Abstract:A method for the chemical modulation of the photoluminescence of a DNA-bound diruthenium(II) complex, [(bipy)2Ru(bpib)Ru(bipy)2]4+ (bipy = 2,2′-bipyridine, bpib = 1,4-bis([1, 10]phenanthroline [5,6-d]imidazol-2-yl) benzene) by the introduction of Cu2+ ion and EDTA has been developed. The diruthenium(II) complex showed strong photoluminescence both in buffer solutions and on an indium-tin oxide (ITO) surface, which was not modulated by Cu2+ or EDTA. The DNA-bound [(bipy)2Ru(bpib)Ru(bipy)2]4+ with a binding constant of 3.8 × 104 M−1 showed an enhancement in the luminescence based on the electrostatic interaction between the complex and DNA. The presence of Cu2+ was found to quench the luminescence of DNA-bound [(bipy)2Ru(bpib)Ru(bipy)2]4+, but the quenched luminescence was recovered by addition of an equimolar concentration of EDTA. Hence, the photoluminescence of DNA-bound [(bipy)2Ru(bpib)Ru(bipy)2]4+ depends strongly on the introduction of Cu2+ and EDTA.
Nuclease Activity of Copper(ii) Complex Containing 1,10-Phenanthroline-5,6-dione and L-phenylalanine Ligands

Abstract:The interaction between CT-DNA and a ternary copper (II) complex, [Cu(phendio)(L-Phe)(H 2 O)](ClO 4 )·H 2 O (CuPP, phendio = 1,10-phenanthroline-5,6-dione, L-Phe = L-phenylalanine), has been conducted by electronic spectra, fluorescence spectroscopy and cyclic voltammetry. It has been found that the max. absorption peak from the electronic spectra is red shifted and the intensity is weakened and that the values of peak current from cyclic voltammetry are decreased significantly in the presence of DNA compared with that in the absence of DNA. At the same time, the complex can quench the emission intensity of EB-DNA system. The existence of the intercalation mode between the complex and DNA was proven. By submarine gel electrophoresis, we found that the copper(II) complex can cleave circular plasmid pBR322 DNA into nicked and linear forms in the presence of ascorbic acid and H 2 O 2 .
DNA Damage by Copper(ii) Complexes: Coordination-Structural Dependence of Reactivities.

Abstract:A variety of copper complexes with different structural features have been shown to bind double-helical DNA with binding constants of 104–107 M−1 and to promote double-strand DNA damage upon reductant/H2O2 activation. The interaction of the Cu complex with DNA results in hyperchromism and shifts to longer wavelengths of the strongest transitions in the Cu complexes, as well as striking hypochromism or hyperchromism of DNA absorption at 260 nm. In the presence of DMPO as the spin trap, the solution of each copper complex exhibits typical four-line ESR spectra of the hydroxyl radical by adding 2-mercaptoethanol and H2O2 to the solution. Quantitation by 2-deoxy-D-ribose shows that the competence of hydroxyl radical generation by the copper complexes upon reductant and H2O2 activation decreases in order, that is, Cu(HTCD)2+∼Cu(Im)4Cl2∼Cu(IDB)(NO3)2>Cu(IDB)Cl2>Cu(IDBt)Cl2. The copper complex-mediated hydroxyl radical, a powerful oxidant that attacks the adjacent DNA, is responsible for the DNA oxidative damage. The λDNA damage chemistry illustrates that the competence and selectivity of double-strand λDNA damage by the copper complexes are dependent on their geometric structures and types of ligands. The decreasing order of the DNA damage capacity by the present complexes is Cu(Im)4Cl2∼Cu(IDB)(NO3)2>Cu(HTCD)2+>Cu(IDBt)Cl2>Cu(IDB)Cl2.
Synthesis, characterization, DNA interaction and cytotoxic activities of copper complexes with ethyl 2-[bis(2-pyridylmethyl)amino]propionate.

