Abstract:Lactoferrin is a multifunctional, iron-binding glycoprotein found in physiological fluids of mammals. Many functions of lactoferrin are thought to be involved in the N-lobe. In the present study, a gene encoding the N-lobe of porcine lactoferrin was cloned and expressed in Pichia methanolica. The gene contains 1038 bp, an “AGG” codon between 1039-1041bp in the PLF gene was replaced with a “TAA” stop codon through primer design and PCR. The sequencing results were aligned with the sequence (Genbank, L77887), indicating that the homology between these two sequences was 99%. The porcine lactoferrin N-lobe (PLfN) was expressed successfully in Pichia methanolica, and was secreted into the culture medium. The protein translated from the new ORF contained 346 amino acid residues, including a 17-amino acid signal peptide, its MW was approximately 42 kD, and it possessed antibacterial activity toward Escherichia coli ATCC25922. The method established in our study will pave the way for efficient industrial production of recombinant PLfN on a large scale for future utilization of this protein as a feed additive.
Optimization of the Fermentation Conditions of Lipase from Bacillus sp.ZJU318 and Characterization of the Enzyme

Abstract:The main factors affecting the lipase production by Bacillus sp.ZJU318 were studied by the method of uniform design.The results showed that the optimal fermentation medium for lipase production was: 3.28% of soybean powder,0.023% of sucrose,0.075% of K2HP04.Under the optimal fermentation conditions 28℃ and initial pH 8～9,the maximal lipase activity could reach 24.7U/mL.The enzyme activity was maximum at pH 9.0 and was stable over a pH range of 7.0-10.0,and the optimum temperature for enzyme reaction was 45℃.Little activity loss(6.2%) was observed after 1h of incubation at 40℃.However,the stability of the lipase decreased sharply at 50℃ and 60℃.

Abstract:The purpose of this study was to optimize the formulation of lactoferrin nano-liposomes using response surface methodology. Response surface methodology based on central composite rotatable design has been successfully used to model and optimize biochemical and biotechnological processes. The mass ratio of phosphatidylcholine and cholesterol (1-8), lactoferrin concentration (2-15 mg/mL), tween80 (0.5-2.5 mg/mL) and the volume ratio of organic phase and aqueous phase (2-10) were selected as independent variables with encapsulation efficiency and particle size as dependent variables. For each response, a 2nd-order polynomial model was developed using multiple linear regression analysis. Applying a desirability function method the optimum parameters were: phosphatidylcholine to cholesterol mass ratio of 5.77, lactoferrin concentration of 11.71 mg/mL, tween80 of 1.18 mg/mL and organic phase to aqueous phase volume ratio of 4.59. At this optimum point, particle size and encapsulation efficiency were found to be 214 nm and 60.21%, respectively. Furthermore, leakage ratio of nano-liposomes was used to determine the influence of storage period. © Maxwell Scientific Organization, 2012.

Abstract:ε-Poly-L-lysine (ε-PL) is a novel food biopreservative with broad antimicrobial activity. In the present study, single-factor experiments were applied to screen optimal carbon and nitrogen sources that affect the yield of a ε-PL-producing strain, Streptomyces violaceusniger, which was obtained in our laboratory recently. Box-Behnken design and response surface methodology (RSM) were adopted to derive a statistical model for optimizing the composition of the fermentation medium. The results showed that the main factors that affect ɛ-PL production were glucose, carbon source; peptone, organic nitrogen source; and (NH(4))(2)SO(4), inorganic nitrogen source. The optimum fermentation medium for ɛ-PL production of S. violaceusniger consisted of (g/L) glucose, 62.49; peptone, 5.71; (NH(4))(2)SO(4), 11.19; MgSO(4), 0.5; FeSO(4), 0.03; ZnSO(4), 0.04; KH(2)PO(4), 1.36; and Na(2)HPO(4).12H(2)O, 3.58. When cultured in the optimum medium, the ε-PL production was an average of 0.349 ± 0.025 mg/mL, which was 1.75 times higher than the initial medium.

