Antitumor Effects of Magnetic Gemcitabine Stealth Nano-liposomes on Human Breast Cancer Xenografts in Nude Mice

Abstract:Objective To study the optimal conditions to prepare magnetic gemcitabine stealth nano-liposomes (MGSL), evaluate their physico-chemical properties and antitumor effects in vivo. Methods MGSL were prepared by reverse-phase evaporation method. The nanoparticle appearance was examined by both scanning electron microscope and atomic force microscope. The particle diameter and particle size distribution of MGSL were determined by a laser light scattering technique. The encapsulation efficiency and drug loading were quantified by HPLC. The magnetic response at room temperature was measured by a magnetometer. The stability of MGSL was also evaluated. The antitumor effect of MGSL on human breast cancer xenografts in nude mice was investigated. Results The shape of MGSL was round or elliptical. The particle diameter of MGSL optimized was 206.6 nm, the encapsulation efficiency was (81.7±5.1)% and drug loading was (10.4±0.7)%. The growth of tumor in the group of MGSL(+) was significantly slowed down as compared with other groups. The tumor volume in MGSL(+) group was smaller than that of other groups from the second day (all P<0.05). The tumor inhibition rate in MGSL(+) group was 87.3%, which was remarkably higher than in other groups (all P<0.05). Conclusion MGSL prepared by reverse-phase evaporation method met with the demand of targeted delivery system, and they might be an effective anti-tumor agent.
