Genetic structure and geographic variance of the Tibetian snowcock Przewalsky’s subspecies Tetraogallus tibetanus przewalskii populations

Abstract:Using polymerase chain reaction(PCR) and direct sequencing methods to analyze the genetic structure and molecular phylogeography of the Tibetian snowcock Tetraogallus tibetanus przewalskii in China, a total 535 bp nucleotides of the mitochondrial DNA (mtDNA) Cyt b genes were sequenced in 51 Tibetian snowcock collected from four geographic populations in the north mountains of Qinghai-Tibetian Plataeu. We defined four geographic regions: Qilian mountain (QL), Chaidamu basin (CD), Kunlun mountain(KL) and Tanggula mountain (TG) based on the geographic distribution of samples. Among the comparable sequences of 535 bp,15 nucleotide sites were variable(2.8% of the entire sequences), and defined 13 haplotypes(25.49% of all the samples). There was high haplotype polymorphism in four geographic populations. The result indicated that the average genetic distance among all haplotypes was 0.23%, and the genetic variation between and within geographic populations were 80.75% and 19.25%, respectively. Molecular variance analysis(AMOVA) and phylogenetic tree analysis demonstrated that haplotype was shared among geographic populations, no geographic clustering was observed, there was distinctive geographic phylogeography between QL and KL, CD (Φ_ ST was 0.345 and 0.365, respectively,P<0.01), but no obvious subdivision among other populations(P>0.05), We estimated the average split time of the haplotypes to be 3.8 million years ago (MYA) from genetic data. Therefore, we suggest that the present distribution pattern of Tibetian snowcock in China results from recent population expansion based on the above-mentioned results.