Abstract:The objective of the current study was to optimize the fermentation conditions of producing fibrinolytic enzyme through single factor experiment and response surface experiment.The optimal fermentation conditions were obtained as follows:the carbon source was bran,the nitrogen source was yeast extract,the inorganic salt was 0.1% MgSO4,the solvent of medium was bean juice,pH was 4.0.Using 2% bran,2% yeast extract,0.1% MgSO4 as the major factor,choosing the right step,designing response surface experiment,the second-order regression model was obtained fitting the experimental data results.The R2 of the model was 0.8709,and was significant at the 90% of the probability.The second-order model predicted that under the conditions of 2.97% bran,2.30% yeast extract paste,0.147% MgSO4,pH 4,the highest fibrinolytic activity predicted was 410 IU/mL.

Abstract:The multifunctional antibacterial peptide lactoferricin-lactoferrampin (LFCA) is derived from bovine lactoferrin. Optimization of the fermentation process should be studied since different microorganisms have their own favorable conditions and processes for growth and the production of metabolites. In this study, the culture conditions of a recombinant strain, pPG-LFCA-E/LR-CO21 (LR-LFCA), expressing LFCA was optimized, utilizing the high-density fermentation process to augment the biomass of LimosiLactobacillus reuteri and the expression of LFCA. Furthermore, an assessment of the protective effect of LR-LFCA on intestinal inflammation induced by lipopolysaccharide (LPS) was conducted to evaluate the impact of LR-LFCA on the disease resistance of piglets. The findings of this study indicate that LR-LFCA fermentation conditions optimally include 2% inoculation volume, 36.5 °C fermentation temperature, 9% dissolved oxygen concentration, 200 revolutions/minute stirring speed, pH 6, 10 mL/h glucose flow, and 50% glucose concentration. The inclusion of fermented LR-LFCA in the diet resulted in an elevation of immunoglobulin levels, significant upregulation of tight junction proteins ZO-1 and occludin, reinforcement of the intestinal barrier function, and significant amelioration of the aberrant alterations in blood physiological parameters induced by LPS. These results offer a theoretical framework for the implementation of this micro-ecological preparation in the field of piglet production to enhance intestinal well-being.

Abstract:Objective To investigate the fermentation conditions of gene engineering E.coli strain for expression of Helicobacter pylori adhesin (N acetylneuraminyl lactose binding haemagglutinin, HpaA). Methods The expression of recombinant fusion protein was analyzed for optimizing the composition of the media, the range of pH, induction time, and other factors in fermentation task. Results Under the optimal fermentation conditions, the cell output and expression rate of HpaA reached 48.2 g/L and 31.5%, respectively. Conclusion Both the recovery rate of gene engineering E.coli strain and expression level of HpaA can be improved under the fermentation condition.

Abstract:Using ordinary nutrient medium as control,Lactobacillus acidophilus was cultured with different carbon source,nitrogen source,and pH.The result showed that the growth of L.acidopholus was greatly influenced by the pH of the culture.Higher pH value than 3.8 might inhibit the growth of L.acidopholus.The growth of L.acidopholus also was restricted by the carbon source and nitrogen source ratio.Higher content of nitrogen source led to excessive growth and lower accumulation of the products,while lower nitrogen source content led to slower growth of the cells.And insufficient carbon source might cause senility and atuolysis of the cells.The best culture conditions were determined as follows:2%glucose,2%peptone,0.1%Nacl,0.001%MnSO4,0.02%MgSO4,0.05%CH3COONa and pH6.0.

Abstract:Plackett-Burman design was used to evaluate the inportance of fermentation medium factors of Bacillus subtilis BSD-2 affecting the inhibitory zone diameter of produced antibacterial peptides. Subsequendy, 7 steepest ascent experiments involving 3 most important factors (i.e., peptone, starch and soybean meal) were conducted for approaching their optimal response region.Finally, the optimal fermentation medium formula was obtained by optimizing the most important factors using central composite design combined with response surface methodology. Results indicated that the optimal fermentation medium formula consisted of 14.29 g/L peptone, 14.07 g/L amylum, 6.49 g/L soybean power, 2.0 g/L CaCO_3 and 1.0 g/L MgSO_4. There was a 1.77-fold increase in the yield of antibacterial peptides obtained using the optimal fermentation medium, compared with that before optimization.

