Abstract:Several new 1,3,4-oxadiazole derivatives containing a pyrazole ring were designed and synthesized from ethyl acetoacetate and triethyl orthoformate as starting materials via multi-step reactions. The compound structures were confirmed by melting point, 1 H NMR and HRMS. They were evaluated for fungicidal and herbicidal activities. Four of the compounds exhibited moderate fungicidal activity against Colletotrichum species. Most of the compounds had moderate-to-good activity as a herbicide.

Abstract:Natural products are a source of many novel compounds with biological activity for the discovery of new pesticides and pharmaceuticals. Quinoxaline is a fused N-heterocycle in many natural products and synthetic compounds, and seven novel quinoxaline derivatives were designed and synthesized via three steps. Pesticidal activities of title quinoxaline derivatives were bioassayed. Most of these compounds had herbicidal, fungicidal, and insecticidal activities. The compounds 2-(6-methoxy-2-oxo-3-phenylquinoxalin-1(2H)-yl)acetonitrile (3f) and 1-allyl-6-methoxy-3-phenylquinoxalin-2(1H)-one (3g) were the most active herbicides and fungicides. Mode-of-action studies indicated that 3f is a protoprophyrinogen oxidase-inhibiting herbicide. Compound 3f also possessed broad-spectrum fungicidal activity against the plant pathogen Colletotrichum species. Some of these compounds also had insecticidal activity. Molecular docking and DFT analysis can potentially be used to design more active compounds.

Abstract:Seven new quinoxaline derivatives 3a-3g were synthesized through two steps convenient approach. Their structures were confirmed by Fourier transform infrared, proton nuclear magnetic resonance, and high-resolution mass spectrometry. The synthesized compounds 3a-3g showed good fluorescent intensity in dimethyl sulfoxide. They were also shown to possess good antifungal activities. The structure-activity relationship was studied by density functional theory calculations.

The structures of the products obtained are characterized by elemental analyses and spectral data and the fragmentation pattern of the mass spectra of 3a-j is discussed. 3a-j have been screened for their antibacterial activity against B. subtilis and E. coli.

Abstract:A series of novel 6-aryl-3-(1-p-chlorophenyl-5-methyl-1,2,3-triazol-4-yl)-s-triazolo-[3,4-b]-1,3,4-thiadiazoles 6a-m have been synthesized in 49-64% yields by the cyclocondensation of 4-amino-3-(1-p-chlorophenyl-5-methyl-1,2,3-triazol-4-yl)-5-mercapto-1,2,4-triazole 5 with various aromatic acids in the presence of POCl3. Biological activity of 6a-m was studied.

Abstract:Ten novel benzoyl ureas containing a 1,3,4-thiadiazole group were synthesized by the reaction of 2-amino-5-substituted phenyl-1,3,4-thiadiazole and 2,6-difluorobenzoyl isocyanate, and structurally confirmed by H-1 NMR, IR, ESI-MS techniques and elemental analysis. Preliminary bioassay test indicated that some compounds showed insecticidal activity. Compounds 3d (3,5-(CH3)(2)) and 3g (4-C4H9) exhibit good insecticidal activity against Aphis craccivora with higer than 90% mortality.

Abstract:Aim To design and synthesize a series of the triazole propanol derivatives containing nitrogen,and to study their antifungal activities.Methods The target compounds were synthesized from 1,3 difluorobenzene in four steps.Results Eleven new compounds were obtained and their structures were confirmed by IR and 1H NMR.Conclusion Among the eleven new compounds,ten compounds had antifungal activities to different degrees.

Abstract:BACKGROUND: Chemical pesticides are the main measures for pest control, but have caused growing resistance of pests and brought a series of environmental problems. Development of high-efficient insecticidal molecules with novel scaffolds is therefore particularly urgent. RESULTS: Based on a [5 + 1] annulation reaction with 5-amino-1H-phenylpyrazole and dialkyl bromomalonate, 27 novel five-substituted 4,5-dihydropyrazolo[1,5-a]quinazolines were designed following the intermediate derivatization method and synthesized. Bioassay results indicated that most of the test compounds displayed good insecticidal activities against Plutella xylostella, Spodoptera frugiperda, and Solenopsis invicta. In particular, the insecticidal activities of compounds 4a, 4f, and 4m against P. xylostella [median lethal concentration (LC50) values ranged from 3.87 to 5.10 mg L-1] were comparable to that of indoxacarb (LC50 = 4.82 mg L-1). In addition, compounds 4a and 9e showed similar high insecticidal activities against Spodoptera frugiperda (mortality rate = 79.63% and 72.12%) at 100 mg L-1, comparable to that of fipronil (mortality rate: 68.44%); compound 9a showed possible delayed toxicity against Solenopsis invicta (mortality rate: 95.66%) after 5 days of treatment at 1.0 mg L-1. CONCLUSION: Due to their high insecticidal activities against P. xylostella, compound 4m, 4a, and 4f could be considered as qualified candidates for novel insecticide. Several other 4,5-dihydropyrazolo[1,5-a]quinazolines with relatively high bioactivity, such as compounds 9a and 9e, are also worth further optimization as potential insecticide or anticide candidates.

Abstract:Five N'-(substituted pyrimidin-2-yl)-N-dichlorochrysanthemoyl thioureas (5a-5e) were synthesized from corresponding dichlorochrysanthemic acid in three steps. And two dichlorochrysanthemoylimino-2H-1,2,4-thiadiazolo[2,3-a]pyrimidines were prepared by oxidizing cyclization of 5a, 5b. The structures of these novel target compounds were confirmed by IR, H-1 NMR and elemental analyses. The preliminary biological activity tests indicated that some target compounds had better insecticidal activity, fungicidal activity and so on mulriple efficacies.

