Abstract:The direct electrochemistry of cytochrome c at microband gold electrode modified with a new promoter 4,6-dimethyl-2-mercaptopyrimidine was investigated. The self-assembly monolayer of 4, 6-dimethyl-2-mercaptopyrimidine on the surface of gold electrode was characterized by IR spectrum and XPS. Cyclic voltammetric experiments indicated that the electrochemistry of cytochrome c at gold electrode modified with this promoter was quasi-reversible with one-electron transfer reaction. The heterogeneous electron-transfer rate constant, k(s), of cytochrome c was evaluated to be ca.6.6x 10(-3)cm/s. The effects of the immersion time and the oxygen in air on the promoter to promote the direct electrochemistry of cytochrome c were discussed.

Abstract:We have successfully constructed a novel gold film with open interconnected macroporous walls of nanoparticles by combining the hydrogen bubble dynamic template synthesis with galvanic replacement reaction. After modified by a self-assembled monolayer (SAM) of 11-mercaptoundecanoic acid (MUA), the three-dimensionally (3D) interconnected macroporous Au film has been used as a biocompatible substrate for the immobilization of cytochrome c. The morphology, structure and electrochemical features of the modified and unmodified macroporous Au films were characterized by field-emission scanning electron microscopy (FESEM), energy-dispersive X-ray (EDX), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The results reveal that the resultant films had a large electroactive surface area for high protein loading, enhanced electron transfer of cytochrome c, retained electrochemical activity, good stability and repeatability. And the excellent electrochemical behaviors could be attributed to the hierarchical structure of the macroporous Au film constructed by nanoparticles.

Abstract:The direct electrochemistry of cytochrome c (cyt c) on a gold electrode modified with 3-mercaptopropylphosphonic acid [HS–(CH2)3–PO3H2, MPPA] self-assembled monolayers (SAMs) was for the first time investigated. Electrochemical measurements and surface-enhanced infrared absorption spectroscopic reveal that the adsorption kinetics of cyt c on the MPPA-SAMs is very fast (saturation adsorption is completed within 5s) and the immobilized cyt c molecules retain their native secondary protein structure. The nature of interaction between cyt c and –PO3H2 groups is mainly the electrostatic interaction. The direct electrochemistry of the immobilized cyt c on the –PO3H2 terminated SAMs with short chain is nearly reversible. Its formal potential (E0′=18±3mV vs. SCE) is very close to that of cyt c in an aqueous solution (E0′=18–22mV vs. SCE). In addition, the electron transfer rate of cyt c immobilized on –PO3H2 terminated SAMs is relatively slow as compared to –SO3H and –COOH terminated SAMs, indicating excess negative charge density on the SAMs surface will decrease the electron transfer rate of cyt c.

Abstract:Electrochemical behavior of cytochrome c at a bis(4-pyridyl)disulphide modified gold electrodes covered with nanoporous alumina templates was investigated. The experimental results show that the nanoporous alumina templates promote further the direct electron transfer of cytochrome c on the modified gold electrode, resulting in a well-defined fast votammetric response.

Abstract:In this work, we design and synthesize 2,2'-(7,9-dimethyl-2,4,6,8-tetraoxo-6,7,8,9-tetrahydropyrimido[5,4-g]pteridine-1,3(2H,4H)-diyl)bis(N,N-bis(2-chloroethyl)acetamide) (PT-MCA) as a novel DNA intercalator and potential antitumor agent. Electrochemical analysis reveals the redox process of PT-MCA on the electrode surface. The bioelectrochemical sensors are obtained by modifying the surface of GCE with calf thymus DNA (ctDNA), poly (dG), poly (dA), and G-quadruplex, respectively. The DNA oxidative damage induced by PT-MCA is investigated by comparing the peak intensity change of dGuo and dAdo and monitoring the peaks of the oxidation products of guanine and/or adenine (8-oxoGua and/or 2,8-oxoAde). UV-vis absorption and fluorescence spectra and gel electrophoresis are further employed to understand the intercalation of PT-MCA into DNA base pairs. Moreover, PT-MCA is proved to exhibit stronger anti-proliferation activity than mitoxantrone against both 4T1 and B16-F10 cancer cells. At last, the oxidative damage of PT-MCA toward ctDNA is not interfered by the coexistence of ions and also can be detected in real serums.

