Abstract:Mixed-mode chromatography (MMC) is a promising technology for protein separation with high adsorption capacity, good selectivity, salt-tolerance, and facile elution. Expanded bed adsorption (EBA) can capture target proteins directly from bioparticle containing crude feedstock without solid-liquid separation. In this study, MMC and ERA were combined to develop a mixed-mode EBA technique for human serum albumin (HSA) separation. Two mixed-mode EBA resins (using quartz-densified agarose beads and tungsten carbide/agarose composite beads, respectively) with. tryptamine as the ligand were prepared (TA-S and TA-T). Static and dynamic adsorption behaviors of HSA were determined under varying conditions and typical adsorption properties of pH dependence and salt-tolerance were found. The performance of the resins in expanded bed was studied, and the dynamic binding capacities at 10% breakthrough (Q(10%) ) of TA-S and TA-T at bed expansion of 1.8 were 27.54 and 18.04 mg/mL settled resin, respectively. Moreover, these two resins were used to separate recombinant HSA (rHSA) from Pichia pastoris culture broth. TA-S showed better separation performance and the purity of rHSA monomer reached to 83.3% with the recovery of 91.6% through a two-step elution process. Total content of rHSA monomer and degraded fragment was 99.8%. The results demonstrate that the combination of MMC and EBA would be a potential platform for protein capture with fewer pretreatments and high process efficiency.
Application of C-18-Modified Mseoporous Sba-15 As the Substrate for High Performance Liquid Chromatography

Abstract:C-18 functionalized mesoporous silica SBA-15 with a high surface area was applied in high performance liquid chromatography (HPLC) to separate small blomolocules: cysteine, glutathione, dopamine and 6-thiopurine. Compared to commercial available C-18 column, C-18-SBA-15 has a much higher surface area, which enhance,,, the capillary HPLC resolution by increasing molecular capacity factors. The uniform distributed mesopores in SBA-15 result in HPLC peaks narrower than that of commercial column. Mesoporous SBA-15 can be used as an excellent HPLC stationary phase for special applications for its low cost, easy synthesis and various group-modifications.
Incorporation of silica nanoparticles into porous templates to fabricate mesoporous silica microspheres for high performance liquid chromatography applications

Abstract:High-performance liquid chromatography is one of the most important analytical tools for the identification and separation of substances. The efficiency of this method is largely determined by the stationary phase of the columns. Although monodisperse mesoporous silica microspheres (MPSM) represent a commonly used material as stationary phase their tailored preparation remains challenging. Here we report on the synthesis of four MPSMs via the hard template method. Silica nanoparticles (SNPs) which form the silica network of the final MPSMs were generated in situ from tetraethyl orthosilicate (TEOS) in the presence of (3-aminopropyl) triethoxysilane (APTES) functionalized p(GMA-co-EDMA) as hard template. Methanol, ethanol, 2-propanol, and 1-butanol were applied as solvents to control the size of the SNPs in the hybrid beads (HB). After calcination, MPSMs with different sizes, morphology and pore properties were obtained and characterized by scanning electron microscopy, nitrogen adsorption and desorption measurements, thermogravimetric analysis, solid state NMR and DRIFT IR spectroscopy. Interestingly, the 29Si NMR spectra of the HBs show T and Q group species which suggests that there is no covalent linkage between the SNPs and the template. The MPSMs were functionalized with trimethoxy (octadecyl) silane and used as stationary phases in reversed-phase chromatography to separate a mixture of eleven different amino acids. The separation characteristics of the MPSMs strongly depend on their morphology and pore properties which are controlled by the solvent during the preparation of the MPSMs. Overall, the separation behavior of the best phases is comparable with those of commercially available columns. The phases even achieve faster separation of the amino acids without loss of quality.
Large-pore Mesoporous Silica Spheres: Synthesis and Application in HPLC