Abstract:Five new 5,7-disubstituted-2-(2,4-dichlorophenoxyacetylimino)-2H-1,2,4-thiadiazolo[2,3-a]pyrmidines were synthesized, and their structures were confirmed by H-1 NMR and elemental analysis. Biological test showed that some of the target compounds displayed herbicidal activity to some extent. In order to investigate the relationship between the structure of these compounds and their herbicidal activities, a crystal structure of the target compound 5b was determined by X-ray diffraction analysis.

Abstract:A series of ( E )-5-{[(1 H -1,2,3-triazol-5-yl)sulfanyl]methyl}-4-(benzylideneamino)-4 H -1,2,4-triazole-3-thiols were synthesized using 4-amino-5-{[(1 H -1,2,3-triazol-5-yl)sulfanyl]methyl}-4 H -1,2,4-triazole-3-thiol as an intermediate compound. The structure of the newly synthesized compounds was confirmed by 1 H and 13 C NMR, IR, and electrospray ionization mass spectra. Six compounds showed promising insecticidal activity against Plodia interpunctella .

Abstract:Fourteen new substituted pyrimidines were synthesized and their structures were confirmed by H-1 NMR, IR, MS spectra and elemental analyses. Their insecticidal, fungicidal and herbicidal activities were examined. Some of these compounds showed good fungicidal activity after introduction of dimethylamino into the 2-position of pyrimidine ring. The compound with the phenoxy substructure at the 4-position of pyrimidine ring also showed good fungicidal activity, e. g. 3b, 3c and 3e. But compounds with methyl at the 5-position on the pyrimidine ring diminished the fungicidal activity. The best substituent on phenyl ring was 2-nitro-4-trifluoromethyl.

Abstract:A facile approach for the synthesis of pyrimidine derivatives containing 1,2,4-triazole moiety via the reaction of substituted 2-phenoxypropane hydrazide with ethyl-N-(5-cyano-2-alkylthio-5-cyano-6-methylthiopyrimidin-4-yl) formimidate in refluxing 2-methoxyethanol is reported. The structures of the title compounds were identified by 1H NMR, 13C NMR, and HRMS. The configuration of compound 5a was confirmed by X-ray crystallography analysis. The preliminary herbicidal test indicated that some of the target compounds exhibited good inhibition activities against Brassica napus (rape) and Echinochloa crusgalli (barnyard grass).Graphical

Abstract:Twenty six novel pyrimidin-4-amine derivatives containing the 1,2,4-oxadiazole motif were synthesized. Their chemical structures were confirmed by 1H nuclear magnetic resonance (NMR), 13C NMR, and highresolution mass spectrography. The insecticidal activity results indicated that some of them possessed excellent insecticidal activity (100%) against Mythimna separate, especially for compounds 6d, 6f, 6o, 6w, 6y and 6z. These compounds exhibited no activity against the insects Aphis medicagini and Tetranychus cinnabarinus. The structure-insecticidal activity relationships are discussed. Density functional theory analysis can potentially be used to design more active compounds. These results provide useful insecticide design information for further optimization.

Abstract:A series of thiazolo[3,2-a]pyrimidine derivatives were synthesized from the reaction of α-bromoacetophenone, aromatic aldehyde, malononitrile and thiourea which was catalyzed by Tröger's base derivative 5,12-dimethyl-3,10-diphenylbis-1H-pyrazol[b,f][4,5]-1,5diazadicyclo[3.3.1]-2,6-octadiene (1). The reaction mechanism was discussed by the H NMR analysis and chemical experiments. The pharmacological activity results of the products indicated that most of products showed high inhibitory on one or more cancer cells in human hepatocarcinoma cell (HepG2), human non-small cell lung cancer cell (247) and human non-small cell lung cancer cell (A549) in vitro. And seven products have antibacterial activity against the methicillin-resistant Staphylococcus aureus. These results showed the great potential of these compounds in drug development.

Abstract:A series of novel 1,2,4-triazole derivatives containing oxime ether and phenoxy pyridine moiety were designed and synthesized. The new compounds were identified by nuclear magnetic resonance (NMR) spectroscopy and high-resolution mass spectrometry (HRMS). Compound (Z)-1-(6-(4-nitrophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-methyl oxime (5a18) was further confirmed by X-ray single crystal diffraction. Their antifungal activities were evaluated against eight phytopathogens. The in vitro bioassays indicated that most of the title compounds displayed moderate to high fungicidal activities. Compound (Z)-1-(6-(4-bromo-2-chlorophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-methyl oxime (5a4) exhibited a broad-spectrum antifungal activities with the EC50 values of 1.59, 0.46, 0.27 and 11.39 mg/L against S. sclerotiorum, P. infestans, R. solani and B. cinerea, respectively. Compound (Z)-1-(6-(2-chlorophenoxy)pyridin-3-yl)-2-(1H-1,2,4-triazol-1-yl)ethan-1-one O-benzyl oxime (5b2) provided the lowest EC50 value of 0.12 mg/L against S. sclerotiorum, which were comparable to the commercialized difenoconazole. Moreover, homologous modeling and molecular docking disclosed possible binding modes of compounds 5a4 and 5b2 with CYP51. This work provided useful guidance for the discovery of new 1,2,4-triazole fungicides.