Abstract:Copper(II) complexes of ethyl 2-[bis(2-pyridylmethyl)amino]propionate ligand (ETDPA) have been synthesized and characterized by elemental analyses, IR spectra, UV spectra and ES-MS. These complexes are stable in air with the formula of [(ETDPA)CuCl2] and [(ETDPA)Cu(phen)](ClO4)2. The DNA-binding properties of the Cu(II) complexes have been investigated by ultraviolet spectroscopy and fluorescence spectroscopy, which showed that the binding mode of the two complexes with DNA was different. The binding mode of [(ETDPA)CuCl2] is not the classical intercalation binding, and the binding mode of [(ETDPA)Cu(phen)](ClO4)2 with DNA is intercalation. The cytotoxic assay shows that the [(ETDPA)Cu(phen)](ClO4)2 is more active on the proliferation of cancer cells than the [(ETDPA)CuCl2].

Abstract:The effects of copper ion on the interaction of [Ru(bpy)2HPIP]2+(bpy = 2,2′-bipyridine, HPIP = 2-(2-hydroxyphenyl) imidazo [4,5-f] [1, 10] phenanthroline) with DNA have been investigated by electronic absorption spectroscopy and fluorescence spectroscopy. HPIP ligand of the complex with an intramolecular hydrogen bond can bind Cu2+ in the absence of DNA, as revealed by the absorbance and fluorescence decrease for [Ru(bpy)2HPIP]2+. The resultant heterometallic complex binds to DNA via intercalation of HPIP into the DNA base pairs and its DNA-binding ability is stronger than [Ru(bpy)2HPIP]2+ itself. The DNA bound [Ru(bpy)2HPIP]2+ cannot bind Cu2+ at low Cu2+ concentration and the intramolecular hydrogen bond in HPIP is located inside the DNA helix. While the Cu2+ concentration is relative high, Cu2+ can quench the fluorescence of DNA bound [Ru(bpy)2HPIP]2+. The quenching reason is proposed.
Synthesis, DNA interaction and anticancer activity of copper(II) complexes with 4'-phenyl-2,2':6',2″-terpyridine derivatives.

Abstract:Three novel copper(II) complexes CuL1Cl2 (1) (L1=4′-(3-methoxyphenyl)-2,2′:6′- 2″-terpyridine), CuL2Cl2 (2) (L2=4′-(4-methoxyphenyl)-2,2′:6′-2″-terpyridine) and CuL3Cl2 (3) (L3=4′-(3,5-dimethoxyphenyl)-2,2′:6′-2″-terpyridine) have been synthesized and characterized. Absorption spectral titration experiments, ethidium bromide displacement assays, and cyclic voltammetric experiments were carried out and the results suggested that these complexes bound to DNA through an intercalative mode. Moreover, these complexes were found to cleave pBR322 DNA efficiently in the presence of glutathione (GSH), and exhibited good anticancer activity against HeLa, Hep-G2 and BEL-7402 cell lines. Nuclear chromatin cleavage was also observed by acridine orange/ethidium bromide (AO/EB) staining assays and comet assays. These results demonstrated that these three Cu(II) complexes caused DNA damage and induced the apoptosis of HeLa cells. Mechanistic investigations revealed the participation of reactive oxygen species which can be trapped by reactive oxygen species (ROS) radical scavengers and ROS sensors.
Luminescence properties of a di-ruthenium(II) complex with an intramolecular hydrogen bond modulated by DNA and copper(II) ion

Abstract:Modulation of the luminescence properties of a di-ruthenium(II) complex [(bpy) 2 Ru(BL)Ru(bpy) 2 ] 4+ (bpy = 2,2′-bipyridine, BL = 2-hydroxyl-5-methyl-1,3-bis([1,10]phenanthroline-[5,6-d]imidazol-2-yl)benzene) by DNA and/or Cu 2+ ion has been investigated. It is found that the ruthenium(II) complex can coordinate to the Cu 2+ ion in both the absence and presence of DNA. Binding to DNA is through electrostatic interactions and the intramolecular hydrogen bond in the complex is located outside of the DNA. The binding constant is 1.6 × 10 4 M −1 . Moreover, it is demonstrated that DNA has the ability to enhance the luminescence intensities of both the di-ruthenium(II) complex and the tri-metallic system generated by chelating with Cu 2+ . Conversely, Cu 2+ ion can quench the luminescence of both the free ruthenium(II) complex and the DNA-bound ruthenium(II) complex.

