Abstract:Current imaging methods can provide little helpful information about the tumor response to effective treatments in a very early stage.However,apoptosis imaging offers an early,noninvasive and dynamic way to monitor the therapy-induced cellular changes,and contributes to knowing the early response in vivo.One of the most prominent characteristics ot apoptosis is the externalization of phosphatidylserine(PS),which resides predominantly in the inner leaflet of the plasma membrane in healthy cells.This feature has made PS a well target to image apoptosis.Radio labeled annexins have shown the ability to selectively combine with PS undergoing apoptosis in response to successful treatment in vivo.As abundant of apoptotic cancer cells can be shown 24 h after effective therapy,apoptosis imaging can help to provide instruction for individualized treatment in clinic.In this review we will provide an overview about the mechanism,applications,limitations,and future perspectives of the apoptosis imaging with annexins in tumor response to successful treatment in both preclinical experiments and clinical trials.
Preparation, in Vitro and in Vivo Evaluation of (99M)tc-Annexin B1: a Novel Radioligand for Apoptosis Imaging.

Abstract:To develop a radiopharmaceutical for apoptosis imaging, Annexin B1, a new Ca2+-dependent phosphatidylserine (PS)-binding protein, was directly radiolabeled with (99m)Tc. This procedure yields up to 96% of radiochemical purity and higher radiolabeling efficiency. The preparation has been found to be sufficiently stable in vitro. Binding assay with human activated platelets indicated that (99m)Tc-Annexin B1 retained its PS binding activity. Biodistribution in mice revealed that (99m)Tc-Annexin B1 rapidly cleared from the blood and predominantly accumulated in the kidney. The increase in hepatic uptake in anti-Fas antibody treated mice correlated to histologic evidence of fulminant hepatic apoptosis. These data suggest that (99m)Tc-Annexin B1 can be used as a novel radiotracer to detect apoptosis in vivo.

Abstract:Objective Annexin B1,a novel Ca2+-dependent phosphatidylserine(PS)-binding protein,has been shown to have a high affinity for PS exposed on the surface of apoptotic cells or activated platelets.The aim of this study was to develop and bioevaluate a Annexin B1 based PS-targeting radiotracer.Methods Annexin B1 was directly radiolabeled with technetium-99m using stannous chloride as reducing agent in the presence of citrate.Binding assay with human activated platelets was used to evaluate the biological activity of 99mTc-Annexin B1 in vitro.Biodistribution of this radioligand was studied in normal mice.The potential of 99mTc-Annexin B1 to detect apoptosis in vivo was evaluated in dexamethasone-inducecd thymus apoptotic mice and anti-Fas antibody induced liver apoptotic mice.Results The labeling procedure yielded 99mTc-Annexin B1 with high radiochemical purity and good in vitro stability.Platelets binding assay indicated that 99mTc-Annexin B1 retained its PS binding activity in vitro.99mTc-Annexin B1 is rapidly cleared from the blood and predominantly accumulates in the kidney.The uptake of mouse thymus increased markedly at 18 hours after the administration of dexamethasone compared with control mice.Radionuclide imaging showed a markedly increased uptake of 99mTc-Annexin B1 in the liver of anti-Fas antibody treated mice compared with the control.Conclusion These data suggest that 99mTc-Annexin B1 retain its in vitro and in vivo biological activities.It may therefore be useful as a novel radioligand for the noninvasive imaging of PS externalization associated with apoptosis.

Abstract:Real-time monitoring of cell apoptosis could provide valuable insights into early detection of therapy efficiency and evaluation of disease progression. In this work, we designed and synthesized a new live-cell-permeable, fluorescent light-up probe for real-time cell apoptosis imaging. The probe is comprised of a hydrophilic caspase-specific Asp-Glu-Val-Asp (DEVD) peptide and a hydrophobic tetraphenylethene (TPE) unit, a typical fluorogen with aggregation-induced emission characteristics. In aqueous solution, the probe is almost nonfluorescent but displays significant fluorescence enhancement in response to caspase-3/-7, which are activated in the apoptotic process and able to cleave the DEVD moieties. This fluorescence "turn-on" response is ascribed to aggregation of cleaved hydrophobic TPE residues, which restricts the intramolecular rotations of TPE phenyl rings and populates the radiative decay channels. The light-up nature of the probe allows real-time monitoring of caspase-3/-7 activities both in solutions and in living cells with a high signal-to-noise ratio. The probe provides a new opportunity to screen enzyme inhibitors and evaluate the apoptosis-associated drug efficacy.
Experimental study on tumor cell apoptosis imaging in vivo with 99Tcm-HYNIC-Annexin Ⅴ in mice tumor-bearing