Abstract:Large-pore mesoporous silica microspheres with tailored pore sizes were prepared via a two-step synthesis process by using a triblock copolymer EO20PO70EO20 as template in combination with a cosurfactant cetyltrimethylammonium bromide (CTAB) and a cosolvent ethanol. The obtained mesoporous silica spheres were characterized by scanning electron microscopy (SEM), transmission electron microscopy (TEM), X-ray diffraction (XRD), and N2 sorption. The pore size of the silica spheres was increased up to 9.8nm with increasing the hydrothermal treatment temperature in the second step to 130°C. The presence of the cosolvent ethanol was found to be essential to the formation of mesoporous silica with a perfectly spherical morphology. Spherical mesoporous silica was successfully functionalized by dimethyloctadecylchlorosilane (C18), leading to the formation of a C18-modified spherical mesoporous silica. It has been shown that the C18-modified mesoporous silica microspheres can be used as a good packing in high performance liquid chromatography (HPLC) to separate both small aromatic molecules and large biomolecules such as proteins.

Abstract:Effective protein adsorption by solid matrices from complex biological samples has attracted attention for broad application in biomedical field. Immobilization of calixarenes to solid supports is an essential process for their application in protein separation and purification. Silica is the most widely used support material in calixarene immobilization. With high concentration of polymer microspheres as templates, the large pore mesoporous silica microspheres with controllable, uniform size and structure were successfully synthesized and the resulting large pore mesoporous silica microspheres were modified with water-soluble tetrasulfonate calix[4]arene of unique hollow cavity-shaped structure. The tetrasulfonate calix[4]arene modified large pore mesoporous silica microspheres (SCLX4@LPMS) were characterized by diverse analytical techniques and their protein adsorption performance were also investigated. The obtained SCLX4@LPMS gave rise to an adsorption efficiency of >90% for cytochrome c and lysozyme within a wide pH range of 3.0-10.0 and possessed remarkably high adsorption capacity of cytochrome c (363.64 mg g(-1)) and lysozyme (166.11 mg g(-1)). The retained cytochrome c and lysozyme can be readily eluted by using phosphate buffer solution containing NaCl as a stripping reagent with the recoveries of 81% and 86% after 5 times enrichment, respectively. The SCLX4@LPMS microspheres have been applied for the selective adsorption of proteins in real samples and had the application potential in protein adsorption, drug delivery, biosensors, and other biomedical fields.
Large‐pore Mesoporous SBA‐15 Silica Particles with Submicrometer Size As Stationary Phases for High‐speed CEC Separation

Abstract:A new mesoporous sphere-like SBA-15 silica was synthesized and evaluated in terms of its suitability as stationary phases for CEC. The unique and attractive properties of the silica particle are its submicrometer particle size of 400 nm and highly ordered cylindrical mesopores with uniform pore size of 12 nm running along the same direction. The bare silica particles with submicrometer size have been successfully employed for the normal-phase electrochromatographic separation of polar compounds with high efficiency (e.g., 210 000 for thiourea), which is matched well with its submicrometer particle size. The Van Deemeter plot showed the hindrance to mass transfer because of the existence of pore structure. The lowest plate height of 2.0 mu m was obtained at the linear velocity of 1.1 mm/s. On the other hand, because of the relatively high linear velocity (e.g., 4.0 mm/s) can be generated, high-speed separation of neutral compounds, anilines, and basic pharmaceuticals in CEC with C-18-modified SBA-15 silica as stationary phases was achieved within 36, 60, and 34 s, respectively.

