Abstract:OBJECTIVE To study the flavonoids of Artemisia anomala S. Moore. METHODS The compounds were isolated by various column chromatography, and their structures were elucidated on the basis of spectral analysis and physical-chemical properties.RESULTS Eleven flavonoids were obtained and identified as isorhamnetin(Ⅰ), kaempferol(2), eupatorin(3), luteolin(4), apige-nin (5), quercetin (6) , cosmosiin (7), kaempferol-3-ο-α-L-rhamnopyranoside (8), luteolin-7-ο-β-D-glucopyranoside (9), quercetin-3-ο-α-L-rhamnopyranoside(10) and avicularin(11). CONCLUSION All the compounds were obtained from this plant for the first time, in addition, compound 8 and 11 were obtained from this genus for the first time.

Abstract:Fourteen known compounds were isolated from the aerial parts of Artemisia anomala and identified as follows:zhepiresinol(1),(-)-lariciresinol(2),ficusal(3),medioresinol(4),(+)-syringaresinol(5),1β,2β-epoxy-3β,4α,10α-trihydroxyguaian-6α,12-olide(6),5β,10α-dihydroxy-1αH-dehydroleucodin(7),3α-chloro-8α-acetoxy-4β,10α-dihydroxy-1β,2β-epoxy-5α,7αHguai-11(13)-en-12,6α-olide(8),13-O-desacetyleudesma-afraglaucolide(9),1,10-seco-4ξ-hydroxy-muurol-5-ene-1,10-diketone(10),1α-hydroxyafraglaucolide(11),aurantiamide(12),indole-3-aldehyde(13),loliolide(14).All these compounds were isolatedfrom this plant for the first time and compounds 1-4,8,10,13 were obtained from genus Artemisia for the first time.

Abstract:AIM: To study chemical constituents of the aerial parts of Artemisia anomala (Asteraceae). METHODS: The constituents were isolated with silica gel, ODS column chromatography and semi-preparative HPLC, and their structures were elucidated on the basis of physical characteristics and spectral data. RESULTS: Eight compounds were obtained, and their structures were identified as 3 beta-ethoxytanapartholide (1), (4S*, 5S*)-dihydro-5-[( 1R*, 2S*)-2-hydroxy-2-methyl-5-oxo-3-cyclopenten-1-yl]-3-methylene-4-(3-oxobutyl)-2(3H)-furanone (2), ligucyperonol (3), cyperusol C (4), santamarin (5), 1 alpha, 2 alpha, 3 alpha, 4 alpha, 10 alpha -pentahydroxyguaia-11(13)-ene-12, 6 alpha-olide (6), balanophonin (7), methyl 3-(2'-hydroxy-4'-methoxyphenyl) propanoate (8). CONCLUSION: Compound 1 was a new artifact, 4 and 8 were isolated from the genus Artemisia for the first time, and compounds 2-3, 5-7 were isolated from this plant for the first time.

Abstract:Four novel cycloartanes, named sphaerophysone A (1), B (2), C (3) and D (4), were isolated from the ethanol extract of Sphaerophysa salsula (Pall.) DC. The structures were elucidated on the basis of spectral evidence, and the stereochemistry of compound 1 was defined by X-ray crystallographic analysis. Sphaerophysone B (2) may be an artifact formed in the isolation procedure.

Abstract:Two new eudesmane sesquiterpenoids artanoate (1) and eudesmanomolide (2) were isolated from the aerial parts of Artemisia anomala S. Moore. Their structures were elucidated as methyl (4R, 5S, 6S, 7S, 10R)-1-oxo-4, 6-dihydroxy-eudesma-2, 11 (13)-dien-12-oate (1) and (1R, 5R, 6R, 10R)-3, 13-diacetoxy-1-hydroxy-3, 7(11)-diene-12, 6-olide (2) on the basis of extensive spectroscopic analyses. Compound 1 showed cytotoxicity against HCT-8 cell lines with IC50 value of 9.13μM, and compound 2 exhibited inhibitory activities against HCT-8 and A549 cell lines with IC50 values of 3.76 and 5.49μM, respectively.

Abstract:Four previously unreported sesquiterpenoid diasteromers, arteannoides U–X (1–4), together with one new norsesquiterpenoid 5 (arteannoide Y) and one undescribed rearranged cadinene sesquiterpenoid 6 (arteannoide Z) were obtained from the dried aerial parts of Artemisia annua (Qinghao). Notably, arteannoides U–X (1–4) are four stereoisomers that possess the same molecules and the same planar connectivity, but differ from each other in configuration at a certain stereocenter. Their accurate structures were unambiguously identified and distinguished by extensive spectroscopic analyses, NMR calculations with DP4+ analysis, electronic circular dichroism (ECD) calculations and X-ray diffraction analyses. Compounds 1, 3, and 4 showed inhibitory activities against the production of inflammatory cytokines (PGE2, NO, IL-6 and TNF-α) in lipopolysaccharide (LPS)-induced RAW 264.7 macrophages.