Abstract:Objective To investigate in vivo the imaging and boidistribution characters of()~(99)Tc~m-HYNIC-Annexin Ⅴ as tumor cell apoptosis imaging agent in tumor-bearing mice.Methods Annexin Ⅴ was labeled with technetium-99m using a hydrazino-nicotinamide(HYNIC) method,then isolated and identified with high performance liquid chromatography(HPLC).One week after being inoculated with S-180 sarcoma in the right upper limbs,the mice were randomized to receive a single dose of cyclophosphamide(150 mg/kg intraperitoneally),and were injected through tail vein with()~(99)Tc~m-HYNIC-Annexin V 72 hours later.The images and radioactivity in tissues were obtained at 5 min,30 min,1 h,3 h and 6 h postinjenction of imaging agent,respectively.The data of radioactivity in tissues were analyzed by the statistic software of SPSS 12.0.Results The labeling efficiency and radiochemical purity of HYNIC-Annexin V labeled with technetium-99m reached 95% and 99%,respectively.Abnormal and marked uptake of radioactivity in tumor tissue in tumor-bearing mice received cyclophosphamide intraperitoneally was found to be at 6 h postinjection.Compared with that in tumor tissue at different time point of postinjection,the ratio(1.59±0.44)%ID/g in tumor tissue at 6 h after injection of imaging agent showed the most important(P0.05).The()~(99)Tc~m-HYNIC-Annexin Ⅴ accumulated mainly in the kidney,lung and liver. It was cleared quickly through blood.In comparison with activity([8.85±1.65]%ID/g) at 5 min postinjection,20 percents of initial activity remained in blood([1.59±0.50]%ID/g) at 30 min.The ratio([3.73±1.42]%ID/g) of tumor/muscle was higher than that of tumor/blood([2.80±0.54]%ID/g).Conclusion (()~(99)Tc~m)-HYNIC-Annexin Ⅴ appears to be a potential and useful molecular probe of tumor cell apoptosis.
Annexin A5-functionalized nanoparticles for in vivo dual modality imaging of apoptosis

Abstract:1477 Objectives Noninvasive detection of apoptosis may play an important role in the evaluation of therapy efficiency and disease progression. In this study, we aim to develop Annexin A5-functionalized nanoparticles for dual optical/nuclear imaging of apoptosis. Methods Poly (ethylene glycol) (PEG)-coated, core-crosslinked polymeric micelles (CCPM) were dual-labeled with a near-infrared fluorescence fluorophores (Cy7) and a radioisotope (111In). CCPM nanoparticles were conjugated with Annexin A5. To evaluate the specificity of Annexin A5-CCPM to apoptotic cells, three different preclinical models of apoptosis, including a tumor model, a liver apoptosis model, and an inflammation model were carried out. Results In mice bearing EL4 lymphoma treated with cyclophosphamide and etoposide, the tumor apoptosis was clearly visualized by both SPECT and fluorescence molecular tomography after i.v. administration of 111In-labeled Annexin A5-CCPM. In contrast, there was little accumulation of this nanoradiotracer in the tumors of untreated mice. The biodistribution data was consistent with the imaging data. In liver apoptosis model, the apoptotic liver induced by anti-Fas antibody was clearly visualized by both nuclear and optical images. In contrast, significantly lower signals were detected in healthy livers of untreated mice after injection of 111In-labeled Annexin A5-CCPM and in apoptotic livers after injection of 111In-labeled CCPM. Similarly, 111In-labeled Annexin A5-CCPM showed a significantly higher uptake in the site of inflammation induced by intramuscular injection of turpentine (4.9 %ID/g) than that in the healthy site (0.6 %ID/g). Moreover, in all three models, histological analysis revealed colocalization between radioactivity and fluorescence signals from the nanoparticles, and drug-induced apoptosis. Conclusions Annexin A5-CCPM allowed visualization of apoptosis by both nuclear and optical techniques. The complementary information acquired with multiple imaging techniques should be advantageous in assessing and validating early response to therapy
Experimental Study on Tumor Response to Chemotherapy with 18F-Sfb-annexin B1