Abstract:Siliceous mesostructured cellular foam (MCF) with highly interconnected porous structure, ultralarge pore size and relatively uniform particle size (3–5μm) was prepared to achieve the mixed-mode and efficient separation of intact proteins. And molecular sieving effect for the first time played an important role in protein separation using mesoporous silica materials as HPLC stationary phase. The spherical silica particles were synthesized via hydrothermal method and the pore size was easily regulated by adding NH4F as well as altering the aging time. After aminopropyl derivatization, the chromatographic performance of functionalized mesoporous silica particles was investigated in comparison with those without modification and commercial NH2 column, and their mixed-mode retention mechanisms were investigated in detail. The superior separation performance for the retention of proteins was obtained on our home-made column in comparison with commercial NH2 column. The influences of aminopropyl derivatization and mobile phase composition on the column property were also investigated. Moreover, the home-made column showed similar performance for separation of polar anilines and neutral PAHs with the commercial column, owing to mixed-mode retention mechanisms including p–π stacking, electron interaction, hydrophobic effect, π–π EDA interaction and hydrogen bonding. All these results indicated that the aminopropyl modified MCF would be promising in the mixed-mode and efficient separation of biomolecules in addition with small molecules.

Abstract:Vigorously developing new high performance liquid chromatography (HPLC) stationary phases to meet the versatile separation requirements is still an important issue in the field of analytical chemistry. Conjugated microporous polymers (CMPs) are a new type of three-dimensional network porous material with high specific surface area, good chemical stability and superhydrophobicity. Herein, we firstly report the synthesis and applications of CMPs@SiO2 material for HPLC stationary phase. The CMPs@SiO2 material can be in situ fabricated via Sonogashira coupling of 1,3,5-triethynylbenzene and 1,4-diiodobenzene on the surface of spherical silica. The morphology and physicochemical properties of the synthesized stationary phase material were investigated by a series of characterization methods. Due to the superhydrophobic nature of the CMPs@SiO2 material, the packed CMPs@SiO2 HPLC column displays ultrastrong chromatographic retention and can be used for separation of both hydrophobic and hydrophilic compounds with good selectivity. Significantly, CMPs@SiO2 column can be performed for separation with pure acetonitrile as the eluent. Thus, the new column was successfully exploited for monitor and analysis of the hydrolysis of silane coupling agents. Furthermore, based on its oleophilicity, this report firstly utilized the CMPs@SiO2 material to identify and analyze the quality of cooking oils through one-step enrichment and subsequent HPLC separation. We will further exploit to fabricate versatile CMPs-based stationary phases, highlighting their potential applications in different separation scopes.

Abstract:Micrometer-sized silica spheres with bimodal macromesoporous structures are synthesized continuously by a modified gelation technology, and a novel coaxial microfluidic device is applied to control the size of the silica spheres. These spheres are used as a new protein adsorbent to realize fast adsorption and separation of protein mixture. BSA and lysozyme (LYS) are as model proteins with different sizes and pIs. High protein adsorption capacity and rapid adsorption rate have been achieved. Fast separation of a binary mixture of BSA and LYS through a short packed column (50 mm x 4.6 mm id) has also been successfully realized. LYS was selectively adsorbed while BSA flowed through the column in 12 s at a flow rate of up to 5.0 mL/min. As a result, an ultrafast adsorption and separation of BSA and LYS was obtained, showing great potential for applications in fast, large-scale protein separation processes of these monodispersed silica spheres.

Abstract:A stationary phase based on sub-2 μm ground silica monolith particles was fabricated by in situ polymerization and applied in micro-column for separation of peptides. The sub-2 μm silica particles were obtained from monolith using sol-gel process followed by grinding and sedimentation to remove the fines. Initially, the silica monolith particles were pretreated with 3-trimethoxysilyl propyl methacrylate to attach double-bonded ligands onto the surface, then a network structure was formed onto the surface of the particle using styrene, N-isopropylacrylamide and ethylene glycoldimethacrylate. The effect of the flow rate of mobile phase on the separation performance was investigated. The stationary phase was characterized by field emission scanning electron microscopy, thermogravimetry, particle size distribution and element analysis. The resultant phase was packed in glass lined stainless steel micro-columns (2.1 mm x 50 mm) and evaluated for fast separation. An average number of theoretical plates as high as 9800 plates/column (5.10 μm plate height) was achieved for five synthetic peptides under the optimized flow rate of 0.15 mL/min. The repeatabilities of column-to-column, intra-day and inter-day through relative standard deviation were found better than 4%, exhibiting satisfactory repeatability of the developed micro-column for fast separation of peptides.This article is protected by copyright. Abstract:Mesoporous SBA-15 silica particles were prepared in 1.6 M HCl with triblock copolymer Pluronic P123 (poly(ethyleneglycol)-block-poly(propyleneglycol)-block-poly(ethyleneglycol)) as the templating agent. Characterization shows that it has a highly ordered hexagonal mesoporous structure running along the same direction, extremely narrow pore size distribution (5.8 nm) and high surface areas (799 m2 g−1). The SBA-15 particles were successfully employed for the normal-phase chromatography separation of six Vitamin E isomers (mobile phase: n-hexane/ether (97/3, V/V); flow-rate: 2.0 mL min−1). The results show that α-, β-, γ-, δ- tocotrierols and α-, β- tocotrienols are base-line separated.