Abstract:Colloidal gold modified carbon paste electrodes were prepared by mixing 24-nm-diameter colloidal Au particles with carbon paste. The modified electrodes displayed a low charging current and a favorable electrochemical response of hexacyanoferrate (111). The direct electrochemical behavior of a horse-heart cytochrome c (cyt.c) adsorbed on this electrode surface is described. It showed a surface-controlled electrode process with the electron transfer rate constant of (1.21 +/- 0.08) s(-1) and alpha of 0.67. Cyclic voltammograms showed small peak-to-peak separations at low scan rates. The adsorbed cyt.c maintained its activity and could also electrocatalyze the reduction of hydrogen peroxide. Since this behavior was quite pronounced the electrode was used for H2O2 detection. The Km,PP value for this sensor was found to be 2.28 +/- 0.17 mM, allowing measurements down to 0.01 mM H2O2.

Abstract:The nano-scale HAp was prepared with the precipitation method. Due to the particular multi-absorbing sites, hydroxyapatite can be used as a novel favorite promoter for the direct electrochemistry of cytochrome c (cytc c). Cyclic voltammogram of cyt c on the hydroxyapatite modified glassy carbon electrodes showed a pair of well-defined and nearly reversible peaks at about 0.074 V (vs. Ag/AgCl) in pH 7.0 PBS buffers, being the characteristic peaks of the cyt c heme Fe(III)/Fe(II) redox couples. The experimental results supported that the electrostatic interaction between cyt c and HAp gave a rapid one-electron diffusion-controlled electron transfer. It provided a good model for the study of redox processes in biological system. Spectral characterization verified the activity of cyt c retained. The effects of scan rate, ionic strength on the electrochemical behavior of cyt c were discussed in details.

Abstract:The monolayer of cytochrome c oxidase maintaining physiological activity and attached covalently to the self-assembled monolayers of 3-mercaptopropionic acid (MPA) on a gold electrode was obtained. The results of cyclic voltammetry show that direct electron transfer between cytochrome c oxidase and the electrode surface is a fast and diffusionless process. MPA has a dual role as both electrode modifier and the bridging molecule which keeps cytochrome c oxidase at an appropriate orientation without denaturation and enables direct electron transfer between the protein and the modified electrode. Immobilized cytochrome c oxidase exhibits biphasic phenomena between the concentration of the electrolyte and the normal potentials; meanwhile its electrochemical behavior is also influenced by the buffer components. The quasi-reversible electron transfer process of cytochrome c oxidase with formal potential 385 mV vs. SHE in 5 mM phosphate buffer solution (pH 6.4) corresponds to the redox reaction of cyt a3 in cytochrome c oxidase, and the heterogeneous electron transfer rate constant obtained is 1.56 s−1. By cyclic voltammetry measurements, it was observed that oxidation and reduction of cytochrome c in solution were catalyzed by the immobilized cytochrome c oxidase. This cytochrome c oxidase¦MPA¦Au system provides a good mimetic model to study the physiological functions of membrane-associated enzymes and hopefully to build a third-generation biosensor without using a mediator.

Abstract:The promotion of horse heart cytochrome c electrochemistry at a gold minigrid electrode in the presence of some amino acids in solution has been investigated by cyclic voltammetry. Of these, Cys, His, Met, Cis and Trp have proved to be effective in facilitating electron transfer between Cyt c and the gold electrode, and good, quasi-reversible redox reactions were achieved. Under alkaline pH conditions, the above amino acids also present a promotion effect on the electrochemistry of Cyt c, but a different cyclic voltammogram appears. The new appearance results from the stability of different conformation states of Cyt c at different pH regions. Trp shows a more pH dependent ability in promoting the electrochemistry of Cyt c at a gold electrode. Probably, the proton dissociation and steric factor of the bulky indole group account for its electrochemical behavior. Abstract:The direct electrochemistry of catalase (Ct) was accomplished at a gold electrode modified with single‐wall carbon nanotubes (SWNTs). A pair of well‐defined redox peaks was obtained for Ct with the reduction peak potential at −0.414 V and a peak potential separation of 32 mV at pH 5.9. Both reflectance FT‐IR spectra and the dependence of the reduction peak current on the scan rate revealed that Ct adsorbed onto the SWNT surfaces. The redox wave corresponds to the Fe(III)/Fe(II) redox center of the heme group of the Ct adsorbate. Compared to other types of carbonaceous electrode materials (e.g., graphite and carbon soot), the electron transfer rate of Ct redox reaction was greatly enhanced at the SWNT‐modified electrode. The peak current was found to increase linearly with the Ct concentration in the range of 8×10 −6 –8×10 −5 M used for the electrode preparation and the peak potential was shown to be pH dependent. The catalytic activity of Ct adsorbates at the SWNTs appears to be retained, as the addition of H 2 O 2 produced a characteristic catalytic redox wave. This work demonstrates that direct electrochemistry of redox‐active biomacromolecules such as metalloenzymes can be improved through the use of carbon nanotubes.

