Abstract:Guided by in vitro immunological tests, three immunomodulating steroidal glycosides, stemucronatosides A (1), B (2), and C (3), were isolated from the roots of Stephanotis mucronata. On the basis of chemical evidence and extensive spectroscopic methods including 1D and 2D NMR, their structures were determined as 12-O-deacetylmetaplexigenin 3-[O-6-deoxy-3-O-methyl-beta-D-allopyranosyl-(1-->4)-O-beta-D-cymaropyranosyl- (1-->4)-beta-D-cymaropyranoside], 12-O-deacetylmetaplexigenin 3-[O-beta-D-thevetopyranosyl-(1-->4)-O-beta-D-cymaropyranosyl- apropyranosyl-(1-->4)-beta-D-cymaropyranoside], and metaplexigenin 3-[O-beta-D-glucopyranosyl-(1-->4)-O-6-deoxy-3-O-methyl-beta-D-allopyranosyl-(1 -->4)-O-beta-D-cymaropyranosyl-(1-->4)-beta-D-cymaropyranoside], respectively. These compounds showed immunomodulating activities in vitro.

Abstract:Three new C(21)-steroidal glycosides stemucronatosides H, I, and J (1-3, resp.) were isolated from the stems of Stephanotis mucronata, together with three known compounds sinomarinoside A (4), sinomarinoside E (5), and stephanoside H (6). Abstract:Stephanotis mucronata (Blanco) Merr. has been used for rheumatoid arthritis in Chinese folk herb medicine. Guided by bioactive test, a novel potent immunosuppressive C(21) steroidal glycoside stemucronatoside K (SMK) was isolated from this plant. Its structure was elucidated on the basis of the chemical evidence and extensive spectroscopic methods. We investigated the immunosuppressive effects of SMK in vitro and in vivo. SMK significantly suppressed concanavalin A (Con A)- and lipopolysaccharide (LPS)-stimulated splenocyte proliferation in vitro in a concentration-dependent manner. ICR mice were immunized subcutaneously with OVA on the first day and administered intraperitoneally with SMK at the doses of 2.5, 5, and 10 mg/kg once daily for 10 days. At 24 h after the last administration, mitogen- and OVA-stimulated splenocyte proliferation, the levels of cytokines from splenocytes, and specific antibody titers in serum were measured. SMK significantly inhibited Con A-, LPS- and OVA-induced splenocyte proliferation in the immunized mice in a dose-dependent manner. OVA-specific IgG, IgG1, and IgG2b antibody titers were significantly reduced by SMK compared with the control group. SMK also significantly decreased OVA-induced interleukin-2 (IL-2), interferon-gamma (IFN-gamma), and IL-4 production from splenocytes in the OVA-immunized mice. These results demonstrated that SMK could suppress the cellular and humoral immune response in mice. This study provided evidence to understand the therapeutic effects of S. mucronata and an immunosuppressive natural product compound to further researches to be developed as immunosuppressant.

Abstract:AIM:To investigate chemical constituents of Gypsophila paniculata.METHODS:The compounds were isolated by silica gel,Sephadex LH-20,Rp-C18 column chromatography and preparative liquid chromatography.Their structures were elucidated by ESI-MS,NMR techniques (1H,13C,HMQC,HMBC,TOCSY) and acid hydrolysis.Moreover their α-glucosidase inhibition activity was evaluated compared with that of their aglycone.RESULTS:Two triterpenoid saponins,caryophyllacosides A (1) and B (2),were isolated from the roots of Gypsophila paniculata,without having recourse to chemical degradation or modification.Both of them were found to have potent α-glucosidase inhibitiory activity.CONCLUSION:The two triterpenoid saponins are new boactive compounds.

