Abstract:The optical properties of porcine liver were studied during the dehydration process at different constant temperature (37degreesC, 40degreesC, 43degreesC, 46degreesC, 50degreesC). Each time after porcine liver samples were placed in thermostat for a different period, the weight of samples were recorded by a precise electronic scale, and the optical properties of corresponding samples were measured by a double integrating sphere system. The results showed that the absorption coefficient mainly depended on dehydration level of tissue. In contrast, the reduced scattering coefficient had more complex relation with the dehydration process, which depended on both temperature and heating time. There is not obvious relation between the reduced scattering coefficient and dehydration level. This study implies that dehydration, temperature and time are important factors altering optical properties of tissue.
Quantitative Analysis of Dehydration in Porcine Skin for Assessing Mechanism of Optical Clearing

Abstract:Dehydration induced by optical clearing agents (OCAs) can improve tissue optical transmittance; however, current studies merely gave some qualitative descriptions. We develop a model to quantitatively evaluate water content with partial least-squares method based on the measurements of near-infrared reflectance spectroscopy and weight of porcine skin. Furthermore, a commercial spectrometer with an integrating sphere is used to measure the transmittance and reflectance of skin after treatment with different OCAs, and then the water content and optical properties of sample are calculated, respectively. The results show that both the reduced scattering coefficient and dehydration of skin decrease with prolongation of action of OCAs, but the relative change in former is larger than that in latter after a 60-min treatment. The absorption coefficient at 1450 nm decreases completely coincident with dehydration of skin. Further analysis illustrates that the correlation coefficient between the relative changes in the reduced scattering coefficient and dehydration is ∼1 during the 60-min treatment of agents, but there is an extremely significant difference between the two parameters for some OCAs with more hydroxyl groups, especially, glycerol or D-sorbitol, which means that the dehydration is a main mechanism of skin optical clearing, but not the only mechanism.
Dehydration Effect on Absorption and Scattering Properties of Porcine Kidney

Abstract:Dehydration effect on the absorption and scattering properties of porcine kidney were studied in vitro.The measurements were performed using a spectrophotometer with an integrating sphere attachment,and the absorption and scattering properties at 532 nm and 1 064 nm were assessed from these measurements using the inverse adding-doubling method.The results of measurement showed that the absorption coefficients and reduced scattering coefficients of porcine kidney tissue at 532 nm and 1 064 nm were all increased with the increase of the dehydration level of porcine kidney at exposing temperature of 37 ℃,obviously.For exposing temperature of 37 ℃,the absorption coefficients of porcine kidney tissue at 532 nm and 1 064 nm were obviously bigger than that of natural porcine kidney tissue at the corresponding wavelength,and for exposing temperature of 37 ℃,the reduced scattering coefficients of porcine kidney tissue at 532 nm and 1 064 nm were also bigger than that of natural porcine kidney tissue at the corresponding wavelength,obviously.For the same dehydration level,the absorption coefficient and reduced scattering coefficient of porcine kidney tissue at 532 nm were obviously bigger than the absorption coefficient and reduced scattering coefficient of porcine kidney tissue at 1 064 nm.The results of study imply the dehydration level,exposing temperature and exposing time were important factors that influence the absorption and scattering properties of porcine kidney tissue.

Abstract:The physical changes of tissue are complicated to evaluate during optical clearing (OC) treatment. Monitoring the changes of optical parameters, including the complex refractive index (CRI), helps people better understand the OC process. From the imaginary part of CRI, we can deduce the extinction coefficient of tissue. Based on the total internal reflection method, the time-dependent CRI of porcine muscle during natural dehydration is well determined. Results show that the real RI increases continuously with the increase of dehydration time, whereas the extinction coefficient initially increases and then decreases. Finally, the extinction coefficient becomes much smaller than the initial value, which demonstrates that better tissue optical clarity is obtained. The change tendency of the extinction coefficient of tissue is used to qualitatively explain the dynamic change of transmittance of a natural dehydrated tissue. Consequently, CRI, especially its imaginary part, is a very useful optical parameter by which to evaluate the OC effect.
Effect of Dehydration on Optical Clearing and OCT Imaging Contrast after Impregnation of Biological Tissue with Biochemical Agents