Abstract:Monodisperse porous silica microspheres with a tunable particle size and pore size were fabricated by utilizing porous polymer microspheres as a novel hard template during the sol-gel process followed by calcination to remove the polymer. The particle size and pore size could be simply tuned by the feature of the polymer template and reaction conditions such as different functionalization of the parent polymer template, particle size of polymer template, and amount of TEOS during the sol-gel process. EDA (ethylenediamine), APTES (3-aminopropyl)triethoxysilane, and TMA (trimethylamine hydrochloride) functionalization of porous poly(GMA-co-EGDMA) microspheres were carried out to study their effect on the synthesized porous silica microspheres. The TMA-functionalized polymer microspheres led to a higher yield, smaller silica nanoparticles, and no self-nucleation of TEOS due to their positive surface charge. Furthermore, no addition of NaOH during TMA functionalization and the amount of TEOS during the sol-gel process played key roles in determining the pore size and particle size of porous silica microspheres. Then, through poly(aspartic acid) coating of the APTES-functionalized monodisperse porous silica microspheres, the modified monodisperse porous silica microspheres were explored as the stationary phase of HPLC for protein separation. The effects of particle size and pore size on the chromatographic behavior were discussed. When the protein mixture composed of transferrin, hemoglobin, ribonuclease A, cytochrome C, and lysozyme was used as the model analytes, the as-prepared silica microspheres exhibited an excellent separation performance with a high protein recovery and good reproducibility.
Rapid Separation of Proteins by Capillary HPLC on a Short Polymethacrylate‐based Strong Cation‐exchange Monolithic Column

Abstract:A polymethacrylate-based strong cation-exchange capillary monolithic column was prepared by in-situ co-polymerization for the fast separation of proteins. Glycidyl methacrylate (GMA) was used as monomer, ethylenedimethacrylate (EDMA) as cross link agent and the mixture of 1-propanol, 1,4-butanediol and water as porogen solvent. The monolith was sulfonated using 1 mol/L Na2SO3 based on a ring opening of epoxides. The influences of the contents of the porogen solvent and GMA and the various concentration ratios of 1-propanol to 1,4-butanediol in the polymerization mixture on the morphology, porosity, globule size, stability and column efficiency were investigated. The morphology and pore size distribution of the monolith were characterized by SEM and mercury intrusion porosimetry, respectively. Using only 1.5 cm length of this monolithic capillary column, four kinds of proteins, trypsin, cytochrome C, lysozyme (egg white) and egg albumin, were successfully separated from each other in 5 min at a high flow rate of 110 mm/s.

