Effects of fibroblast growth factor on the proliferation of elastic chondrocyte and formation of matrix

Abstract:Aim: To observe the effects of fibroblast growth factor on the cartilage cell proliferation and synthetic extracellular matrix. Methods: The experiment was conducted at the Laboratory of Oral and Maxillofacial Surgery, Fourth Military Medical University and Center of Tissue Engineering, Southern Hospital, Southern Medical University between March 2003 and December 2004. The chondrocytes were isolated from rabbit auricular cartilage by enzyme digestion and mixed with 12 g/L alginate, with the cell concentration of 10 10 L -1, and the 125 mmol/L CaCl 2 solution was dropped by injection syringe to prepare the alginate beads of chondrocytes and cultured with DMEM medium of foetus ox serum with the volume fraction of 0.1 in vitro added with 0, 10, 50 and 100 μg/L basic fibroblast growth factor (bFGF) for 14 days to performed the biochemical analysis. The total amount of DNA and glycosaminoglycan and the content of glycosaminoglycan of unit DNA were observed in every group. Results: 1 The total amount of DNA:It was (3.4 4±0.23), (4.70±0.22), (6.60±0.35) and (6.82±0.33) μg, respectively in the 0, 10,50 and 100 μg/L fibroblast growth factor concentration group. 2 Total amount of glycosaminoglycan: It was (15.35±0.80), (20.63±0.72), (29.72±2.08) and (31.11±2.39) μg, respectively in the 0, 10, 50 and 100 μg/L fibroblast growth factor concentration group. 3 Content of glycosaminoglycan of unit DNA: It was (4.47±0.09), (4.39±0.13), (4.50±0.13) and (4.55±0.18) g/g, respectively in the 0, 10, 50 and 100 μg/L fibroblast growth factor concentration group. Conclusion: The fibroblast growth factor accelerates significantly the proliferation of chondrocytes, and the total content of synthetic glycosaminoglycan also enhances obviously, but the synthetic amount of glycosaminoglycan of unit DNA has insignificant changes. It is indicated that fibroblast growth factor affects the biological character of the chondrocytes.
