Abstract:Several aromatic aldehydes and sodium bisulfite were mixed to prepare hydroxyl sulfonates,which were added to o-phenylenediamine or substituted o-phenylenediamine in refluxing ethanol to afford 2-substi-tuted benzimidazoles through oxidation by air directly.One of the fifteen synthesized compounds,2-(4-chlorophyenyl)-6-methoxyl-1H-benzimidazole has not been reported.All the compounds were identified by IR,1H NMR spectra and elemental analysis.The method is simple,mild,eco-friendly and requires no catalyst.

Abstract:An efficient protocol for chemoselective synthesis of 2-substituted benzimidazoles from a variety of aliphatic/aromatic/heteroaryl aldehydes and o-phenylenediamine derivatives promoted by NaHSO3 in water had been developed. The amount of NaHSO3 had a great effect on the reaction selectivity of 2-substituted benzimidazole and 1,2-disubstituted benzimidazole when the reaction was carried out in water. When the amount of the NaHSO3 was more than 11 equivalents, the 2-substituted benzimidazole could be highly selectively formed as the sole product. NaHSO3 was firstly reacted with aldehyde to form the aldehyde sodium bisulfite, which reacted with o-phenylenediamine to form the 2-substituted benzimidazole and inhibited the formation of 1,2-disubstituted benzimidazole. This protocol solved the poor selectivity problem appearing in traditional method when cyclocondensation between o-phenylenediamine and aldehydes. The method also had advantage of simple work up by filtrating the single 2-substituted benzimidazole precipitates from reaction mixture at the end of the reaction without further purification. In addition, the method was applicable to both electron-rich and electron-poor starting materials, which was successfully used for synthesizing nine novel 2-substituted benzimidazole derivatives containing a 1,2,3-triazole moiety. They were characterized by NMR, IR and HRMS spectrum. Moreover, this method had been applied to a large scale synthesis of 2-substituted benzimidazole derivatives.

Abstract:Here, a mild and general oxidative radical relay carbocyclization reaction with 2-arylbenzoimidazoles and cyclic ethers is reported. This method provides an efficient access to a wide range of structurally diverse benzimidazo[2,1-a]isoquinoline-6(5H)-ones under metal-free conditions. The wide substrate scope, good functional group tolerance, and scale-up operation of this method are expected to promote its potential applications in biotechnology and pharmacy.

Abstract:Herein, an atom-economical and eco-friendly electrochemical oxidation/cyclization of glycine derivatives through intramolecular Shono-type oxidative coupling is disclosed, leading to a variety of 2-substituted benzoxazoles in 51-85% yields. This oxidative cyclization proceeded in transition metal- and oxidant-free conditions and generated H-2 as only a byproduct. Additionally, gram-scale reactions and a broad substrate scope demonstrated the synthetic usefulness of this protocol.

Abstract:An efficient and convenient synthesis of benzoxazole/benzothiazole-substituted esters in a one-pot strategy is reported. In this investigation, a selective construction of C-N and C-S bonds via simple addition is performed. Thus, using substituted 2-aminophenols/2-aminobenzenethiols, TMTD (tetramethylthiuram disulfide) and α,β-unsaturated esters as starting substrates, C-N and C-S bonds can be selectively constructed by means of the Michael addition reaction. This protocol features high selectivity, high atomic economy, mild conditions, good functional tolerance and good to excellent yields, showing the potential value for the preparation of some biologically and pharmaceutically active compounds.

Abstract:A novel synthetic method for 1,2,6-trisubstituted benzimidazole derivatives was developed, which started from 3-fluoro-4-ratrobenzonitrile and aromatic amines to give the intermediates N-substituted-2-nitro-4-cyanoaraline (1) with high yield and short reaction time under microwave irradiation condition. Then, compound 1 reacted with different aldehydes in the presence of Na2S2O4 to provide the final compounds. Meanwhile, the process of the reactions was optimized in this study and the optimized conditions are described as: dimethyl sulfoxide (DMSO)-H2O as solvent, n(compound 1) : n(4-chlorobenzaldehvde) : n(sodium dithiorate)=1 : 1 : 9 at 80 degrees C for 2 h. Employing the same synthetic method, 21 1,2,6-trisubstituted benzimidazole derivatives were synthesized.