Abstract:Background and purpose: One of the main mechanism of chemotherapy is inducing tuomr apoptosis. Molecular imaging can allow noninvasively and dynamically monitor tumor apoptosis in vivo, and help to drug screening and therapeutic evaluation. The purpose of this study was to evaluate the feasibility of 18F-SFB-Annexin B1 in detecting apoptosis at an early phase after chemotheraphy. Methods:Annexin B1 was labeled with 18F using SFB as a chelating agent. Tissue distribution of 18F-SFB-Annexin B1 was studied in healthy mice by the dissection method. W256 tumor-bearing rats were injected with 18F-SFB-Annexin B1 intravenously at 24 h after the treatment of cyclophosphamide (CTX 200 mg/kg) or saline. Then imaging was acquired at 1, 2, 3, and 4 h postinjection on a PET/CT, and the tumor-to-muscle ratio of SUVmax (T/M) and the AI from TUNEL testing were compared. Results: 18F-SFB-Annexin B1 had a radiochemical pruity (RCP)>95%. Biodistribution of this probe showed a predominant uptake in the kidney, then was liver, spleen, and myocardium, rapid clearance from blood and urinary was observed. The radios of T/M were 4.38±0.56, 6.75±1.16, 6.44±1.12, 4.81±0.17, respectively at 1, 2, 3, 4 h post injection of the chemotherapy group, much higher than that of the saline group (2.35±0.14, 2.99±0.55, 3.04±0.41, 2.33±0.47, respectively). The differences between the two groups were significant (F=23.790, 16.913, 14.046, 77.517, respectively, all P<0.05). TUNEL staining revealed that chemotherapy treatment significantly increased the percentage of apoptosis cells with an AI of (21.00±0.04)%in the chemotherapy group, higher than that in the saline group (8.58±0.01)%, the difference was significant (F=21.539, P<0.05). The radios of T/M were significantly correlated with the values of AI (r=0.91, P<0.05). Conclusion: 18F-SFB-Annexin B1 can be used to apoptosis imaging and early therapeutic evaluation in vivo because it can reflect apoptosis at an early stage after chemotheraphy.

Abstract:Background: In this study, a new method for the analysis of cell cycle specificity of apoptosis was designed by using a modified annexin V and propidium iodide (API) method. Methods: Cells of the human promyelocytic HL-60 line treated with camptothecin (CPT) or ultraviolet light (UV) were labeled with fluorescein isothiocyanate-conjugated annexin V and prefixed with 1% methanol-free formaldehyde on ice, and their DNA was stained stoichiometrically with propidium iodide in the presence of digitonin. Cellular green and red fluorescences were measured by flow cytometry. Results: Cell cycle specificity of apoptosis obtained by the API method and those analyzed for the presence of DNA strand breaks by using terminal deoxynucleotidyl transferase (TdT) assay were similar: CPT- or UV-induced apoptosis preferentially in S- or G(1)-phase cells, respectively. When the internucleosomal DNA degradation was prevented by the serine protease inhibitor N-tosyl-L-phenylalanine chloromethyl ketone, apoptotic cells could not be detected by the TdT assay but were identified by the API method. Conclusions: The API method, similar to the TdT assay, accurately detects the cell cycle phase specificity of apoptosis. Also, the API method appears to detect earlier stages of apoptosis than the TdT assay. (C) 2004 Wiley-Liss, Inc.

