Abstract:Genetic variation between three Oryza rufipogon populations from Chafing of Hunan Province in China was surveyed at DNA level by analysis of RAPD fragments. The three populations were identified as the population PH that was a re-introduced population from ex situ conserved plants of O. rufipogon, the population PO that was a natural population with limited disturbances from human activities, and the population PI that was a population at the vicinity of rice fields. Twenty-four RAPD primers with ideal amplified DNA products screened from 384 primers were used for analysis of the genetic variation. A total of 170 fragments were scored and 78.82% of them were polymorphic. The total gene diversity (Ht) of the three O. rufipogon populations was 0.227, indicating high level of genetic variation of O. rufipogon in the Chaling populations. The three populations all showed relatively high level of within-population genetic diversity. Among them, the population PI has the highest level of genetic diversity (Ae = 1.364, PPB = 64.12%, He = 0.308, and Ht = 0.215, respectively), the population PR has the lowest levels of genetic diversity (Ae = 1.268, PPB = 48.24%, He = 0.301, and HI = 0.156, respectively), and the population PO has the intermediate level of genetic diversity (Ae = 1.342, PPB = 58.23%, He = 0.285, and Ht = 0.206, respectively). The results suggested that introgression between cultivated rice and O. rufipogon probably altered the genetic structure of the wild rice. The rev-introduced population PR restored 75% of the original genetic variation, compared with the original population PO, indicating that the collection of germplasm or in situ conservation of o. rufipogon should include as much genetic variability as possible. The genetic differentiation (Gst) of the Chaling O. rufipogon populations was 0.155 and the gene flow (Nm) was 2.72, showing low level of differentiation among populations. In addition, a high level of genetic variability (80.19%) was found within populations from AMOVA analysis. This was probably attribute to relatively high outcrossing rate (23.3%) of the Chaling O. rufipogon. This study suggested that in situ conservation site of o. rufipogon should be separated from cultivated rice with a distance long enough to avoid the introgression of cultivated rice, and that re-introducing O. rufipogon to its original site was an effective approach to its in situ conservation.