Abstract:Monodispersed core-shell silica spheres with fibrous shell structure and tunable pore size were prepared by using a one-pot oil-water biphase method. The pore size could be tuned from 7 nm to 37 nm by using organic solvents with different polarities as oil phase. The spheres synthesized by using benzene as organic solvent had the maximum pore size of 37 nm and possessed a surface area of 61 m(2) g(-1). The obtained wide pore core-shell silica spheres were applied for rapidly separating small molecules, peptides, small proteins, and large proteins with molecular weight up to 200 kDa. Since the pore size of the core-shell silica spheres was sufficiently large for the free access of all the solutes, sharp and symmetric peaks were obtained. The separation performance was as high as 264,531 plates m(-1) for fluorene. The great efficient separation demonstrates that the wide pore core-shell silica spheres have a great potential for rapid analysis of both small and large solutes with high performance liquid chromatography. Exploiting styrene-maleic acid copolymer grafting chromatographic stationary phase materials for separation of membrane lipids

Abstract:High performance liquid chromatography-mass spectrometry is one of the most commonly used strategies for lipid analysis. The development of versatile chromatographic stationary phases to meet the increasing demands for separation of complex lipids is very important. Styrene-maleic acid (SMA) copolymer is an amphiphilic polymer, which has been proven to have the ability to solubilize lipid molecules of various structures. In this study, styrene-maleic anhydride copolymer coated silica was first prepared by the thiol-ene click reaction. With L-cysteine hydrochloride or dodecanol as the post-modification reagents, Sil-SMA-amino acid and Sil-SMA-dodecanol stationary phase materials were further successfully fabricated via nucleophilic ring-opening reaction. The Fourier-transform infrared, thermogravimetric analysis, and elemental analysis results confirmed the two stationary phase materials were successfully prepared. Furthermore, both the Sil-SMA-dodecanol column and the Sil-SMA-amino acid column possessed reversed-phase/hydrophilic interaction/ion exchange mixed-mode retention mechanisms. The column efficiency of the Sil-SMA-derivatives columns reached 77,300 N/m. Based on the mixed-mode retention characteristics, the Sil-SMA-derivatives columns achieved both the lipid classes and species separation via a single column. The Sil-SMA-amino acid column was further successfully used to separate lipid extract from gastric cancer cell membrane. All these results demonstrated that the SMA-based stationary phase materials have a good potential for use in lipid separation.

Abstract:Based on a previous study of protein digestion inside the nanoreactor channels of the mesoporous molecular sieve silicate SBA-15 (Chem. Eur. J. 2005, 11: 5391), we have developed a highly efficient enrichment and subsequent tryptic digestion of proteins in SBA-15 for matrix-assisted laser desorption/ionization mass spectrometry with time-of-flight/time-of-flight analyzer (MALDI-TOF/ TOF) peptide mapping. The performance of the method is exemplified with myoglobin and cytochrome c. First, protein adsorption isotherms for two standard proteins with a range of initial concentration of proteins were investigated at room temperature. The results revealed that the kinetic adsorption rate of a protein within SBA-15 was independent of initial protein concentration, and a 15-min protein enrichment within SBA-15 could be enough for protein identification in biological samples. It was noticed that no washing steps were needed to avoid protein loss due to desorption from the mesochannels into solution. Second, protein digestion inside the channels of SBA-15 was also optimized. After adsorption of proteins into SBA-15 in 15 min, the trypsin solution (pH 8) was directly added to the SBA-15 beads with immobilized proteins by centrifugation, and then the digestion was performed for 15 min at 37 degrees C. It was observed that a higher peptide sequence covering of 98% for myoglobin was obtained by MALDI-TOF/TOF analysis, compared to in-solution digestion. So the protein digestion inside SBA-15 was proved to be significantly faster and yielded a better sequence coverage. The new procedure allows for rapid protein enrichment and digestion inside SBA-15, and has great potential for protein analysis. Copyright (c) 2006 John Wiley & Sons, Ltd.

Abstract:This paper presents the integrated results of a series of new methods for preparing mesoporous silica spheres as high-performance liquid chromatography (HPLC) packing materials. The separation performance of the mesoporous spheres materials has also been determined. Micrometer-sized silica spheres with uniform spherical morphologies and ordered mesostructures were first successfully synthesized by the method employing a water-soluble polymer-assisted assembly. Then the templates for getting ordered mesoporous materials with high-density silanol groups were removed by using hydrothermal oxidation. Finally the silica spheres were functionalized with C8 alkyl groups by surface modification under hydrothermal conditions. The resultant functionalized silica spheres were demonstrated to be excellent packing materials for HPLC.