Dexamethasone-induced murine thymus apoptosis and fas-mediated murine liver apoptosis models were used to investigate the ability of radiolabeled annexin B1 to detect apoptosis in vivo. The labeling procedure yielded a compound with up to 98% radiochemical purity and good in vitro stability. The in vitro binding assays indicated that 99m Tc-HYNIC-annexin B1 retain its PS-binding activity. Biodistribution of the compound in mice showed that 99m Tc-HYNIC-annexin B1 is rapidly cleared from the blood and predominantly accumulates in the kidney. The marked increase in dexamethasone-treated murine thymus uptake and fas-mediated murine liver uptake correlated with histologic evidence of apoptosis. These data suggested that 99m Tc-HYNIC-annexin B1 retain its in vitro and in vivo biological activities. This radiotracer may therefore be useful as a novel radioligand for the noninvasive detecting of PS externalization associated with apoptosis.
Study of the Relationship Between Apoptosis and Intracellular Ph in Single Living Cells Using a Three-Channel Real-Time Fluorescence Imaging Method

Abstract:Using a Dual-View wavelength splitter, a custom-made filter block and only a single intensified charge-coupled device (ICCD), a three-channel real-time fluorescence imaging method for single living cell research was established based on an ICCD fast fluorescence micro-imaging system. The relevant calibrating method for images was also developed to eliminate the influence introduced by spectral crosstalk. Double-labeled with two fluorescent indicators, Annexin V-FITC and SNARF-1, mouse thymocyte was treated with S-nitrosoglutathione (GSNO) and the relationship between their apoptosis and intracellular pH changes was studied in real-time at the single living cell level. The results not only showed the different regularities of intracellular pH changes between apoptosis induced by GSNO and spontaneous apoptosis, but also provided evidence for the application of the three-channel real-time fluorescence imaging method to bio-medical studies.

Annexin A5-conjugated polymeric micelles for dual SPECT and optical detection of apoptosis.

Abstract:Imaging of apoptosis can allow noninvasive assessment of disease states and response to therapeutic intervention for a variety of diseases. The purpose of this study was to develop and evaluate a multimodal nanoplatform for the detection of apoptosis. Methods: To modulate the pharmacokinetics of annexin A5, a 36-kDa protein that binds specifically with phosphatidylserine, annexin A5 was conjugated to polyethylene glycol-coated, core-cross-linked polymeric micelles (CCPMs) dually labeled with near-infrared fluorescence fluorophores and a radioisotope (In-111). To evaluate the specificity of the binding of annexin A5-CCPM to apoptotic cells, both fluorescence microscopy and cell-binding studies were performed in vitro. Pharmacokinetics, biodistribution, dual nuclear and optical imaging, and immunohistochemical studies were performed in 2 xenografted tumor models to evaluate the potential applications of annexin A5-CCPM. Results: In cell-based studies, annexin A5-CCPM exhibited strongly specific binding to apoptotic tumor cells. This binding could be efficiently blocked by annexin A5. In mice, annexin A5-CCPM displayed a mean elimination half-life of 12.5 h. The mean initial concentration in blood was 22.4% of the injected dose/mL, and annexin A5-CCPM was mainly distributed in the central blood compartment. In mice bearing EL4 lymphoma treated with cyclophosphamide and etoposide and in mice bearing MDA-MB-468 breast tumors treated with poly(L-glutamic acid)-paclitaxel and cetuximab (IMC-C225) anti-epidermal growth factor receptor antibody, the tumor apoptosis was clearly visualized by both SPECT and fluorescence molecular tomography. In contrast, there was little accumulation of this nanoradiotracer in the tumors of untreated mice. The biodistribution data were consistent with the imaging data, with tumor-to-muscle and tumor-to-blood ratios of 38.8 and 4.1, respectively, in treated mice, and 14.8 and 2.2, respectively, in untreated mice bearing EL4 lymphoma. Moreover, further studies demonstrated that the conventional Tc-99m-labeled hydrazinonicotinamide annexin A5 and the plain CCPM control exhibited significantly lower uptake in the tumors of the treated mice than annexin A5-CCPM. Immunohistochemistry staining study showed that radioactivity count correlated with fluorescence signal from the nanoparticles, and both signals colocalized with the region of tumor apoptosis. Conclusion: Annexin A5-CCPM allowed visualization of tumor apoptosis by both nuclear and optical techniques. The complementary information acquired with multiple imaging techniques should be advantageous in assessing and validating early response to therapy.

