Effects of Chronic Salt Loading on Oxidative Stress and Endothelial Dysfunction in Spontaneously Hypertensive Rats

Abstract:Background High salt intake increase the risk of cardiovascular disease,the mechanis by which may be mediated by oxidative stress and endothelial dysfunction.Objective To investigate the change of oxidative stress and endothelial dysfunction in spontaneously hypertensive rats(SHR) during long term chronic salt loading.Methods Forty-eight male SHR(5 week old) were randomly to receive high salt(4% NaCl,n=24) or normal salt(0.4% NaCl,n=24).Six rats were sacrificed every 4 weeks and the aortae were obtained to measure the eNOS expression by immunohistochemistry.Serum samples were collected to measure the concentration of serum super oxide dismutase(SOD),methane dicarboxylic aldehyde(MDA) and nitric oxide metabolites(NOx).Results Compared with normal salt group,the systolic blood pressure was significantly higher than that in normal salt group [4 weeks(158.0±13.6) vs(145.0±10.9);8 weeks(175.0±18.5) vs(149.0±8.1);12 weeks(178.0±14.8) vs(161.0±15.3);16 weeks(178.0±17.1) vs(159.0±17.1)mmHg,P0.05].Concomitantly,the levels of serum NOx(P0.01),MDA(P0.05) and eNOS(P0.01) were also higher.There were non-signifantly increasing tendency of the increment of oxidative stress products with aging.Conclusion Salt increase the risk of cardiovascular disease associaled with increasing oxidative stress and nitric oxide generation.
