Hypoxia-induced gene expression of receptor Tie2 in endothelial cells cultured from human cerebral arteriovenous malformation

Abstract:Objective To investigate the gene expression of receptor Tie2 in cultured endothelial cells from human cerebral arteriovenous malformations ( cAVM) under hypoxic conditions. Methods The cAVM endothelial cells were growed by seeding vascular tissue blots and culturing for 60 hours and purified with cell - scraping technique. The expression of factor Ⅷ-relative antigen (FⅧ-RA) in cultured cells was observed with immunocytochemistry. The level of Tie2 protein and mRNA in cells were determined by immunocytochemistry and RT-PCR analysis respectively when they were exposed to hypoxic conditions ( 95% N2,5% CO2; two hours, four hours, eight hours) or maintained in basal condition. Results Under inverted microscope the cultured cells showed contact inhibition and a rounded cobblestone appearance. More than 95% of them were stained strongly with antibodies against FⅧ-RA. Significant Tie2 mRNA and protein accumulated when these cells were exposed to hypoxia . Conclusion These findings suggested hypoxia may induce gene expression of receptor Tie2 in endothelial cells at the transcriptional level, which may play an important role in the pathogenesis and development of vascular malformations in central nervous system.