Abstract:The interaction between salmon testes DNA (st-DNA) and a series of Cu(II) polypyridyl complexes, i.e. [Cu(dmbpy)(NO3)2] (1) (dmbpy = 4,4'-dimethyl-2,2'-bipyridine), [Cu(bpy)(NO3)2] (2) (bpy = 2,2'-bipyridine), [Cu(phen)(NO3)2] (3) (phen = phenanthroline), [Cu(terpy)(NO3)2]·H2O (4) (terpy = 2,2':6',2″-terpyridine), [Cu(dpq)(NO3)2] (5) (dpq = dipyrido-[3,2-d:2',3'-f]-quinoxaline) and [Cu(dppz)(NO3)2] (6) (dppz = dipyrido[3,2-a:2',3'-c]phenazine) was studied by UV/Vis absorption, Circular Dichroism, Linear Dichroism, EPR, Raman and (UV and vis) resonance Raman spectroscopies and viscometry. These complexes catalyse enantioselective C-C bond forming reactions in water with DNA as the source of chirality. Complex 1 crystallizes as an inorganic polymer with nitrate ligands bridging the copper ions, which adopt essentially a distorted square pyramidal structure with a fifth bridging nitrate ligand at the axial position. Raman spectroscopy indicates that in solution the nitrate ligands in 1, 2, 3 and 4 are displaced by solvent (H2O). For complex 1, multiple supramolecular species are observed in the presence of st-DNA in contrast to the other complexes, which appear to interact relatively uniformly as a single species predominantly, when st-DNA is present. Overall the data suggest that complexes 1 and 2 engage primarily through groove binding with st-DNA while 5 and 6 undergo intercalation. For complexes 3 and 4 the data indicates that both groove binding and intercalation takes place, albeit primarily intercalation. Although it is tempting to conclude that the groove binders give highest ee and rate acceleration, it is proposed that the flexibility and dynamics in binding of Cu(II) complexes to DNA are key parameters that determine the outcome of the reaction. These findings provide insight into the complex supramolecular structure of these DNA-based catalysts.

Abstract:To investigate the effect of intercalative ligand on their biological activity, five new complexes [Ru(bpy)2(dpq)](ClO4)2 (bpy=2,2′-bipyridine, dpq=dipyridoquinoxaline), [Ru(bpy)2(dicnq)](ClO4)2 (dicnq=pyrazino[2,3-f]phenanthroline-2,3-dicarbonitrile), [Ru(bpy)2(dppp3)](ClO4)2 (dppp3=pyrido[3′,4′:5,6]pyrazino[2,3-f][1,10]phenanthroline), [Ru(bpy)2(dppp2)](ClO4)2 (dppp2=pyrido[2′,3′:5,6]pyrazino[2,3-f][1,10]phenanthroline) and [Ru(bpy)2(OCH3-dppz)](ClO4)2 (OCH3-dppz=11-methoxy-dipyrido[3,2-a:2′,3′-c]phenazine) have been synthesized and characterized in detail by 1H NMR spectroscopy, mass spectrometry and elemental analysis. Their interaction with calf thymus DNA and the inhibitory activity towards topoisomerase II and T7 RNA polymerase have been examined by a series of experimental methods. The results suggest that a high DNA-binding affinity is important for their topoisomerase II and DNA transcription inhibition. Intercalative ligands with more extended aromatic area, better conjugate effect and presence of electron-withdrawing substituted groups are beneficial to the DNA intercalation and enzymatic inhibition of their polypyridyl Ru(II) complexes.
Effects of Copper Ions on DNA Binding and Cytotoxic Activity of a Chiral Salicylidene Schiff Base

