Abstract:The nucleocapsid protein 409 bp gene of porcine reproductive and respiratory syndrome virus(PRRSV) was amplified by RT-PCR.The gene was cloned into prokaryotic exprssing pGEX-KG vector,the recombinant plasmid pGEX-KG-N was transformed into E.coli BL21.The results of SDS-PAGE and Western blot indicated that the nucleocapsid protein gene was expressed in a high level and the recombinant fusion protein,which was about 41 000,had immunologically reactive activity.The recombinant plasmid was induced to express large-scale GST-N fusion protein,the induced recombinant bacteria were lysed by freeze-thaw and sonication.With the GST-Protein Purtification Kit,the target protein was purified by affinity chromatography,there is only one purpose bands,purity above 90%.Using recombinant nucleocapsid protein as antigen,an indirect ELISA for the detection of antibdoies against PRRSV was developed,and the ELISA kits were maded.The optimal coating concentration of antigen was determined by checker board titration and amounted to 2.5 mg/L of GST-N perwell,the serum sample for testing was diluted to 1:100 for detection.The sealing buffer was 1%BSA,and the cutoff was determined to be 0.224.Comparing this indirect ELISA with HerdChek ELISA kit,its sensitivity and specificity were 90%,95%,respectively,the correlation between the two ELISA methods was 92%.Meanwhile,it was confirmed that there was no cross reaction between the antibodies against other porcine viruses:PPV,PRV,JEV,PCV,CSFV.The variant coefficient of ELISA test kit in a batch and between batches varied from 3.44% to 6.34% and from 5.04% to 7.64%,respectively.The ELISA kit was stable at-20℃ at least for one year.The detection of serum samples of 350 sows by the ELISA revealed that the positive rate of sow serum reached 88.6%.All data in this study demonstrated that the recombinant nucleocapsid protein-based indirect ELISA would be an useful tool for routine diagnostics,epidemiological surveys and outbreak investigations.
