Abstract:Epigenetic infractions could have a profound impact on the tumorigenicity. E74 like ETS (Erythroblast Transformation Specific) transcription factor 3 (ELF3) which encodes an epithelial-restricted member of the ETS transcription factor family, is long suspected to be a prominent player in breast cancer. However, ELF3 operates differently in various cancer types. Its precise role in breast cancer propagation remains elusive. Hence in this analysis, to resolve the functionality of ELF3 in breast cancer pathology, we have studied ELF3 expression, mutations, function networks and regulation in data from patients with breast invasive carcinoma (BRCA), procured from various different public databases. The present study protrudes staggered evidence on the prominence of ELF3 in BRCA and its potential role as a novel biomarker. ELF3 expression was assessed using Oncomine and UALCAN analysis, and data concerning ELF3 genetic alterations and associated survival rates were further analyzed using c-BioPortal. Further, LinkedOmics was used to indicate correlations between ELF3 and other pertinent differentially expressed genes. Gene enrichment analysis of ELF3 associated genes was conducted using the Web-based Gene SeT AnaLysis Toolkit. The expression levels of ELF3 were considerably increased in patients with BRCA and also in various molecular subtypes of breast cancer cells. ELF3’s behavior was permuted in 82 of 996 (8%) BRCA patients. Expression of this gene is ascribed to functional networks pertaining to biochemical pathways such as ‘cell adhesion molecule binding’, ‘histone deacetylase binding’, ‘chromosome segregation’, ‘DNA-binding transcription activator activity’ and ‘Cell cycle’ pathways. The present study revealed the expression pattern variations and potential changes in bio-molecular pathways of ELF3 in BRCA, explicating the pernicious nature of ELF3 and establishing a genomic foundation for further investigation of the role of ELF3 in breast carcinogenesis.