Abstract:An efficient and green method, combining enzymatic and visible-light catalysis for synthesis of the widely applicable 2-substituted benzothiazoles, has been developed. This method features a relay catalysis protocol consisting of biocatalytic promiscuity and visible-light-induced subsequent oxidization of 2-phenyl benzothiazolines. The whole reaction process is very high-efficiency, achieving 99% yield in just 10 minutes, under an air atmosphere, nearly 100% atomic utilization, and the 2-substituted benzothiazole products were obtained in good to excellent yields with a wide range of substrates. This reaction is the other example of combining the non-natural catalytic activity of hydrolases with visible-light catalysis for organic synthesis and the catalytic system does not require additional oxidants or metals, which is good for the environment.

Abstract:An effective method for the synthesis of benzazoles through the electrochemical oxidative cyclization of o‐aminophenol/o‐aminothiophenol/o‐phenylenediamine and aldehydes was developed. A series of benzazoles were efficiently synthesized by using this method in good yields. Furthermore, this method was applied to the synthesis of drug thiabendazole in a gram‐scale, which proved the practicality of the method. This approach avoids using catalysts and oxidants, and has the advantages of wide substrate range, mild reaction conditions and good tolerance of functional groups. Finally, the possible reaction mechanism was proposed and supported by cyclic voltammetry (CV) and the control experiments.

Abstract:We developed an efficient and convenient method for preparing N-substituted 2H- 1,4-benzoxazin-3-(4H)-ones from 2-halophenols via a nucleophilic substitution with 2-chloroacetamides followed by a Cul-catalyzed coupling cyclization. A broad spectrum of substrates can be effectively employed to afford the desired products in good yields. Since this method involves simple reaction conditions, a short reaction time, and a broad substrate scope, it is particularly attractive for the efficient preparation of biologically and medicinally interesting molecules.

Abstract:A condition-controlled strategy for selectively synthesis of 2-acylbenzothiazoles and bibenzo[b][1,4]thiazines from aryl methyl ketones and disulfanediyldianilines was realized using I2/DMSO or I2/MeCN systems, respectively. The desired products were synthesized in only 15 min with moderate to excellent yields (50%-90%) under microwave-assisted, metal-free conditions. The strategy provides a great advantage for selective synthetic applications in the efficient synthesis of benzothiazoles and bibenzothiazines heterocycle compounds.

Abstract:A sequential asymmetric conjugate addition/cyclisation of α-bromohydroxamates with para-quinone methide derivatives has been developed, which provides enantioenriched 1,4-benzoxazepines in generally high yields (up to 95%) and good enantioselectivities (up to 97 : 3 er). This protocol not only offers a novel and straightforward strategy for constructing chiral 1,4-benzoxazepines, but also demonstrates the potential of α-bromohydroxamates as three-atom synthons in asymmetric cyclisation reactions.

Abstract:We herein present a novel and an unexpected method for the synthesis of α-ketothioesters by the reaction of pyrrolidine with α-oxodithioester followed by treatment with methyl iodide in methanol. One of the synthetic applications of α-ketothioesters for the synthesis of 2-acylbenzimidazoles by acid-catalyzed cyclocondensation with benzene-1,2-diamines is described. The probable mechanisms of formation of α-ketothioesters and 2-acylbenzimidazoles are proposed.

Abstract:An efficient, economical, and convenient method was developed for the preparation of 2-substituted (N, O, C) benzothiazoles from N'-substituted-N-(2-halophenyl)thioureas, O'-substituted-N-(2-halophenyl) carbamothioates, or N-(2-halophenyl) thioamides via a base-promoted cyclization in dioxane without any transition metal. A one-pot variant combining the synthesis of the thiourea and the cyclization was also demonstrated. High yields were obtained, and a variety of functional groups were tolerated under these conditions. Transition-metal-free, mild reactive conditions, wide application scope, and shorter reaction times make this method superior to the reported methods for the synthesis of 2-substituted benzothiazoles and suitable for combinatorial format.

Abstract:We present an improved copper-catalyzed cyclization for an efficient synthesis of benzimidazoles from o -bromoarylamine and nitriles, under mild and ligand-free conditions. The optimal conditions yielded exceptional products of up to 98%, demonstrating the broad applicability of this synthetic strategy in generating a wide range of valuable imidazole derivatives. This methodology enables the efficient synthesis of various substituted benzimidazole derivatives and offers an environmentally friendly alternative to conventional methods. By eliminating the use of harsh reagents and high temperatures associated with traditional synthesis approaches, this method proves to be more efficient and robust. Notably, we successfully applied this synthetic approach to the synthesis of bendazol and thiabendazole, yielding 82% and 78%, respectively, on a 100 gram scale.
