Abstract:Objective To express HSPB7 protein possessing biological activities in E.coli and prepare the polyclonal antibody against HSPB7.Methods The full length gene fragment of HSPB7 was amplified from rat heart by RT-PCR,then was cloned into pET43.1a(+)prokaryotic expressing vector.pET-43.1a-HSPB7 was induced by IPTG in E.coli BL21(DE3)and HSPB7 protein was expressed withoyt tag.After purification,the purified protein was detected by MS/MS and biological activity assay.The purified protein was used to immunize New Zealand rabbits for obtaining the antiserum.The titers and specificity of the antibody were measured by Western blotting.Results Rat HSPB7 gene was cloned and the plasmid pET-43.1a-HSPB7 expressing a 18 600 Da protein was constructed.The purified protein was characterized by ESI-Q-TOF-MS/MS assay.HSPB7 has molecular chaperon activities which were temperature-dependent in vitro.The antibody against HSPB7 showed high titer and specificity,and could recognize native rat HSPB7 protein.Conclusion The purified HSPB7 protein with biological activities and the anti-HSPB7 polyclonal antibody are obtained,which provide reliable tools for the further research of HSPB7.
