Analysis of Co2+ Tolerance on Transgenic Tobacco Over-expressing A Ferritin Gene

Abstract:Objective: A Ferritin gene (NtFer1, GenBank accession No.ay083924) from Tobacco was directly cloned and transformed into the pBI121 binary vector. Recombinant plasmid was then transformed into Agrobactrium tumefaciens by electroporation. Tobacco (Nicotiana tobacum) primary transformants were produced by leaf disc transformation. Kanamycin tolerance analysis of T2 generation of transgenic tobacco showed that most of them had 3:1 separation ratios. PCR-Southern indicated that exogenous gene was integrated into the genome of transgenic tobacco plants, and and Northern hybridization indicated that exogenous ferritin gene correctly over-expressed. The physiologic characters of transgenic tobacco responsed to 150μmol·L-1 Co2+ tolerance was studied. The results showed that enhanced growth of transgenic tobacco was observed at the early development stages, so that plant height and fresh weights were significantly greater than those of non-transformant. And the activity of POD, SOD and concentration of chlorophyll were increased, whereas the accumulation of MDA were decreased in transgenic tobacco than in non-transformant.These results demonstrated that the exogenous ferritin over-expression could increase the ability of cleaning up reactive oxygen more than that of non-transformant, and showed a stronger tolerance to Co2+ stress.
