Homocysteine Induces Activation of the Signal Pathway of Protein Kinase C and NF-κB in Endothelial Cells

Abstract:Objective To explore the role of the signal pathway of protein kinase C(PKC) and nuclear factor-κB(NF-κB)in regulating the pathogenic process of hyperhomocysteinemia.Methods Human umbilical vein endothelial cells(HUVEC) were treated with Hcy,with or without PKC inhibitor,for different periods of time.Dynamic changes of NF-κB activation were measured by electrophoretic mobility shift assay(EMSA) and TransAMTM NF-κB P65 assay system.Western blotting was performed to detect protein expression.Results Treatment with high concentrations of Hcy induced HUVEC NF-κB activation accompanied by an increased level of phosphorylated PKC expression.The NF-κB activation reached its maximum after stimulation by high concentrations of Hcy for 30 min and 6 h.Meanwhile PKC reached its maximum after stimulation by Hcy for 30 min.Western blotting showed that Hcy-induced NF-κB activation was due to the increased phosphorylation of the inhibitory protein(IκBα),while PKC inhibitor almost completely blocked the NF-κB activation as well as the phosphorylation of IκBα.Conclusion Hcy may induce the activation of the signal pathway of PKC and NF-κB in endothelial cells and PKC inhibitor can block the above effects significantly.