Abstract:A chiral Schiff base HL N-(5-bromo-salicylaldehyde)dehydroabietylamine (1) and its chiral dinuclear copper complex [Cu2L4]·4DMF (2) have been synthesized and fully characterized. The interactions of 1 and 2 with salmon sperm DNA have been investigated by viscosity measurements, UV, fluorescence and circular dichroism (CD) spectroscopic techniques. Absorption spectral (Kb=3.30 × 10(5)M(-)(1) (1), 6.63 × 10(5)M(-)(1)(2)), emission spectral (Ksv=7.58 × 10(3)M(-)(1) (1), 1.52 × 10(4)M(-)(1) (2)), and viscosity measurements reveal that 1 and 2 interact with DNA through intercalation and 2 exhibits a higher DNA binding ability. In addition, CD study indicates 2 cause a more evident perturbation on the base stacking and helicity of B-DNA upon binding to it. In fluorimetric studies, the enthalpy (ΔH>0) and entropy (ΔS>0) changes of the reactions between the compounds with DNA demonstrate hydrophobic interactions. 1 and 2 were also screened for their cytotoxic ability and 2 demonstrates higher growth inhibition of the selected cancer cells at concentration of 50 μM, this result is identical with their DNA binding ability order. All the experimental results show that the involvement of Cu (II) centers has some interesting effect on DNA binding ability and cytotoxicity of the chiral Schiff base.

Abstract:1,10-Phenanthroline-5,6-diol (dpcatH2) was prepared conveniently from 1,10-phenanthroline-5,6-dione using hydrazine sulfate as reductant, and a new bimetallic Cu(II) complex, [(phen)Cu(dpcat)Cu(phen)](ClO4)2, where phen denotes 1,10-phenanthroline was synthesized by a “one-pot” method. Presence of calf thymus DNA would result in an evident change in the electronic absorption, quenching effect on the fluorescence, protection from fluorescence quencher ferrocyanide and disappearance of redox process of the complex, and the viscosity of DNA would be enhanced by addition of the complex. All these facts suggest that the dicopper(II) complex binds to double helix DNA by classical intercalation.
Oxidative DNA Cleavage Promoted by Polynuclear Copper Complexes Bearing Iminodiacetate Chelator

Abstract:The rigid-spaced polycopper(II) complexes of different benzyl polyiminodiacetic acid (IDA) ligands, Cu2L1 (complex 1, meta-dicopper complex), Cu2L2 (complex 2, para-dicopper complex) and Cu3L3 (complex 3) (H4L1, N,N,N′,N′-m-xylylenediamine tetraacetic acid; H4L2, N,N,N′,N′-p-xylylenediamine tetraacetic acid; H6L3, N,N,N′,N′,N″,N″-mesitylenetriamine hexaacetic acid) were prepared to explore the chelator’s role in determining the DNA cleavage behavior of polycopper complexes by comparing these complexes of IDA chelator with their analogues of bis(2-pyridylmethyl)amine (BPA) chelator. PolyCu(IDA) complexes 1–3 show the synergic effect between copper(II) centers. Their DNA cleavage activity follows an order of 3>1>2, which is consistent with their DNA binding abilities. However, this consistence was not observed in the case of polyCu(BPA) complexes according to our previous study. In addition, both the DNA binding ability and cleavage efficiency of polyCu(IDA) complexes are distinctly lower than those of polyCu(BPA) complexes. Furthermore, both the meta- and para-dicopper complexes are able to mediate linear DNA from supercoiled DNA at high complex concentration for polyCu(IDA) complexes, while for their analogues with BPA chelators, only the meta-dicopper analogue could induce the linear DNA from supercoiled DNA. These results suggested that the replacement of BPA chelator with the anionic IDA chelator alters distinctly the DNA cleavage behavior of rigid-spaced polycopper complexes.
DNA-Binding and Cytotoxicity of Copper(I) Complexes Containing Functionalized Dipyridylphenazine Ligands

