Abstract:Lactobacillus is the biggest genus of the lactic acid bacteria,classification and identification take an important part in the field of Lactobacillus research.However,many groups of species alike in taxonomy in the Lactobacillus are difficult to be differentiated accurately in phenotypic and physiological characteristics.In addition to that,the traditional 16S rDNA sequence homology analysis fails to identify the resembling Lactobacillus casei group and Lactobacillus plantarum group to species/sub-species level respectively.Nowadays,many advanced molecular technologies and methods are applied in identification and classification of closed lactic acid bacteria.This review refer to the related articles combined with the experiments,to summarize 9 molecular technology methods on identification and classification of L.casei group and L.plantarum group,such as sequence analysis of Protein-coding genes,Amplified Fragment Length Polymorphism (AFLP),Species-specific PCR etc.This is significantly important to research lactic acid bacteria on the polyphonic taxonomy as well as industrial production.

Abstract:Recent years have seen an explosion in the development and application of molecular tools on identification and characterization of lactic acid bacteria.Research shows that genetic variation is the root cause of genetic diversity.Detection and analysis of genetic variation can help us understand the molecular basis of various biological phenomena in lactic acid bacteria.Since the entire lactic acid bacteria kingdom cannot be covered under sequencing projects,molecular markers and their correlation to phenotypes provide us with requisite landmarks for elucidation of genetic variation.Genetic or DNA based marker techniques such as 16S rRNA,16S-23S rRNA,RAPD,PCR-PFLP,AFLP and rep-PCR are routinely being used in evolutionary,taxonomical,phylogenic and genetic studies of lactic acid bacteria.With the advancement of molecular biology,these molecular techniques have been well established and their advantages as well as limitations have been realized.Hence,the aim of this review is to provide an overview of some rapid and reliable molecular methods used for identifying and characterizing species and strains of LAB associated with food and industry.Their principles,methodologies,advantages,disadvantages as well as applicability are detailed.

Abstract:Abstraet:The accurate classification and identification of probiotic strains are the premise for further application of the strains in the development of functional foods and pharmaceuticals.For the high diversity and complexity of the microbial communities,traditional identification techniques such as morphological and cultivation characteristics as well as physiological and biochemical tests were time-consuming and in low accuracy.With the development of molecular biotechnology,the use of modem molecular markers was recognized as a new direction for classification of probiotic microbes.This paper provided an overview of several types of molecular markers that were commonly used in classification of the probiotic microbes including molecular hybridization markers,PCR amplification markers and novel molecular markers.Using classification and identification of probiotics as example,the basic principles,features and current developing status of molecular markers technology were summarized for providing the basis for further utilizations.

Abstract:Metabolomics is an important branch of systematic biology.It has received great attention around the world,especially in microbiological research field,and new progress has been also made.The metabolomics of lactic acid bacteria has become one of the hottest research topics.This review summarizes the major techniques involved in the research procedures of the metabolomics of lactic acid bacteria,including sample preparation,analysis and identification of metabolites and data analysis.The applications of the metabolomics of lactic acid bacteria are also given based on several typical examples.At the same time,we discussed the potential problems of the metabolomics of lactic acid bacteria,and points out the future of this research.

Abstract:With the advances and cost reducing of DNA-sequencing, genome determination for many microorganisms becomes available. For the research of lactic acid bacteria (LAB), this progress will not only facilitate to understand their nature of hereditary constitution, but also gradually leads to the possibility for analysis and further control of their biological functions. At present, the whole genome for 22 strains of LAB have been completed and published, at least another 12 strains of LAB are under sequencing. Based on the published data, we described the research progression of LAB genomics from four aspects, including general features, diversity of metabolism, evolution and colinearity.

Abstract:Lactic acid bacteria (LAB) are important probiotic microorganisms used in food fermentation industry.Molecular biological manipulation of these microorganisms has great potential for the development of food industry and human health.Based on the characteristics of lactic acid bacteria and the development of molecular biology,this review aims at the basic principles of a food-grade vector system for LAB,the components of a food-grade plasmid,the strategies to construct and transform a food-grade plasmid,and recent applications of food-grade plasmids.This review highlights that food-grade molecular biology could have a major positive impact on further development and application of LAB.

Abstract:Lactic acid bacteria (LAB) are widely used for the production of a variety of foods and feed raw materials where they contribute to flavor and texture of the fermented products. In addition, specific LAB strains are considered as probiotic due to their health-promoting effects in consumers. Recently, the genome sequencing of LAB is booming and the increased amount of published genomics data brings unprecedented opportunity for us to reveal the important traits of LAB. This review describes the recent progress on LAB genomics and special emphasis is placed on understanding the industry-related physiological features based on genomics analysis. Moreover, strategies to engineer metabolic capacity and stress tolerance of LAB with improved industrial performance are also discussed.

Abstract:This review deals with recent research advances on the biodiversity of Chinese traditional foods lactic acid bacteria (LAB). Traditional foods like yoghourt, kumiss, kefir and fermented camel milk are abundant in LAB. The distribution of LAB taxa is highly variable from one traditional food to another. Isolation of LAB taxa mainly depends on the sample plating methods, incubation media and conditions. It is therefore difficult to define associations between population composition and either the type of traditional food or the geographic location. Identification of isolated strains needs a ployphasic approach, including a combination of phenotypic and genotypic methods, among which the latter often based on the polymerase chain reaction (PCR) and encompassing 16S rRNA sequencing and DNA-DNA hybridizations. The main obstacle in current identification approach of LAB strains is the lack of a robust and exchangeable identification system for all LAB species. Some proposal are come up in this paper : 1) to establish a reliable and scientific system in sample collection, strain isolation and identification. 2) to isolate new characteristic strains. 3) to explore and develop new products contained LAB. 4) to intensify the research on the function and mechanism of isolated strains.

Abstract:DNA microarray technology emerged since the last century 90's allows a parallel analysis for thousands of genes in a single experiment.With the many strain's whole genome sequenceing,DNA microarray technology as a primary high-throughput tool has been widely applicated in monitoring of gene expression patterns,such as gene function prediction,detection of gene mutations,polymorphism analysis and so on.More attention has been taken on the application of gene chip technology in the research of lactic acid bacteria(LAB),as for several LAB strains genome sequencing completed and a lots of sequences of EST,16S/16S-23S rDNA obtained.The basic principle of the gene chip and the application progress of DNA microarray in identification and expression analysis of LAB were introduced,which would provide reference for further application and exploitation of LAB DNA microarray.

Abstract:The bacteria which can ferment carbohydrates to produce large amounts of lactic acid are called as lactic acid bacteria.They are probiotics beneficial to people and livestock and poultry.Lactic acid bacteria of animal intestines have a better adaptability because it has resistance to the acid.It could survive easily in the gut to resist pathogenic microorganism adhesion.This paper reviewed several methods of isolation and cultivation of lactic acid bacteria of animal intestines and summarized the better media to isolate lactic acid bacteria.This report summarized the methods to identificate actic acid bacteria in the intestines of animals, including morphological identification,biochemical identification and molecular biological identification methods,and focused on the 16 S rDNA of molecular biological identification.It has important significance to establish the methods for isolation and identification of lactic acid bacteria in the animal intestines and study the composition of microbial flora in the intestines of animals.
