodotoxin (TTX), a potent neurotoxin, has recently been detected in marine fauna and waters in multiple locations outside its historical range and is projected to increase in both stability and bioavailability in acidifying oceans. We report the first biosensor designed to detect sub-ng mL-1 concentrations of TTX directly in natural seawater. The compact sensor chip employs a competitive enzyme strategy with a glucose oxidase-TTX conjugate that generates an amperometric response in the presence of a ferricyanide redox mediator. The biorecognition layer synthesis is confirmed by attenuated total reflection Fourier transform infrared spectroscopy. Cyclic voltammetry and electrochemical impedance spectroscopy are used to probe the stability and hydrophilicity of the biorecognition layer. We investigate the effects of nonspecific adsorption on sensor results and find that bovine serum albumen is an excellent blocking agent for preventing NSA when incubating with filtered seawater samples. The biosensor has a limit of detection of 0.053 ng mL-1 and a sensitivity of 87 nA ng-1 mL. Requiring no extraction or preconcentration steps with a time-to-results of under an hour, our biosensor provides a new tool for detecting TTX in natural waters, which can lead to more proactive monitoring and higher throughput testing.

Abstract:Tetridotoxin (TTX), Saxitoxin(STX) are important tool-drugs in clarifing the sodium channel structure. The parameters of electronic structure molecular geometry and molecular configuration of TTX, STX.and its derivatives were studied. It is shown that active guanidinium group and hemiketal ring in TTX and two guanidinium grouop in STX form two high polar Planar orthoriangle configuration with a positive net charge carbon centers. Three apices nitrogen or oxygen atoms of triangle posses strong negative charge respectively.Molecular electrostatic potential of some active site of TTX. STX and its derivatives were calculated and electrostatic potential contour maps were obtained. The fitting of TTX and STX molecular structres made by technology of molecular graphics shows very similar electronic and stereo characteristics both in TTX and STX. The acting mechanism and relationship between the molecular configurations and the toxicities of TTX,STX, and its derivatives were discuseed.

In order to protect the public safety and health, there have been more and more researches on the detection of marine biotoxins.