Abstract:In this study, culture conditions, including dissolved oxygen (DO) content, presence of osmoprotectants, residual glucose concentration, and ammonium sulfate-feeding strategies, were investigated for decreasing the inhibition effects of acetic acid, ammonium, and osmotic stress on l-lysine fermentation by Escherichia coli. The results revealed that higher DO content and lower residual glucose concentration could decrease acetic acid accumulation, betaine supplementation could enhance osmotic stress tolerance, and variable speed ammonium sulfate-feeding strategy could decrease ammonium inhibition. Thus, with 25 % DO content, 0–5.0 g/L of residual glucose concentration, and 1.5 g/L of betaine supplementation, 134.9 g/L of l-lysine was obtained after 72 h of culture, with l-lysine yield and productivity of 45.4 % and 1.9 g/(L · h), respectively.

Abstract:Strains producing β-glucosidase were isolated by solid medium containing esculin and ferric.Then six strains having high β-glucosidase activity were selected with p-nitropheny l-β-D-glucopyranoside as substrate.Strain having high ginsenoside bioconversion was finally chosen with Rbl as substrate and converted products were determined by TLC and HPLC.β-glucosidase activity of this strain was 0.803 U/mL and ginsenoside Rd conversation rate was 97.234 2％.The selected strain was identified as Saccharomycopsisfibuligera based on the morphology and nucleotide sequence analysis of the 18S rDNA.Fermentation medium was optimized to contain lactose as carbon source and corn flour as nitrogen source,and Ca2+ as metal ion.Finally inducer was optimized to promote the ginsenoside bioconversion.Results indicated that inducer had little impact on the bioconversion but rutin,a kind of inducer,could promote the bioconversion of Rd to CK.The selected strain with high ginsenoside bioconversion had good potential application.
Optimization of Nutrient Factors in Medium for Lactobacillus Fermentum Growth

Abstract:In this study, the optimal basal medium and the optimal carbon source, nitrogen source and growth factor in it were chosen for Lactobacilus fermentum growth, and the optimal addition amounts of the three nutrient factors were investigated using orthogonal array design. Results indicated that MRS medium was the most suitable for Lactobacilus fermentum growth, and its optimal formula consisted of 2.0% glucose, 0.5% beef extract, 0.5% casein peptone, 2% corn syrup, and other unchanged basic ingredients. The number of living Lactobacilus fermentum cells was over 1010 CFU/mL after 16 h to 18 h of cultivation in optimized MRS medium at 37 ℃, which exhibited a 10-fold increase compared with that in original MRS medium.

Abstract:Bovine lactoferrin peptide has been shown to be a broad-spectrum antimicrobial peptide. Based on the relationship between the structure and function of antimicrobial peptides, the antimicrobial peptide databases and protein analysis software were used to optimize the design of bovine lactoferricin peptide (LfcinB). The designed bovine lactoferricin-derived peptide (LfcinBD) gene fragment was inserted into a pPIC9K-His plasmid to construct a recombinant expression vector. After linearization of the Recombinant plasmid, Pichia pastoris GS115 cells were transfected with linearized recombinant plasmid by using electroporation and LfcinBD gene expression was induced with methanol. After the fermentation, supernatant was separated by low-temperature high-speed centrifugation. Ultrafiltration and freeze drying of the fermentation supernatant were performed, purified. Experimental results showed that the LfcinBD had stronger bacteriostatic activity against Staphylococcus aureus than the natural bovine lactoferrin peptide (LfcinB) produced under the same fermentation conditions. The effective expression of the optimized bovine lactoferricin-derived peptide was detected using SDS-PAGE electrophoresis. This study lays the foundation for further exploration to improve the biological activities of antimicrobial peptides.

Abstract:This experimental work was primarily on the fermentation techniques of lactic acid,trying to enhance the production of lactic acid by mutation of the strain and optimization of the culture medium.The final concentration of L-lactic acid in the broth was 18.5 g/L using the basic medium with the original strain,and the conversion rate based on glucose was 50.27%.A high-yield strain was screened out by laser mutation from original strain,which was named DBLB28.The culture medium was optimized after screening out the high-yield strain.Several factors that had important effects on lactic acid fermentation were chosen by orthogonal experiment design.Then they were optimized by response surface methodology(RSM),and the regression equation was proved to be effective after analysis of variance.Finally,the optimized condition was as follows:glucose concentration 68.6 g/L,beef extract concentration 22.4 g/L,peptone concentration 23.2 g/L.The concentration of lactic acid production was 29.0 g/L from the regression equation,and the data from experiment was 29.4 g/L.

