Abstract:The determination of nitroaromatic compounds in aqueous solution was investigated at beta-cyclodextrin (beta-CD)/silver nanoparticle (AgNPs) composite modified ITO electrodes. This method relies on the different reduction potentials for the various nitroaromatic isomers, the different binding strengths of the nitroaromatic isomer guests to the beta-CD host, and excellent electron transfer ability of AgNPs. After incubation in a solution with different single nitroaromatic compounds, reduction peaks in the range from -550 to -913 mV were observed at the modified electrode, depending on the nitroaromatic compound present. The sensor exhibited selectivity for some isomers in a solution containing a mixture of nitroaromatic compounds. In particular, the sensor shows specificity for 4-nitroaniline and 1-chloro-2-nitrobenzene over other nitroaniline isomers and nitrochlorobenzene isomers, respectively. The results show that all the nitroaromatic compounds, 2-nitroaniline, 3-nitroaniline, 4-nitroaniline, 1-chloro-2-nitrobenzene, 1-chloro-3-nitrobenzene, and 1-chloro-4-nitrobenzene, species.
Determination of 14 nitrosamines at nanogram per liter levels in drinking water.

Abstract:N-Nitrosamines, probable human carcinogens, are a group of disinfection byproducts under consideration for drinking water regulation. Currently, no method can determine trace levels of alkyl and tobacco-specific nitrosamines (TSNAs) of varying physical and chemical properties in water by a single analysis. To tackle this difficulty, we developed a single solid-phase extraction (SPE) method with high-performance liquid chromatographytandem mass spectrometry (HPLCMS/MS) for the determination of 14 nitrosamines of health concern with widely differing properties. We made a cartridge composed of a vinyl/divinylbenzene polymer that efficiently concentrated the 14 nitrosamines in 100 mL of water (in contrast to 500 mL in other methods). This single SPEHPLCMS/MS technique provided calculated method detection limits of 0.01-2.7 ng/L and recoveries of 5393% for the 14 nitrosamines. We have successfully demonstrated that this method can determine the presence or absence of the 14 nitrosamines in drinking water systems (eight were evaluated in Canada and the U.S.), with occurrence similar to that in other surveys. N-Nitrosodimethylamine (NDMA), N-nitrosodiphenylamine, and the TSNA 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanol were identified and quantified in authentic drinking water. Formation potential (FP) tests demonstrated that NDMA and TSNA precursors were present in (1) water samples in which tobacco was leached and (2) wastewater-impacted drinking water. Our results showed that prechlorination or ozonation destroyed most of the nitrosamine precursors in water. Our new single method determination of alkylnitrosamines and TSNAs significantly reduced the time and resource demands of analysis and will enable other studies to more efficiently study precursor sources, formation mechanisms, and removal techniques. It will be useful for human exposure and health risk assessments of nitrosamines in drinking water.
Confirmation of N-(nitrosomethyl) Urea As a Nitrosourea Derived by Nitrosation of Fish Sauce.

Abstract:N-(nitrosomethyl) urea (NMU) was characterized in the carcinogenic nitrosated fish sauce (NFS) with high performance liquid chromatography (HPLC)-photohydrolysis-pyrolysis-thermal energy analysis recently. We used HPLC-electronic spray ionization-mass spectrometry and HPLC-diode array detection to confirm NMU in NFS further. It was observed that the corresponding chromatographic fraction of NMU of NFS showed the same mass spectrum (m/z 64, 102, and 145) and ultraviolet-absorbance (lambda max = 230 nm) as those of authentic NMU. These results confirmed that the component of NFS was NMU.

Abstract:To characterize chemical carcinogens in acidic-nitrosated fish sauce sample, N-nitrosamides in the sample were separated by two kinds of reversed-phase HPLC columns, and with detection by photolysis-pyrolysis-thermal energy analyzer. A strong chromatographic peak at t(R) 12 or 4.5 min, same as that for N-(nitrosomethyl)urea (NMU), was obtained on PRP-1 or C-18 HPLC column from fish sauce sample with 10 mM trifluoroacetic acid as the basic mobile; acetonitrile, as organic modifier after the sample was nitrosated by 5 mmol/L of sodium nitrite (final concentration) at 37 degrees C and pH 2.0 for 1 h. No response above t(R) could be observed from the nitrosated sample in the detection system without photolysis. Such a peak could not be obtained from the unnitrosated fish sauce either. These results indicated that the component was NMU. Furthermore, this component, NMU, could also be detected in the nitrosated human gastric juice sample spiked with fish sauce. The formation of NMU in the sample was pH- and nitrite-dependent. This paper provides direct evidence that NMU formation could occur in fish sauce from the high-risk area for stomach cancer and in the fish sauce spiked human gastric juice during nitrosation under simulated gastric conditions.