Abstract:There are two events happening simultaneously after the impregnation of tissue with the hyperosmotic agents. The diffusion of the agents into tissue reduces the refractive index mismatch between the scattering centres and background medium. And in the meantime, the hyperosmotic characteristics of agents cause tissue dehydration. Both processes are believed to have the effects on reducing light scattering of biological tissue. In order to investigate the role of dehydration in optical clearing effect on the tissue with the application of hyperosmotic agents, the skin tissues with the application of glycerol are dynamically imaged with optical coherence tomography (OCT). We demonstrate experimentally that both the depth and the contrast of OCT imaging of tissue can be enhanced by the application of agents. Higher concentration of agents causes more water loss of skin tissue and a stronger optical clearing effect. We suggest that the dehydration induced by the agents contributes to the enhancement of both OCT imaging depth and contrast. OCT imaging movies of optical clearing effect of hyperosmotic agents on the skin tissues in vitro are provided.

Abstract:Dehydration is an important mechanism in the process of tissue optical clearing, often accompanied by microstructural changes such as the decrease in scattering coefficient and thickness. In this paper, the dynamic process of tissue dehydration is taken as an example, and a method to extract tissue characteristics based on the regularity of the sequential polarization image as well as Monte Carlo (MC) simulation database is developed and validated. Three temporal polarization parameters are extracted based on the polarization image sequence features of four tissue samples, and then the MC simulation dataset of the influence of different tissue features on three polarization parameters are used as a reference, which not only reveal the most significant microstructural differences between tissue types, but also quantitatively evaluate the interstitial birefringence and fiber content. Specifically, the birefringence of porcine fat, porcine skin and two kinds of skeletal muscle are assessed to be 0, 1.7 × 10 −5 and 2.8 × 10 −3 respectively by ( DoLP-DoCP ) ∆3 or δ ∆3 , and the main reason for the difference of the trend of polarization parameter between porcine fat and skin is further determined to be birefringence rather than cylindrical scattering. In addition, the scattering coefficient ratio of cylindrical scatterers and spherical scatterers is evaluated to be roughly 9:1, 4:1 and 2:1 for porcine skin, porcine skeletal muscle and chicken skeletal muscle respectively. The consistency of experiments and simulations on polarization imaging to monitor dynamic tissue process have verified the potential of this temporal polarization characterization in more biomedical applications.

Abstract:Based on the optical characteristic parameters and thermal characteristic parameters of bio-tissue and the relation between blood perfusion and bio-tissue's thermal damage, the transmission of laser through the biotissue, transient heat response and thermal damage zone of tissue are numerical simulated. By analyzing the effect of laser power input and the size of laser head on the curative effect of thermotherapy, some suggestions useful for tumour clinical treatment were proposed and discussed.

Abstract:Glycocalyx is located outside the vascular endothelial cells playing an important role in vascular homeostasis. However, lacking efficient detection methods is one of the biggest obstacles to study the glycocalyx. In this study, three dehydration methods were used to compare the preservation of HUVEC, aorta and kidney glycocalyx by transmission electron microscope. The chemical pre-fixation was performed by lanthanum nitrate staining, and the mice aorta and renal glycocalyx were prepared by different dehydration methods such as ethanol gradient, acetone gradient and low temperature dehydration. HUVEC glycocalyx was prepared by acetone gradient and low temperature dehydration. Low temperature dehydration method preserves HUVEC and mice aortic glycocalyx completely, which had a certain thickness and presented a needle-like structure. But for mice kidney, the acetone gradient dehydration preparation method could better preserve the glycocalyx integrity than other two methods. In conclusion, low temperature dehydration method is suitable for HUVEC and aortic glycocalyx preservation, acetone gradient dehydration method is more suitable for kidney glycocalyx preservation.

