Effect of angiogenesis on the mild hypothermia in cerebral ischemia in rat

Abstract:Objective To investigate the effects of mild hypothermia on the angiogenesis in cerebral ischemia and study the mechanism of mild hypothermia in cerebroprotective effect.Methods The twenty SD rats were assigned into normal temperature group and mild hypothermia group (each group n=10),following establishment of a permanent middle cerebral artery occlusion (MCAO) model with nylon suture embolization. The rats of normal temperature group were fed in the room temperature, and the temperature of the rats of mild hypothermia group was controlled at the range 33~35 ℃ after two houroperation and persisted 72 h. Seven days later, the rats were sacrificed. The infarct volume was measured by 2% TTC staining technique, then the vascular density were measured by immumohistochemistry methods.Results The infarct volume of mild hypothermia group[( 153.25 ± 23.14 )mm3] was significantly smaller than that in the normal temperature group[( 253.45 ± 36.21 )mm3](P< 0.01 ).Compared with the normal temperature group, expression of VEGF protein in the mild hypothermia group decreased ( 36.07 /HPF vs 24.02 /HPF )(P< 0.01 ) and the numbers of vessels of the two groups were no significantly difference( 39.19 /HPF vs 39.96 /HPF)(P> 0.05 ). Conclusions Mild hypothermia can decrease the infarct volume, but it has no effect on angiogenesis.