Abstract:Objective To optimize the fermentation procedure of recombinant human granulocyte colony-stimulating factor and to obtain high cell density and high level expression of the aim protein.Methods With orthogonal experiment,study the influence of different concentration potassium,sodium salt and magnesium salt on plasmid stability and expression of aim protein in a 14L fermentor.Results Under high concentrations of salt in the culture,the specific growth rate of strains and plasmid stability were significantly improved,so as to improve the purpose of protein expression.Conclusion Gain a efficient fermentation method to produce recombinant human granulocyte colony-stimulating factor.

Abstract:Lactobacillus acidophilus strains have limiting factors such as low cell density and complex nutritional requirements in industrial production, which greatly restricts their industrial application. In this study, fermentation conditions for L. acidophilus were optimized and transcriptomic analysis used to understand growth mechanisms under high-density fermentation conditions. We found that L. acidophilus IMAU81186 has strong tolerance to gastrointestinal juice. In addition, its optimal culture conditions were 3% inoculum (v/v); culture temperature 37 °C; initial pH 6.5; and medium composition of 30.18 g/L glucose, 37.35 g/L soybean peptone, 18.68 g/L fish peptone, 2.46 g/L sodium citrate, 6.125 g/L sodium acetate, 2.46 g/L K2HPO4, 0.4 g/L MgSO4·7H2O,0.04 g/L MnSO4·5H2O, 0.01 g/L serine, and 0.3 g/L uracil. After optimization, viable counts of IMAU81186 increased by 7.03 times. Differentially expressed genes in IMAU81186 were analyzed at different growth stages using transcriptomics. We found that a single carbon source had limitations in improving the biomass of the strain, and terP and bfrA were significantly down-regulated in the logarithmic growth period, which may be due to the lack of extracellular sucrose. After optimizing the carbon source, we found that adding 12 g/L sucrose to the culture medium significantly increased cell density.

Abstract:Human lactoferrin (HLF), an essential nutrient found in breast milk, possesses antibacterial, anti-inflammatory, and immune-enhancing properties. In this study, the effects of three constitutive promoters (P21, P43, and Pveg) and three inducible promoters (Pgrac100, PxylA, and Ptet*) on the expression of HLF were compared using Bacillus subtilis G601 as the host strain. The results showed that the highest expression of HLF, reaching 651.57 μg/L, was achieved when regulated by the Ptet* promoter. Furthermore, the combinational optimization of ribosome binding site (RBS) and signal peptides was investigated, and the optimal combination of RBS6 and SPyycP resulted in increased HLF expression to 1 099.87 μg/L, with 498.68 μg/L being secreted extracellularly. To further enhance HLF secretion, the metal cations-related gene dltD was knocked out, leading to an extracellular HLF level of 637.28 μg/L. This study successfully demonstrated the secretory expression of HLF in B. subtilis through the selection and optimization of expression elements, laying the foundation for the development of efficient B. subtilis cell factories for lactoprotein synthesis.

Abstract:The main indicators for industrial production of high-quality lactic acid at elevated temperatures are high titer, productivity, yield, and optical purity. However, no such strains have been reported to meet all these requirements simultaneously. In this study, a high optical purity L-lactic acid producing strain is developed through the CRISPR-Cas9 gene editing platform. Further, adaptive evolution was used to breed and select a high-performance strain (NCBIO01-M2-ldhL1-HT) that could efficiently produce L-lactic acid at a high temperature of 45℃. This strain produced 221.0 g/L of L-lactic acid in open fermentation with high initial glucose concentration. Also, L-lactic acid productivity and yield was above 7.5 g/L/h and 0.96 g/g respectively, as well as the optical purity of L-lactic acid in the fermentation broth exceeded 99.1%. In short, this breeding strain possess high potential to be considered for the commercial production of polymer-grade L-lactic acid.

Abstract:As a common expression host, Escherichia coli has received more and more attention due to the recently developed secretory expression system, which offers advantages like reduced downstream bioprocesses and improved product quality. These advantages, coupled with high-density fermentation technology, make it a preferred system for large-scale production of many proteins utilized in industry and agriculture at a reduced process cost. To improve the secretion efficiency of target proteins, various strategies, including signal peptide optimization, periplasmic leakage, and chaperones co-expression have been developed. In addition, the optimization of the fermentation conditions such as temperature, inducer, and medium were also taken into account for the extracellular production in the high-density fermentation to reduce the cost of production. Here, these strategies ranging from genetic engineering to fermentation optimization were summarized for the future guidance of extracellular production of recombinant proteins using E. coli.