Abstract:The optical clearing technique is able improve light penetration depth by applying optical clearing agent (OCA) with high refractive indices and hyperosmolarity. However, the mechanism of tissue optical clearing is not much clear. In this work, both physical and physiological mechanisms of optical clearing are discussed. For physical mechanisms, a tissue phantom, Intralipid, was mixed with OCAs, and then the reduced scattering coefficient was theoretically predicted and experimentally measured. It was found that the physical mechanisms for optical clearing are due to the structure change of scatters or the enhancement of the background refractive index. For physiological mechanisms, the dehydration of porcine skin in vitro caused by topical application of OCA was investigated with NIR reflectance spectrum analysis. The results showed that dehydration is major factor during the initial few minutes, and some other changes of tissue structure maybe occurs with increase of time. And the microstructure and ultrastructure of rat skin was examined after in vivo application of glycerol, we found that both the thickness of skin and the size of fiber reduce. This work is helpful for clarifying the mechanisms of tissue optical clearing, which will contribute to the improvement and clinical application of optical clearing technique.
Controlling Optical Properties of Bio-Tissue by the Use of Biocompatible Hyperosmotic Agents

Abstract:Previous results of tissue optical clearing have qualitatively indicated that optical properties of bio-tissue can be changed by the use of biocompatible hyperosmotic agents. In this talk, it is our aim to try to quantitatively evaluate how relative between changes of optical properties and hyperosmotic agents. Tissue-like turbid media, i.e. intralipid-10% suspension and porcine skin tissues, medicated without and with biocompatible hyperosmotic agents, have been investigated within NIR range. Optical parameters (absorption coefficient, scattering coefficient and anisotropy factor) of samples have been calculated by Inverse Adding-Doubling (LAD) modelling method. The results have demonstrated that optical property parameters (absorption coefficient, scattering coefficient and anisotropy factor) of tissue samples have changed with the course of the administration of biocompatible hyperosmotic agents into intralipid-10% suspension and porcine skin tissues respectively. This reveals better refractive index matching environment has been gradually created within tissue. Further, the results have showed the potential capabilities to quantitatively control optical properties of bio-tissue in this way. Therefore, this would facilitate explanation of the mechanism of tissue optical clearing by the use of biocompatible hyperosmotic agents.
Optical Properties of Native and Coagulated Human Liver Tissues at Argon Ion Laser - an in Vitro Study Using the Double-Integrating-Sphere Technique

Abstract:We examine the optical properties of native and coagulated human liver tissues in vitro at four different wavelengths of argon ion laser, 476.5, 488, 496.5 and 514.5 nm, and the differences in the optical parameters of the tissues between 630nm and 790nm. The optical properties, mu(a), mu(s) and g, of liver tissue were determined by measuring the diffuse reflectance, diffuse transmission and collimated transmission in a double integrating sphere set-up, and the inverse adding-doubling algorithm was used to calculate the optical properties from the measurements. The investigative results showed that there were significant differences respectively in the absorption coefficients, scattering coefficients and anisotropy factors between native and coagulated human liver tissues at the same laser wavelength (P < 0.01), these large differences of the absolute values in optical properties indicate that there were large differences in compositions and structures between both, and large differences in compositions and structures induce large differences in absorption and scattering properties between both, and these differences vary with a change of laser wavelength. The scattering coefficients for per tissue type exceed the absorption coefficients by at least one order of magnitude. Hence the conclusions may be helpful to diagnostics and therapeutic applications in liver tissue.
Short-term and long-term effects of optical clearing agents on blood vessels in chick chorioallantoic membrane.