Abstract:Two new glycoalkaloids, manshurienine C (1) and 8-oxotetrahy-corydalmine-1-O-β-D-glucopyranoside (2), along with eleven known alkaloids (3–13), were isolated from the tubers of Stephania succifera. Their structures were elucidated by spectroscopic methods, including NMR and MS spectra. Compounds 5, 6, 8, and 10 showed in vitro antibacterial activity against methicillin-resistant Staphylococcus aureus (MRSA).
Comparison of immunosuppressive activity of stephanoside E and its aglycone from Stephanotis mucronata in vitro

Abstract:Stephanoside E (STE) and its aglycone Stephanthraniline A (STA) isolated from the stems of Stephanotis mucronata (Blanco) Merr. were evaluated for their suppressive potentials on Th1 and Th2 immune responses in vitro, and the effects of the C-3 sugar chains on their activities were discussed. STE and STA significantly inhibited Con A and LPS-induced splenocyte proliferation, and reduced the production of Th1/Th2 cytokines (IL-2, IFN-γ, IL-4, and IL-10) from Con A-stimulated splenocytes in a concentration-dependent manner. The mRNA expression levels of Th1/Th2 cytokines (IL-2, IFN-γ, IL-4, and IL-10) and transcription factors (T-bet and GATA-3) in Con A-stimulated splenocytes were also suppressed by STE and STA. The suppressive effect of STA on Th1 and Th2 immune responses in vitro was stronger than that of STE. These findings suggested that both STE and STA could simultaneously inhibit Th1/Th2 immune responses, and the glycosyl formation at the position C-3 of STA could decrease the immunosuppressive activity in vitro.

Abstract:The stems of the Chinese traditional medicine Stephanotis mucronata were screened for immunologically active pregnane glycosides using high-performance liquid chromatography (HPLC) coupled with electrospray ionization tandem mass spectrometry. In the mass spectra of pregnane glycosides, predominant [M+Na](+) ions were observed and used to determine the molecular masses, while fragmentation reactions of the [M+Na](+) ions were recorded to provide information on the primary sequences of oligosaccharide chains in terms of classes of monosaccharide. Fragment ions from the side-chain cleavage of aglycone portions can provide mass information about side-chain substitutions. To further confirm the fragment ion structures, Fourier transform ion cyclotron resonance tandem mass spectrometry (MSn) with low-energy collision-induced dissociation was performed using samples collected from HPLC fractions, which provided accurate elemental compositions of fragment ions. Based on fragmentation patterns and comparison with standards, ten pregnane glycosides were identified as stemucronatosides C, D, F, and G, mucronatosides A, B, and C, stephanoside E, and two glycosides that are identified in the S. mucronata extracts for the first time. The latter two pregnane glycosides are 12-O-cinnamoyldeacetylmetaplexigenin-3-O-6-deoxy-3-O-methyl-beta-D-allopyranosyl-(1 -> 4)-beta-D-cymaropyranosyl-(1 -> 4)-beta-D-cymaropyranoside and 12-O-cinnamoyl-20-O-acetyl (20S)-pregn-6-ene-3 beta,5 alpha,8 beta,12 beta,14 beta,17 beta,20-heptaol 3-O-beta-D-thevetopyranosyl-(1 -> 4)-beta-D-cymaropyranosyl-(1 -> 4)-beta-D-cymaropyranoside. Stemucronatoside L, a pregnane glycoside from the roots of Stephanotis mucronata, inhibits Th1/Th2 immune responses in vitro

Abstract:Stemucronatoside L (SML), isolated from Stephanotis mucronata, could suppress the activation of T cells in vitro. However, the mechanisms responsible for its immunosuppressive activity remain poorly understood. The purpose of this study was to investigate whether SML could suppress Th1/Th2 immune responses and to characterize the cellular mechanisms involved. Effects of SML on T-lymphocyte subsets and the production of Th1 cytokines IL-2 and IFN-gamma, and Th2 cytokines IL-4 and IL-10 from ConA-stimulated mice splenocytes were detected by flow-cytometric analysis and ELISA method, respectively. Furthermore, effects of SML on mRNA expression level of Th1/Th2 cytokines and transcription factors T-bet and GATA-3 were evaluated by RT-PCR analysis. SML not only significantly decreased the percentage of CD4(+) T cells and the CD4(+)/CD8(+) ratio, but reduced the production of Th1/Th2 cytokines in a concentration-dependent manner. The mRNA expression levels of Th1/Th2 cytokines and transcription factors (T-bet and GATA-3) were also suppressed by SML. These results suggested that SML could simultaneously inhibit Th1/Th2 immune responses by suppressing gene expression of Th1/Th2 cytokines and transcription factors.