Abstract:The present study is to investigate the chemical constituents from the aerial parts of Artemisia myriantha. The chemical constituents were isolated by column chromatographies over silica gel, Sephadex LH-20, ODS and Semi-prep HPLC, and the structures were identified by NMR and MS data. Thirteen known compounds were isolated and identified as: blumenol A (1),(+)-dehydrovomifoliol (2),(+)-3-hydroxy-β-ionone (3),(3R, 6R, 7E)-3-hydroxy-4, 7-megastigmadien-9-one (4),(-)-10-oxo-isodauc-3-en-15-oic acid (5),isoerivanin (6),eudesmafraglaucolide (7), artanomalide A (8),13-acetoxy-3β-hydroxy-germacra-1(10) E,4E,7(11)-trien-12,6α-olide (9),13-acetoxy-3β-tigloyl-germacra-1(10) E, 4E, 7(11)-trien-12, 6α-olide (10),13-acetoxy-3β-(3-methylbutanoyl)-germacra-1(10)E, 4E, 7(11)-trien-12, 6α-olide (11),3,9-diacetoxy-13-hydroxy-1(10), 4, 7(11)-germacratrien-12,6α-olide (12), and 8α-angeloyloxycostunolide (13). Compounds 1-6 and 13 were obtained from the genus Artemisia for the first time, and 7-9 and 12 were isolated from this plant for the first time. Compound 8 exhibited selective cytotoxicity against human colon cancer (HCT-8) and human gastric cancer (BGC-823) with IC₅₀ values of 2.33 and 4.53 μmol•L ⁻¹, respectively.

Abstract:A novel dimeric guaianolide with an unprecedented skeleton, named artanomadimer A (1), and five new analogues, artanomadimers B–F (2–6), were isolated from the aerial parts of Artemisia anomala. Their structures and stereochemistry were elucidated by extensive spectroscopic methods, and the absolute stereochemistry of compound 4 was confirmed by X-ray crystallographic analysis. Artanomadimer A (1) is probably formed through a Diels–Alder reaction with the new carbon–carbon bond formation of C-11/C-2′ and C-13/C-5′ based on its structure. A cytotoxic evaluation showed that compounds 1 and 6 exhibited significant inhibitory effects against the cell growth of BGC-823 tumor cell lines with IC50 values of 2.71 and 6.25μM, respectively.

Abstract:Thirteen new sesquiterpenoids, arteannoides F-R (1–13), along with 13 known analogues (14–26), were isolated from the dried aerial parts of Artemisia annua L. Their structures, including absolute configurations, were unambiguously determined by a combination of physical data analyses (HRESIMS, 1D and 2D NMR, and ECD) as well as the crystal structures of 1, 5, 6, 15, 19, and 23. Among the isolated compounds, 1 features an unusual 11-oxatricyclo[6.2.1.04,9]undecan-2-ene ring system, 5 possesses an uncommon 4,11-ether bridged tricyclic framework, whereas 6 is a new eudesmane-type sesquiterpenoid formed via rearrangement of its carbon backbone. The systemically anti-inflammatory activities of all isolates were evaluated by measuring their inhibitory effects on PGE2, NO, TNF-α, and IL-6 production in LPS-stimulated RAW 264.7 macrophages. Moreover, the structure activity relationships of some compounds are summarized, this study will provide new structural templates for discovering potential anti-inflammatory agents.

Abstract:Four new polyketides including two arthrinic acid derivatives (1-2), one phenolic derivative (3) and (S)-3-hydroxy-6-(2-hydroxypropyl)-5-methyl-2H-pyran-2-one (4) along with one methyl ester of arthrinic acid (5) were isolated from the culture broth of Arthrinium sp., which was an entophytic fungus of clam worm. Their structures were identified on the basis of HR-ESI-MS and NMR spectral analyses together with advanced Mosher's method. In the assay of inhibiting the prostate cancer PC3 cell line, none of the isolated compounds showed significant cytotoxicity.

Abstract:In this study, eight undescribed sesquiterpenoids (artemvulactone A-G and artemvulemdiol A), and two undescribed triterpenoids, (3S)-dammar-20,25-diene-3-hydroxy-24-one and (3S,23E)-dammar-20,23-diene-25- methoxy-3-ol were isolated from the leaves of Artemisia vulgaris L., together with ten known sesquiterpenoids and three known triterpenoids. The structures of these undescribed terpenoids were determined by extensive spectroscopy methods, including 1D and 2D-NMR, HRESIMS, IR, UV, X-ray diffraction, and ECD. The absolute configurations of artemvulactone A, artemvulactone D, and artemvulactone E were determined by X-ray diffraction (Cu Kα). All isolates were evaluated for their anti-inflammatory efficacy by detecting the expression of inflammatory mediator NO in LPS-induced RAW264.7 cells, and the results indicated that artemvulactone E exhibited significant anti-inflammatory effect with an IC50 value of 0.9 ± 0.2 μM. Furthermore, artemvulactone E could reduce LPS-induced COX-2 protein expression dose-dependently by western blotting experiments.