Abstract:Highly dispersed mesostructured cellular foam silica spheres of relatively uniform micrometer size (3.5-4.5 μm) were successfully prepared using a triblock copolymer EO(20)PO(70)EO(20) as the structure-directing agent accompanied by TMB and K(2)SO(4). Both two additives have effective influence and K(2)SO(4) have better performance than other inorganic salts such as KCl and NaCl. The pore size and surface area were tuned by TMB and NH(4)F. The resultant S-MCFs were modified with C(18) group before being used as the HPLC stationary phase for effective separation of aromatic compounds and phthalic acid esters. The perfect spherical morphology and large pore volume gave rise to low and stable back pressure. High surface area, ultralarge pore size and unique pore structure would permit high flow rates and afford the possibility for fast separation.
Chitosan-based membrane chromatography for protein adsorption and separation.

Abstract:A chitosan-based membrane chromatography was set up by using natural chitosan/carboxymethylchitosan (CS/CMCS) blend membrane as the matrix. The dynamic adsorption property for protein (lysozyme as model protein) was detailed discussed with the change in pore size of the membrane, the flow rate and the initial concentration of the feed solution, and the layer of membrane in membrane stack. The best dynamic adsorption capacity of lysozyme on the CS/CMCS membrane chromatography was found to be 15.3mg/mL under the optimal flow conditions. Moreover, the CS/CMCS membrane chromatography exhibited good repeatability and reusability with the desorption efficiency of ~90%. As an application, lysozyme and ovalbumin were successfully separated from their binary mixture through the CS/CMCS membrane chromatography. This implies that such a natural chitosan-based membrane chromatography may have great potential on the bioseparation field in the future.

Abstract:Background: The fabrication technique of capillary column is the key to the development and application of capillary liquid chromatography (cLC) to improve separation efficiency for analytes. The capillary monolithic column possessed three-dimensionally connected porous or channel structures. Unique porous structure endows excellent permeability and high performance in diverse fields, especially in separation. Thereinto, organic monolithic columns have attracted widespread attention due to their advantages of simple preparation and excellent biocompatibility. However, their separation selectivity needs to be further developed and regulated to apply the separation of more diverse samples. Results: A novel polymeric monolithic column was prepared via thermally initiated in situ copolymerization of 2-methyladamantan-2-yl acrylate (MADA) with ditrimethylolpropane tetraacrylate (DTTA) in fused silica. The prepared poly(MADA-co-DTTA) monolith showed adjustable permeability, developed porous structure and high thermal stability. Consequently, it exhibited excellent separation capability of small molecules (alkylbenzenes and polycyclic aromatic hydrocarbons). Especially, when acetonitrile/water (60/40, v/v) was used as the mobile phase, the theoretical plate numbers reached 84,000 plates m-1 for butylbenzene at a linear velocity of 0.5 mm s-1. Most importantly, the hydrophobicity of the poly(MADA-co-DTTA) monolithic column was regulated via host-guest interaction between adamantyl group and cucurbit [7]uril (CB[7]). Additionally, the poly(MADA-co-DTTA) monolith was further adopted for the analysis of the tryptic digest of proteins from HeLa by cLC-MS/MS. The 33,783 unique peptides and 5,299 proteins were identified on the monolith, which exhibited great separation ability for complex samples. Significance and novelty: Due to abundant pore structure and good chemical properties, the poly(MADA-co-DTTA) monolithic column exhibited high performance for the separations of small molecules and biological sample. Meanwhile, owing to the existence of adamantyl-group, CB[7] was immobilized on the poly(MADA-co-DTTA) monolithic column to fabricate poly(MADA-co-DTTA)-CB[7] by host-guest interaction. It is possible to adjust the surface chemistry of the monolithic materials to accommodate more complex analytes.