Abstract:Two new stilbenoids, stemobenoids A (1) and B (2), together with three known compounds were obtained from the roots of Stemona tuberosa. The structures of the new compounds were established by extensive spectroscopic analysis, including HRMS, 1D and 2D NMR data. Compounds 1 and 2 displayed potent quinone reductase inducing activity in Hepa 1c1c7 cells.

Abstract:Stephanthraniline A (STA) isolated from the stems of Stephanotis mucronata (Blanco) Merr. was evaluated for their suppression on T cells' immune responses in vitro and in vivo. In vivo, oral administration of STA significantly inhibited T cell-mediated delayed-type hypersensitivity (DTH) response. In vitro, STA has inhibitory effects on T cell proliferation induced by CD3/CD28 cross-linking or Con A; additionally, CD4+ T cells are more sensitive to this inhibition than CD8+ T cells. STA also suppressed the production of cytokines (IL-2, IFN-γ, IL-4 and IL-17) and mRNA expression of the genes associated with T cell activation, proliferation and differentiation. Our data indicate that STA inhibits the proliferation of T cells by inducing cell cycle arrest but not inducing apoptosis. The inhibitory mechanism of STA on T cells was correlated with the gene change related to multi-signal transduction pathways. Furthermore, we also provided lines of evidence that STA, distinct from glucocorticoids, did not activate the glucocorticoid receptor. These findings would be beneficial for further understanding the therapeutic effects of S. mucronata in the treatment of autoimmune diseases. It also suggested the potential of the natural steroid STA as the effective candidate compounds for use in the treatment of inflammatory and autoimmune diseases.
Steroidal glycosides from Yucca rostrata and Dracaena braunii and their cytotoxic and antimicrobial evaluation

Abstract:Steroidal glycosides are known to possess a wide range of biological activities such as cytotoxicity and antimicrobial effects. To isolate more of these glycosides to get a better understanding at their structure/activity relationship, and to complete chemotaxonomic information, a cultivated Yucca rostrata was studied, along with a wild and a cultivated Dracaena braunii . Thirteen spiro- and furostan glycosides were isolated from those plants and their structure were elucidated using 1D and 2D NMR. Since they were already reported from other plant species, it allowed us to get an idea on which type of structure might be specific to which taxon. The compounds from D. braunii were tested for their antimicrobial activity against Candida albicans , C. glabrata and Staphylococcus aureus Only compound 12 ( CAS 39491-39-9) showed a weak activity on S. aureus with a MIC of 50 μg/mL. The compounds from Y. rostrata were tested for their cytotoxicity against a CT26 (murine colorectal carcinoma) cell line. The spirostan glycosides 4 (CAS 119459-80-2) and 5 (CAS 164592-97-6) showed an interesting activity with an IC 50 of 2.38 μM and 2.26 μM, respectively. Comparing the structures of the active versus the inactive ones, we were able to formulate some hypotheses regarding the structure/activity relationships, in accordance with the current literature.

Abstract:Objective:To study the chemical constituents from the tuberous roots of Stephania succifera.Methods:The compounds were isolated and purified by various chromatographic methods,and their structures were elucidated by spectroscopic methods and physicochemical properties.Results:Eleven alkaloids were isolated and identified as dehydrocrebanine(1),L-tetrahydropalmatine(2),crebanine(3),(-)-sukhodianine-β-N-oxide (4),promucosine (5),(-)-sukhodianine (6),4-hydroxycrebanine (7),N-methylasimilobine-2-O-β-D-glucopyranoside (8),palmatine (9),(+)-stepharine (10) and (-)-asimilobine (11).Conclusion:Compound 5 is isolated from Stephania genus for the first time,and compounds 5,7,8 and 10 are isolated from this plant for the first time.