Abstract:In the current study, further chemical investigation of the fruiting bodies of Fomes officinalis led to isolate seven new 24-methyl-lanostane triterpenoids, named officimalonic acids I−O (1–7). Their structures were elucidated based on the analysis of spectroscopic data (HR-MS, 1D and 2D NMR, UV, IR). Compounds 1−3 possessed an unusual C-23 spirostructure moiety, while compounds 4−7 had 23,26-lactone unit. Anti-inflammatory assay revealed that compounds 3 and 5 exhibited significant inhibitory activities against NO production in LPS-induced RAW 264.7 cells and cyclooxygenase (COX-2).

Abstract:A phytochemical reinvestigation of the aerial parts of Artemisia sieversiana gave a new guaianolide and two known flavones (chrysosplenetin and 5-hydroxy-3',4',6,7-tetramethoxyflavone). Antifungal fractions derived from the chloroform extract of A. annua afforded two cadinane derivatives (arteannuin B and artemisinin), oleanolic acid, beta-sitosterol, stigmasterol, and the four flavones artemetin, bonanzin, eupalitin and chrysosplenetin. Their structures were elucidated by spectral methods. All isolates from the two species were tested in vitro for antifungal activity. Arteannuin B, a main sesquiterpenoid in A. annua, showed antifungal activity against one human (Candida albicans, MIC: 100 micrograms/ml) and four plant pathogenic fungi (Gaeumannomyces graminis var. tritici, Rhizoctonia cerealis, Gerlachia nivalis and Verticillium dahliae, MICs: 150, 100, 150 and 100 micrograms/ml, respectively) whereas others showed no antifungal activity. The MIC value of ketoconazole to C. albicans was 1.0 microgram/ml, and those of triadimefon to G. graminis var. tritici and R. cerealis 150 and 100 micrograms/ml.

Method: The chemical constituents were isolated by repeated silica gel chromatography, medium pressure column chromatography, and semi-preparative HPLC, and their structures were elucidated by spectroscopic analyses and comparison of NMR data with those reported in literature. Result: 15 compounds were isolated and identified to be 5-O-[(E)-Caffeoyl] quinic acid(l), 1,3-di-O-caffeoylquinic acid(2), 4 5-di-O-caffeoylquinic acid(3), 3, 5-di-O-caffeoylquinic acid (4), 3, 4-di-O-caffeoylquinic acid (5), methyl-3,4-di-O-caffeoylquinic acid(6), methyl-3,5-di-O-caffeoylquinic acid(7), 3,6'-O-diferuloylsucrose(8), 5'-β-D-glucopyranosyloxyjasmonic acid(9), Scopoletin(10), scoparone (11), 4-O-β-D-glucopyranosyl-2-hydroxyl-6-methoxyacetophenone (12), chrysosplenol D (13), casticin (14), chrysosplenetin(15). Conclusion: Compounds 2, 6, 8 and 9 are obtained from the Artemisia genus for the first time. Compounds 7 and 15 are obtained from this plant for the first time.

Abstract:Two new dimeric compounds of the alternariol class, (±)-alternarlactones A () and B (), were isolated along with 11 known compounds from the fungus P1210. Their structures were elucidated with the assistance of long-range HSQMBC to address inadequate cross-peaks in HMBC that result from the highly dense quaternary carbons, as well as theoretical calculations. All isolated altenuisol derivatives were screened for their antiparasitic activities, which provide a preliminary structure-activity relationship of this class of compounds against neglected tropical diseases.
Anti-inflammatory discovery of sesquiterpenoids and a jasmonic acid derivative from Artemisia stolonifera

Abstract:Eleven previously undescribed sesquiterpenoids (8-18), one undescribed jasmonic acid derivative (35) and 28 known compounds were isolated from the leaves of Artemisia stolonifera. Undescribed compounds with their absolute configurations were determined by extensive spectroscopic analysis, single-crystal X-ray diffraction and ECD calculation. Compound 8 was identified as a rare sesquiterpenoid featuring a rearranged 5/8 bicyclic ring system, whereas compound 17 was found to be an unprecedented monocyclic sesquiterpenoid with methyl rearrangement. Evaluation of biological activity showed that compounds 1-5 and 7 displayed cytotoxicity against six tumor cells. In the meantime, compounds 11, 12, 18 and 35 exhibited inhibitory effects against LPS-stimulated NO production in RAW 264.7 macrophage cells and reduced the transcription of IL-6 and IL-1β in a dose-dependent manner at 25, 50 and 100 μM. Moreover, the anti-inflammatory-based network pharmacology and molecular docking analyses revealed potential target proteins of 11, 12, 18 and 35.