Abstract:A set of copper(I) coordination compounds with general formula [CuBr(PPh3)(dppz-R)] (dppz-R = dipyrido[3,2-a:2',3'-c]phenazine (Cu-1), 11-nitrodipyrido[3,2-a:2',3'-c]phenazine (Cu-2), 11-cyanodipyrido[3,2-a:2',3'-c]phenazine (Cu-3), dipyrido[3,2-a:2',3'-c]phenazine-11-phenone (Cu-4), 11,12-dimethyldipyrido[3,2-a:2',3'-c]phenazine (Cu-5)) have been prepared and characterized by elemental analysis, H-1-NMR and P-31-NMR spectroscopies as well as mass spectrometry. The structure of Cu-1 was confirmed by X-ray crystallography. The effect of incorporating different functional groups on the dppz ligand on the binding into CT-DNA was evaluated by absorption spectroscopy, fluorescence quenching of EtBr-DNA adducts, and viscosity measurements. The functional groups affected the binding modes and hence the strength of binding affinities, as suggested by the changes in the relative viscosity. The differences in the quenching constants (K-sv) obtained from the fluorescence quenching assay highlight the importance of the functional groups in altering the binding sites on the DNA. The molecular docking data support the DNA-binding studies, with the sites and mode of interactions against B-DNA changing with the different functional groups. Evaluation of the anticancer activities of the five copper compounds against two different cancer cell lines (M-14 and MCF-7) indicated the importance of the functional groups on the dppz ligand on the anticancer activities. Among the five copper complexes, the cyano-containing complex (Cu-3) has the best anticancer activities.
Effect of Adenine Moiety on Dna Binding Property of Copper(Ii)-Terpyridine Complexes

Abstract:Two novel copper(ii) terpyridine complexes, [Cu(atpy)(NO(3))(H(2)O)](NO(3)).3H(2)O () and [Cu(ttpy)(NO(3))(2)] () (atpy = 4'-p-N9-adeninylmethylphenyl-2,2':6,2''-terpyridine; ttpy = 4'-p-tolyl-2,2':6,2''-terpyridine) have been prepared and structurally characterized by X-ray crystallography. Both complexes show a CuN(3)O(2) coordination in a square pyramidal (4 + 1) geometry with terpyridine acting as an equatorial ligand. For complex , intermolecular AA base pairing interaction is observed between N(6) and N(1) of adjacent adenines with N(6)N(1) of 3.027(7) A. A molecular dynamics simulation of the DNA binding of two complexes showed that the adenine moiety plays an important role in the intercalation of into DNA. This is verified by UV, fluorescence, circular dichroism and flow linear dichroism studies. The promotional effect from the adenine moiety to the intracellular DNA binding of complex is also confirmed by the inductively coupled plasma mass (ICP-MS) spectrometry data which showed a significant higher copper content in DNA isolated from complex treated MCF-7 and HeLa cells.

Abstract:Mononuclear copper(II) complex [Cu(dppt)2(H2O)2](ClO4)2 (1), where dppt is bidentate NpyNtz donor asymmetric ligand (pyridyl-triazine) has been isolated. The X-ray crystal structure of 1 possesses a CuN4O2 chromophore with elongated octahedral geometry. The electronic and EPR spectral properties demonstrate that the solvent molecules strongly interacted in the axial position thereby weakens the CuN4 coordination plane. Absorption and emission spectral and electrochemical measurements clearly show the partial intercalative binding of 1 to calf thymus (CT) DNA. Remarkably, it exhibits potent cytotoxicity (IC50, 3.73 M) against human cervical carcinoma cells (HeLa), which is 4.5 times better than cisplatin and is non-toxic (IC50, >500 M) to normal mouse embryonic fibroblasts cells (NIH 3T3). It blocks cell cycle progression of HeLa cells in G1 phase. FACSverse analysis of 1 is suggestive of ROS (reactive oxygen species) generation and absolutely induces apoptotic cell death in HeLa cells.

Abstract:A novel, yet effective method for identifying DNA-binding modes of [Ru(bpy)(2)dmt](2+) (where bpy = 2,2'-bipyridine and dmt = 2,3-dimethyl-1,4,8,9-tetra-aza-triphenylene) on an indium tin oxide electrode has been successfully developed by introducing Cu2+ ion and ethylenediaminetetraacetic acid. The results from emission spectra and fluorescence microscopic images suggested that [Ru(bpy)(2)dmt](2+) not only associates with Cu2+ ion in both the absence and presence of DNA but also shows strong affinity with DNA in the presence of Cu2+. Evidence for the strong binding of [Ru(bpy)(2)dmt](2+) to DNA was determined from the interface studies using electrochemical methods. The present study suggests that a combination of photoluminescence measurement with electrochemical methods identifies the DNA-binding behavior of luminescent molecules with redox activities. [Ru(bpy)(2)dmt](2+) binds to DNA via an intercalative mode.