Abstract:Two new steroidal saponins, named 26-O-β-d-glucopyranosyl-22ξ-hydroxy-(25R)-furost-5-en-3β, 26-diol, 3-O-β [xylopyranosyl (1→3) α-l-rhamnopyranosyl (1→2) β-d-glucopyranoside] (1) and 3-O-β-d-xylopyranosyl (1→3) α-l-rhamnopyranosyl (1→3) β-d-glucopyranoside diosgenin (2), along with protobioside (3), diosgenin (4), β-sitosterol (5) and β-Sitosterol-3-O-β-d-glucopyranoside (6) were isolated from the rhizomes of Polygonatum verticillatum. The structures were elucidated based on physicochemical parameters and spectroscopic analysis. NMR (1 & 2D techniques) was employed to estimate compounds (1-6) in the parent extract (E1) and its fractions (E2 and E4). Cytotoxicity studies of parent extract (E1) and isolates were evaluated against A549 and MCF-7 cell lines. Compounds 1 and 2 exhibited potent cytotoxic activity in both the cell lines and induces apoptosis. These findings will provide two new potent anticancer molecules for the chemical repository.

Abstract:Two new hasubanan alkaloids, stephalonester A (1) and stephalonester B (2), together with four known compounds, stephalonine E (3), longanone (4), cephatonine (5), and prostephabyssine (6) were isolated from the whole plant of Stephania longa. Their structures were determined by HR-ESI-MS, 1 D and 2 D NMR, ECD calculations, as well as by comparison with literature values. All compounds were evaluated for their anti-inflammatory activity in vitro. Compounds 4, 5, and 6 exhibited significantly inhibitory effects on TNF-α and IL-6 production with IC50 values range from 6.54 to 30.44 µM.Graphical

Abstract:Three previously undescribed steroidal constituents including two sterols ( 1 – 2 ) and one pregnane-type steroidal glycoside ( 6 ), along with nineteen known ones ( 3 – 5, 7 – 22 ), were isolated from the 80% alcohol extraction of Solanum nigrum L. Their structures and the absolute configurations were established by analysis of the extensive spectroscopic data ( 1 H/ 13 NMR, 1 H 1 H COSY, HSQC, HMBC, and NOESY), and/or by comparisons of the experimental electronic circular dichroism (ECD) spectra with those calculated ones by TDDFT method. Further, a MTT assay was applied to demonstrate that compounds 1 – 4 , 6 – 12 , 18 , and 22 exhibited significant cytotoxic activities against SW480 cells, and compounds 1 – 4 , 6 – 14 , and 16 – 22 showed significant cytotoxic activities against Hep3B cells.

Abstract:Eurysoloids A (1) and B (2), two novel diastereomeric sesterterpenoids possessing a pentacyclic 5/6/5/10/5 framework with an unusual macrocyclic ether system, were isolated from Eurysolen gracilis Prain. Their structures were unambiguously determined by spectroscopic, single-crystal X-ray diffraction and DP4+ analyses. A plausible biosynthetic pathway for compounds 1 and 2 was proposed. Abstract:Two new phenolic glycosides, Diplostephioside D ( 1 ) and Diplostephioside E ( 2 ) together with ten known phenols ( 3 – 12 ) were isolated from the dried roots of Iris domestica (L.) Goldblatt & Mabb. Among them, compounds 4 – 9 have not been reported previously from the family Iridaceae. The chemical structures were elucidated based on extensive spectroscopic experiments, including 1D and 2D NMR as well as HR-ESI-MS, and comparison with those reported in the literature. All of the isolated compounds were evaluated for their cytotoxic activity against HeLa cells. Compounds 1 , 2 , 4 , 8 and 12 showed cytotoxic activities with IC 50 values ranging from 13.52 ± 1.52 to 53.33 ± 1.40 μM.