Abstract:There is an increasing need to develop non-invasive molecular imaging strategies for visualizing and quantifying apoptosis status of diseases (especially for cancer) for diagnosis and monitoring treatment response. Since externalization of phosphatidylserine (PS) is one of the early molecular events during apoptosis, Annexin B1 (AnxB1), a member of Annexins family with high affinity toward the head group of PS, could be a potential positron emission tomography (PET) imaging probe for imaging cell death process after labeled by positron-emitting nuclides, such as 18 F. In the present study, we investigated a novel PET probe, 18 F-labeled Annexin B1 ( 18 F-AnxB1), for apoptosis imaging. 18 F-AnxB1 was prepared reliably by conjugating AnxB1 with a 18 F-tag, N -succinimidyl 4-[ 18 F]fluorobenzoate ([ 18 F]SFB), in a radiolabeling yield of about 20 % within 40 min. The in vitro binding of 18 F-AnxB1 with apoptotic cells induced by anti-Fas antibody showed twofold increase compared to those without treatment, confirmed by flow cytometric analysis with AnxV-FITC/PI staining. Stability tests demonstrated 18 F-AnxB1 was rather stable in vitro and in vivo without degradation. The serial 18 F-AnxB1 PET/CT scans in healthy rats outlined its biodistribution and pharmacokinetics, indicating a rapid renal clearance and predominant accumulation into kidney and bladder at 2 h p.i. 18 F-AnxB1 PET/CT imaging was successfully applied to visualize in vivo apoptosis sites in tumor induced by chemotherapy and in kidney simulated by ischemia–reperfusion injury. The high-contrast images were obtained at 2 h p.i. to delineate apoptotic tumor. Apoptotic region could be still identified by 18 F-AnxB1 PET 4 h p.i., despite the high probe retention in kidneys. In summary, we have developed 18 F-AnxB1 as a PS-specific PET probe for the apoptosis detection and quantification which could have broad applications from disease diagnosis to treatment monitoring, especially in the cases of cancer.

Abstract:Apoptosis or programmed cell death is an important form of cell death.Apoptosis is involved in numerous human pathological conditions,such as neurodegenerative diseases,ischemic damage,autoimmune disorders,and many types of cancer.Visualization of apoptosis is enormously beneficial in clinical diagnosis,development of new therapies,and therapeutic evaluation.The traditional methods of apoptosis detection include optical microscopy,TdT-mediated-dUTP nick end labeling analysis,and flow cytometry.However,these invasive techniques restrict the conduct of follow-up studies.Apoptosis imaging in living subjects has contributed to nondestructive observation and in understanding the biological process of apoptosis.The developments in PET and SPECT technologies,including the synthesis of targeted radionuclide tracers,led nuclear medicine into a new era of molecular imaging.The development and application of PET and SPECT as apoptosis imaging probes rendered the non-invasive detection of apoptosis in vivo a reality.This article reviewed the recent advances in apoptosis imaging using radionuclide-labeled tracers.

Abstract:Abstract Apoptosis was a natural, non-inflammatory, energy-dependent form of programmed cell death (PCD) that can be discovered in a variety of physiological and pathological processes. Based on its characteristic biochemical changes, a great number of apoptosis probes for single-photon emission computed tomography (SPECT) and positron emission tomography (PET) have been developed. Radionuclide imaging with these tracers were potential for the repetitive and selective detection of apoptotic cell death in vivo, without the need for invasive biopsy. In this review, we overviewed molecular mechanism and specific biochemical changes in apoptotic cells and summarized the existing tracers that have been used in clinical trials as well as their potentialities and limitations. Particularly, we highlighted the clinic applications of apoptosis imaging as diagnostic markers, early-response indicators, and prognostic predictors in multiple disease fields.

Abstract:At present,there are many methods for detecting apoptosis in vitro,but these methods just are traumatic in material extraction and tissue biopsy by sacrificing of animals in vitro,which limits their clinical application and transformation.In vivo detection methods have become the focus of current research because of non-invasive and real-time monitoring of apoptosis in vivo.Radionuclide apoptotic cell imaging technology among has good research prospects in non-invasive,early stage,dynamic,sensitive,quantitative,and detection intravital.It is the most widely studied and mature technology for detecting apoptosis intravital.Radionuclide apoptosis imaging has been widely used in the detection of apoptosis in cardiovascular diseases,central nervous system diseases,organ transplant rejection,and the evaluation of efficacy and prognosis of malignant tumor after radiotherapy and chemotherapy.This article reviews the progress of radionuclide imaging probes in apoptosis.