Abstract:A near-reversible cyclic voltammetric response of cytochrome b5 and its mutants, E44A, E56A and E44/56A, was observed at a gold electrode modified with thioglycolic acid. The electron transfer between the negatively charged protein and negatively charged electrode was promoted by the multivalent cations such as Mg2+ or Cr(NH3)63+ ions. When the protein solution was titrated by Mg2+ ion, the conditional reduction potential EO′ shifted to the positive direction and reached a plateau after Mg2+ ion was more than 20 mM. Under this condition, the EO′ of wild type cytochrome b5 and its mutants are −6 mV (vs. SHE, wild type Cyt b5), −7 mV (Cyt b5 E44A), −2 mV (Cyt b5 E56A), −3 mV (Cyt b5 E44/56A) respectively. The conditional reduction potentials of cytochrome b5 and its mutants obtained by spectroelectrochemical titration in the presence of 0.01 mM Ru(NH3)63+, pH 7.0, I = 0.1 M phosphate buffer are +5 mV (vs. SHE, wild type Cyt b5), +6 mV (Cyt b5 344A), +6 mV (Cyt b5 E56A), + 7 mV (Cyt b5 E44/56A) respectively. The results demonstrate that the mutagenesis at surface residues Glu44 and Glu56 does not alter the reduction potential of cytochrome b5 significantly. However, the studies on binding between cytochrome b5 and Mg2+ ion by the electrochemistry and NMR show that although the Mg2+ ion has the same interaction with these negatively charged residues and similar structural perturbation, the chelation of Mg2+ ions by the heme propionate appears a stronger influence on the heme, shifting the reduction potential of proteins.

Abstract:The pH-dependent redox processes of cytochrome c (cyt c) immobilized on a gold electrode that was coated with a self-assembled monolayer (SAM) of mercaptounadecanoic acid (MUA) were studied by electrochemical methods combined with quartz crystal microbalance (QCM) and surface enhanced infrared absorption (SEIRA) spectroscopy. Variation of the solution pH in the range from 4.0 to 10.0 determines the surface charge of the SAM, for which an apparent pK(a) of 6.0 was determined, whereas the structure of the electrostatically bound cyt c remains largely unchanged. Thus, the pH-dependence of the interfacial redox process reflects the electric-field control of cyt c immobilization which in turn has a pronounced impact on the electron transfer process. In the pH range between 7.0 and 4.0, the electrostatic interactions with the cationic protein are weakened due to the protonation of the carboxyl headgroups of the SAM such that the immobilized protein remains highly mobile and can rapidly adopt the orientation which is most favorable for electron transfer. Thus, the rate constant for direct electron transfer remains unchanged in this pH range, but it decreases upon increasing the pH above 7.0. The dramatic slowdown of the interfacial electron transfer is attributed to the increased strength of electrostatic binding which traps the protein in an orientation that is unfavorable for electron exchange with the electrode. The present study demonstrates that the solution pH is an important parameter that allows for optimizing interfacial electron transfer processes of electrostatically bound proteins.

Abstract:The electrochemistry of cytochrome C was investigated as a spectrographic graphite electrode. In phosphate buffer solution (pH = 7.0), cytochrome C showed stable and quasi-reversible response. The formal potential E0' was 0.015 V (at 25-degrees-C, vs. SCE) and the heterogeneous electron transfer rate constant k(s) obtained varied form 1.10 X 10(-3) cm . s-1 to 1.80 X 10(-3) cm . s-1. The thermodynamic parameters of the electron transfer reaction of cytochrome C was also estimated. Furthermore, the effect of the various electrode surface states on the electrochemistry of cytochrome C was discussed.

Abstract:Self-assembled monolayers (SAMs) of thiol-derivatized tetraphenylporphyrin (SH-TPP) and cobalt tetraphenylporphyrin (SH-CoTPP) have been prepared to examine the structure and direct electron transfer (ET) on a gold electrode in an aqueous solution. The cyclic voltammetry peaks of the porphyrin were obviously observed in aqueous solution. An interesting phenomenon was that direct ET cannot occur in phosphate-buffered solutions when a highly oriented and tightly packed monolayer is in existence, but the relatively low-density SH-TPP SAMs exhibited good electrochemical response. The electron transfer rate constants were obtained from the impedance spectra (Cole-Cole plot), upon application of different biasing potentials to the modified electrode. We propose a new kinetic model of the ET processes of SAMs of thiol-derivatized porphyrin and apply the density functional theory (DFT) to study the ET of porphyrin molecules on Au surfaces.