Abstract:The tissue optical clearing technique shows great potential in optical diagnosis and therapy. However, short-term and long-term effects of optical clearing agents on blood vessels, which are relevant to the safety of clinical applications, have not been clarified. We used laser speckle contrast imaging to monitor the changes in blood vessels in chick chorioallantoic membrane (CAM) after application of glycerol or glucose. The changes in morphology of vessels and blood flow velocity were measured. Long-term effects on blood vessels were investigated by observing the function and the development of blood vessels. The results show that glycerol reduces the local blood flow velocity and constricts and even blocks vessels quickly. At 2 days, the blood flow velocity is recovered to different extents, and new blood vessels develop but are fewer in number. Glucose induces slow changes in blood flow or vessels. However, most blood vessels are blocked, and no new blood vessel develops at 2 days. The effects depend on the dosage of agents, including volume and concentration, and decrease with the dosage of agents. Therefore, short-term effects of glucose on blood vessels are slighter than those of glycerol, but long-term effects of glucose are greater. Abstract:In present work, we studied the effect of optical clearing on porcine skin in vitro with glycerol by Raman microspectroscopy, denoted as RM, at various time intervals of 0, 15, 30, 45, 60, and 75 min respectively. The results showed that the addition of glycerol significantly improved the depth of RM measurement, and enhanced the recovery of skin tissue Raman spectra that were not overlapped with the glycerol Raman spectra over time. Moreover, it was found that the Raman signals resembled the native spectrum of the molecules in porcine skin with a negligible frequency shift. Furthermore, we evaluated the extent of optical clearing in porcine skin by utilizing various concentrations of 40%, 60%, and 80% glycerol solution. The results demonstrated that with the increase of concentration of glycerol, the optical clearing of porcine skin was much improved.
Using Low-Frequency Ultrasound to Improve the Optical Clearing of Porcine Skin

Abstract:The glycerol used as an enhancer for tissue optical clearing technique has been researched. However, using it and a physical way of ultrasound enhance optical clearing of tissue reported a few. We researched that the ultrasound whether can improve the optical clearing of dealt with 80% glycerol tissue. The fresh porcine skins divided into four groups. The first group was not dealt with by ultrasound and 80% glycerol, the second group was dealt with by only ultrasound, the third group was dealt with by 80% glycerol and no by ultrasound, and the fourth group was dealt with by both 80% glycerol and ultrasound. And we measured changes in optical scattering of the porcine skins under treatment with OCT. From the OCT images show that the fourth group changed very faster than the other's during the 0~15 min. And it can be clearly seen that there is a significant improvement in the light penetration depth and imaging contrast in a shorter time. It is possible that the low-frequency ultrasound can make disordering of the stratum corneum lipids of the porcine skin (because the cavitation has happened), and improve the speed of 80% glycerol through the stratum corneum of skin. These results proved that using 80% glycerol with the ultrasound can better improve the optical clearing of tissue.
Optical Transport Characteristics of Human Tissues in Vitro at 808 Nm Linearly Polarized Laser Irradiation

Abstract:In this paper, a double-integrating-spheres system, basic principle of measuring technology of ray radiation, optical model of biological tissues were used to study the optical properties of human tissues. The results of measurement showed that the parameters of optical properties of human normal bladder and human bladder cancer tissue at 808 nm laser and the linearly polarized laser in Kubelka-Munk two-flux model had prominent distinction (P0.01). The parameters of optical properties of human normal bladder tissue for 808 nm laser and the linearly polarized laser had all distinction. Absorption coefficient and effective attenuation coefficient among all parameters of optical properties of human normal bladder tissue had obviously distinction. All parameters of optical properties of human bladder cancer tissue for 808 nm laser and the linearly polarized laser had not prominent distinction(P0.05) which showed that the depolarization of human bladder cancer tissue to 808 nm linearly polarized irradiation was very big. The parameters of optical properties of human normal small intestine tissue at 808 nm laser and the linearly polarized laser irradiation had all prominent distinction (P0.01), which showed that the depolarization of human normal small intestine tissue to 808 nm linearly polarized irradiation was smaller. Distribution of light intensity of three kinds of human tissues at 808 nm laser and the linearly polarized laser irradiation in Kubelka-Munk two-flux model had obviously dissimilitude.

Abstract:Laser therapy has been widely used to treat port-wine stains (PWS) and other cutaneous vascular lesions via selective photothermolysis. Animal models are a valuable tool for investigating thermal responses beneath the skin. However, in previous animal experiments, such as the dorsal skin chamber model, one side of the skin was removed, resulting in the loss of mechanical support for the target blood vessel. In this study, the optical clearing technique was applied to the dorsal skin, allowing direct observation of real thermal responses within the tissue without removing the covering skin. The target blood vessels were irradiated with a pulsed 1064 nm Nd: YAG laser. The corresponding thermal responses were recorded using a CCD camera. Additionally, variations in skin reflectance spectra were measured before and after laser irradiation. Due to the optical clearing and reflectance spectra measurement, vessel responses such as contraction, reperfusion, and full occlusion were correlated with specific variation patterns in reflectance spectral signals.

