The molecular biology identification of a hydrolyzed polycrylamide (HPAM) degrading bacreria strain H5 and biodegradation product analysis

Abstract:A hydrolyzed polyacrylamide (HPAM)-degrading bacteria strain H5 was isolated from curing pot in HPAM distribution station of Daqing Oil Field using Hungate Anaerobic Technique. The pictures of Atomic Force Microscope(AFM) shown that this is long bacillus with width 0.32~2.9μm and length 0.53~5.12μm,which can reduce sulfide and produce H2S, strictly anaerobic bacteria. Alignment of 16S ribosomal DNA sequence suggests that comparability between the bacteria H5 and Clostridium bifermentans(AY781385) reaches to 99%. Fatty acid distributed at iso-C15:0 ,C15:0,C16:0,C16:1ω7cand iso-C17:0,content being 39.41 %,7.38 %,10.90%,8.75 % and 7.11% respectively,73.55% of all fatty acid. The isolate was identified as a new species belongs to fusobacterium, temporarily named Clostridium bifermentans H5. Analysis results of Infrared Spectroscopy (IR) showed that the bacteria can use HPAM as only carbon source, hydrolyzing amide to carboxyl group by the molecule chain amide hydrolysis, degrading the side-chain and changing some function group, resulting in obviously decreased viscosity. Using GC-MS, determined HPAM degrading products of low-molecular weight compounds were polyacrylamide fragments with duplet bond, epoxy as well as carbonyl group, but most of them were acrylamide oligomer derivatives.The removing efficiency for HPAM reached 52.5%.