Abstract:The direct electrochemistry of cytochrome C551 from pseudomonas aeruginosa has been investigated at indium-doped tin oxide (ITO) optically transparent electrodes without any surface promoters or mediators. The cyclic voltammograms show a quasi-reversible electrode reaction. Plots of peak reduction current against square root of scan rate are linear over the range 5 mV · s-1 to 100 mV · s-1. The diffusion coefficient of C551 is calculated from the slope of the line. The rate constant for heterogeneous electron transfer is determined by measuring cyclic voltammetry peak separation as a function of scan rate.

Abstract:With the advantages of in situ analysis and high surface sensitivity, surface-enhanced infrared absorption spectroscopy in attenuated total reflection mode (ATR-SEIRAS) combined with electrochemical methods has been employed to examine the interfacial direct electron transfer activity and adsorption kinetics of cytochrome c (cyt c). This work presents data on cyt c adsorption onto negatively charged mercaptohexanoic acid (MHA) and positively charged 6-amino-1-hexanethiol (MHN) self-assembled monolayers (SAMs) on gold nanofilm surfaces. The adsorbed cyt c displays a higher apparent electron transfer rate constant (33.5 +/- 2.4 s(-1)) and apparent binding rate constant (73.1 +/- 5.2 M-1 s(-1)) at the MHA SAMs surface than those on the MHN SAMs surface. The results demonstrate that the surface charge density determines the protein adsorption kinetics, while the surface charge character determines the conformation and orientation of proteins assembled which in turn affects the direct electron transfer activity.

Abstract:To combine voltammetric techniques with surface-enhanced resonance Raman scattering (SERRS), cytochrome c (cyt c) was immobilized on a roughened silver electrode chemically modified with a self-assembled monolayer (SAM) of 4-mercaptopyridine (PySH). All measurements were performed on the same electrode in a homemade spectroelectrochemical cell suitable for such applications. Cyt c on a PySH-SAM shows a quasi-reversible, monoelectronic, adsorption-controlled CV response with a formal reduction potential of -0.061 V (vs SCE), which is comparable to the values found for native cyt c adsorbed on different SAMs. SERRS spectra proved that cyt c adsorbed on a PySH monolayer is present in the native conformer (the B1 state). Voltammetric and SERRS experiments at high ionic strength revealed that the interaction between the SAM and the protein is electrostatic in nature. In conclusion, PySH was found to be suitable for adsorption of cyt c at SERRS-active silver surfaces. In comparison with other SAMs, PySH requires less time (10 min vs 12-18 h) to form a long-time durable and reproducible coating on the roughened electrode surface.

Abstract:It is proposed that direct electron transfer is dependent on the orientation of the heme plane in cytochrome c (cyt c) assembled on electrode surfaces. Orientation with the heme plane in cyt c parallel to the gold electrode surface favors the direct electron transfer, while vertical orientation of the heme plane makes the direct electron transfer difficult. A preferable electron transport pathway for cyt c is through the axial ligand (His-18) of the heme center rather than the porphyrin ring.

Abstract:The dependence of formal potential (E(0)') on temperature for cytochrome c in phosphate buffer solution at various pH was investigated by the microband gold electrode modified with a new promoter 4,6-dimethyl-2-mercaptopyrimidine (DMMP). The temperature dependence of the E(0)' showed a biphasic character in slightly alkaline solution with a turning point at ca. 45 degrees. However, the E(0)' values decreased monotonically with the increasing temperature in acidic and neutral media. The thermodynamic parameters of the electron transfer reaction for cytochrome c at the chemically modified microband electrode were estimated. The E(0)' and the changes of standard Gibb's free energy (Delta G(0)), entropy (Delta S-0) and enthalpy (Delta H-0) for the reduction of ferricytochrome c at 25 degrees C and pH 7.0 were 0.272V (vs. NHE) -26.2 kJ mol(-1), -123.7 J mol(-1) K-1 and -63.1 kJ mol(-1), respectively. The effect of buffer component on the E(0)' was also studied in binding (phosphate) and nonbinding (Tris/cacodylic acid) neutral buffer media. The results showed that the E(0)' in Tris/cacodylic acid buffer solution shifted positively by about 10 mV in comparison with that in phosphate buffer solution.