Thermal Effects on the Light Propagation Characteristics of Kidney Tissue in Vitro

Abstract:Thermal effects on the optical properties of porcine kidney tissue at 532 nm and 1 064 nm was studied in vitro.The measurements were performed using a spectrophotometer with an integrating sphere attachment,and the optical properties were assessed from these measurements using the inverse adding-doubling method.The results of measurement showed that the absorption coefficients and reduced scattering coefficients of porcine kidney tissue at 532 nm and 1 064 nm vary with a change of exposing temperature obviously.Within exposing temperature range from 25 ℃ to 80 ℃,the absorption coefficients and reduced scattering coefficients of porcine kidney tissue at 532 nm were significantly bigger than that of porcine kidney tissue at 1 064 nm respectively.Maximal absorption coefficients of porcine kidney tissue at 532 nm and 1 064 nm were respectively 1.121 mm-1 and 0.269 mm-1 at exposure temperature of 80 ℃,and minimal absorption coefficients were respectively 0.131 mm-1and 0.019 mm-1 at exposure temperature of 25 ℃,and maximal reduced scattering coefficients of porcine kidney tissue at 532 nm and 1 064 nm were respectively 2.905 mm-1 and 0.705 mm-1 at exposure temperature of 80 ℃ and 70 ℃ respectively,and minimal reduced scattering coefficients were respectively 0.391 mm-1 and 0.184 mm-1 at exposure temperature of 25 ℃.The results of study imply the exposing temperature were important factors that influence the absorption and scattering properties of porcine kidney tissue.

Abstract:In recent years, the tissue optical clearing (OC) technique in the biomedicine field has drawn lots of attention. Various physical and chemical methods have been introduced to improve the efficacy of OC. In this study, the effect of the combination of glycerol and ultrasound treatment on OC of in vitro porcine muscle tissues has been investigated. The refractive index (RI) matching mechanism of OC was directly observed based on the derivative total reflection method. A theoretical model was used to simulate the proportion of tissue fluid in the illuminated area. Moreover, the total transmittance spectra have been obtained by a spectrometer over the range from 450 nm to 700 nm. The administration of glycerol and ultrasound has led to an increase of the RI of background medium and a more RI matching environment was achieved. The experimental results support the validity of the ultrasound treatment for OC. The RI matching mechanism has been firstly quantitatively analyzed based on the derivative total reflection method.

Abstract:A double-integrating-spheres system and Inverse Adding-doubling method were used in this experiment.The optical properties of native and coagulated human liver tissue at 532 nm and 1 064 nm of KTP/YAG laser were study.The results of experiment showed that there is a 23.5 percent increase in the absorption coefficient for coagulated human liver tissues at 532 nm of KTP laser in comparison with the absorption coefficient for native human liver tissues at the same laser wavelength(P0.05),and there is a 34.3 percent decrease in the absorption coefficient for coagulated human liver tissues at 1 064 nm of Nd:YAG laser in comparison with the absorption coefficient for native human liver tissues at the same laser wavelength(P0.05).There is a 4.50 times increase in the scattering coefficient for coagulated human liver tissues at 532 nm to compare with the scattering coefficient for native human liver tissues at the same laser wavelength(P0.05),and there also is a 6.41 times increase in the scattering coefficient for coagulated human liver tissues at 1 064 nm to compare with the scattering coefficient for native human liver tissues at the same laser wavelength(P0.05).there is a 5.47 percent decrease in the anisotropy factor for coagulated human liver tissues at 532 nm to compare with the anisotropy factor for native human liver tissues at the same laser wavelength(P0.05),and there also is a 1.95 percent decrease in the anisotropy factor for coagulated human liver tissues at 1 064 nm to compare with the anisotropy factor for native human liver tissues at the same laser wavelength(P0.05).
