Isolation and Identification of the SCQ Strain of Porcine Reproductive and Respiratory Syndrome Virus

Abstract:The obvious CPE could not be seen until the fourth passage when Mark-145 cells were vaccinated with PRRSV positive serum, which were tested by ELISA kit(offered by IDEXX company).And when the cells were passaged blindly in the sixth passage,70%-80% cells showed obvious CPE in 2-3 days after infected by the suspicious stock.This indicated that PRRSV titres increased quickly. Virions could be observed in the infected cells under the electric microscope.The diameter of the virions and core is about 40-80nm and 20-40nm respectively.The TCID50 is10-4.46/0.05ml.The neutrinization index is 100 when the isolate strain met the specific PRRSV positive sera.So the isolate was designated PRRSV-SCQ strain.Based on the sequence of ATCC VR-2332 and RT-PCR, the ORF5 fragment was amplified following the templet of the total RNA extract of the infected cells by PRRSV-SCQ.Sequencing result showed that ORF5 fragment of PRRSV-SCQ is more similar to ATCC VR-2332 strain rather than LV strain in genetype.