Abstract:Two new dicopper(II) complexes bridged by N,N'-bis(dipropylenetriamine)oxamide (H2oxdipn), namely, [Cu2(oxdipn)](pic)2(1) and [Cu2(oxdipn)(ClO4)2] (2), where pic represents picrate ion, have been synthesized and characterized by elemental analyses, molar conductance measurements, IR and electronic spectral studies, and X-ray single crystal diffraction. In both dicopper(II) complexes, the two copper(II) ions are bridged by trans-oxdipn ligand with the Cu⋯Cu separations of 5.2536(15) and 5.231(2)Å, respectively. The copper(II) ion in complex 1 has a square-planar coordination geometry, while that in 2, a square-pyramidal. Linked with classical hydrogen bonds, the molecules of complex 1 consist of a one-dimensional chain, while complex 2 molecules result in a two-dimensional structure. Numerous hydrogen bonds link complex 1 or 2 into a 2-D infinite network. In vitro cytotoxicity experiment shows that the two dicopper(II) complexes exhibit cytotoxic effects against the selected tumor cell lines. The reactivity towards herring sperm DNA (HS-DNA) and bovine serum albumin (BSA) reveals that the two dicopper(II) complexes can interact with the DNA in the mode of intercalation, and effectively quench the intrinsic fluorescence of BSA via a static mechanism. The influence of different counterions in this kind of dicopper(II) complexes on DNA/BSA-binding properties, and the in vitro cytotoxic activities was investigated.

Abstract:In this paper, macrocyclic copper coordination compound was synthesized. Interaction of this complex with DNA was studied by means of UV-Visible, fluorescence spectroscopy. In addition, the cleavage of DNA induced by this complex studied by agarose gel electrophoresis showed the activity of chemical nuclease of this complex, it has been estimated the molecular weight of products.

Abstract:A total of eight diruthenium sawhorse and cationic complexes incorporating carboxylated chalcones with phenyl or ferrocenyl units have been synthesized and characterized. The electrochemical behavior of the sawhorse and cationic complexes incorporating the ferrocenyl moiety have been studied by cyclic voltammetry. For the cationic complexes both the diruthenium core and the ferrocenyl unit undergo oxidation, in contrast only the ferrocenyl moieties for the sawhorse complexes were oxidized. The enhanced solubility of the cationic complexes over the neutral sawhorse compounds in aqueous medium enabled DNA binding studies to be undertaken for the former. All of the cationic complexes could bind to DNA, and displayed better selectivity for quadruplex over duplex DNA. These results indicate that diruthenium cationic complexes are able to bind to quadruplex DNA and have the potential to function as telomerase inhibitors.

Abstract:Three new copper(II) complexes of N-benzyl di(pyridylmethyl)amine (phdpa) were synthesized and characterized by spectroscopic methods. The interaction between CT-DNA and the complexes was studied by UV and fluorescence titration methods. It was found that the complex [(phdpa)Cu(H(2)O)Ac)](Ac), with the non-planar aromatic heterocyclic ring ligand (phdpa), showed good anticancer properties and could cause the fragmentation of the nucleus, although its interaction with CT-DNA was weaker than that of 1,10-phenanthroline (phen) -based copper(II) complexes. The anticancer activities of copper(II) complexes with phdpa and phen based ligands are correlated to their binding constants with DNA, but phen-based copper(II) complexes did not cause the nucleus fragmentation of HeLa cells. [(phdpa)Cu(H(2)O)Ac)](Ac) can noticeably decrease the oxygen content of a culture solution and of HeLa cells, which make it a new nucleus and oxygen related anticancer copper(II) complex. Information obtained here would be helpful in the design of new antitumor complexes in oxidative therapy.