Abstract:The tobacco-specific nitrosamine 4-(methylnitrosamino)-1-(3-pyridyl)-1-butanone (NNK) is classified as a Group 1 human carcinogen. It is metabolically activated by P450 enzymes to intermediate methylate and pyridyloxobutylate DNA, resulting in the formation of DNA adduct that is critical for the carcinogenicity of NNK. To directly and objectively examine the DNA adduct formation profiles without the complexity of factors in vivo, in the present study, five kinds of methyl DNA adducts were first identified in the incubation model of NNK established with human lung epithelial cells (BEAS-2B). The level of methyl DNA adducts and metabolites of NNK were quantitatively analyzed, respectively. With the increase of exposure time and dose, the level of methyl DNA adducts and metabolites increased. Furthermore, with the changes of the activity of P450 enzymes, which is the main enzyme regulating the α-hydroxylation of NNK, we found the levels of both methyl adducts and metabolites formed via α-hydroxylation in experimental groups showed the same trend compared with those in control group, while the metabolites formed via other pathways changed in the opposite trend. The result proves that the methyl adducts induced by NNK generate via α-hydroxylation pathway in BEAS-2B cells.

Abstract:Nitrosamine is one of the three strong carcinogenic contaminants in food and widely present in pickled and heat-treated food.A new solid phase extraction(SPE)-HPLC method based on the HPLC-MS identification for the determination of nine nitrosamines has been established.The method could be used for the direct and fast detection of 9 nitrosamines in pickles.The conditions for chromatographic separation and detection have been optimized.Four SPE columns with single packing materials(Al2O3,C18,Silica,and Diatomite) and two SPE columns with mixed dual packing materials(Al2O3-C18,Al2O3-Silica) have also been investigated.As a result,the recoveries for determination of 9 nitrosamines by HPLC with SPE column of mixed Al2O3-C18 for cleanup were in the range of 75.7% to 116.0% and the limit of detection was 0.20-0.49 mg/L.On the basis of the above methods,the concentrations of nitrosamines in pickled vegetables of six varieties produced from different origins were investigated.The occurrence rate of N-nitrosodiethylamine(NDMA) and NDEA in all samples was 37.5% and 25.0%,relatively higher than that of other nitrosamines.The highest content of NDEA occurred especially in potherb mustard was up to 59.1 mg/L,and N-nitrosomorpholine(NMOR) has not been detected in all samples.

Abstract:Nitrosamines (NAs) as a group of emerging nitrogenous disinfection byproducts were present in drinking water at ng/L levels. Accurate measurements of NAs at such a trace level in samples is a challenging task. Solid phase extraction (SPE), which is used in the sample pretreatment, plays a critical role in the analysis of NAs in water. In this study, a highly selective and sensitive method for the determination of five less polar NAs, namely nitrosodiethylamine nitrosopiperidine, nitrosodi-n-propylamine, nitrosodibutylamine and nitrosodiphenylamine, in water and beverage samples was developed. A new molecularly imprinted polymer (MIP) was synthesized and used as sorbents in SPE for the sample preparation. Prepared samples were analyzed using high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). Satisfactory recoveries were obtained at three different concentrations (5, 20, and 50 ng/L, n = 3) in the range of 93-107% with relative standard deviations of 3.1-9.8%. Limit of detection and limit of quantitation for the five NAs were in the range of 0.2-0.7 ng/L and 0.6-2.1 ng/L, respectively. Method precisions ranged from 4.9% to 10.5%. This novel method of MIP-SPE coupled with HPLC-MS/MS was successfully applied to the determination of these five NAs in different types of water and beverages samples.

Abstract:A method was developed to separate the tobacco-specific nitrosamines N'-nitrosonornicotine (NNN), 4-(N-nitrosomethylamino)-1-(3-pyridyl)-1-butanone, and other two N-nitrosamines N-nitrosodimethylamine, N-nitrosopyrrolidine which exist in tobacco by high performance liquid chromatography photohydrolysis-pyrolysis energy analysis. The ratio of peak areas of NNN and NaNO2 with concentration of NNN presents linear relationship in the range of 1 similar to 10 mg/L. The absolute detection limit is 6ng. Tobacco sample has been separated and determined, the concentration of NNN in the sample was 5.4 mg/L. The method is advisable, efficient to quantify NNN in tobacco.

Abstract:As highly toxic substances, N-nitrosamines (NAs) have been proved to cause carcinogenesis and mutagenesis in humans. Therefore, to carefully monitor safety and preserve human health, the development of rapid, accurate, and high-sensitivity determination methods of NAs is of substantial importance. This review provides a current-status comprehensive summary of the pretreatment and determination methods of NAs in various samples since 2010. Common pretreatment methods that have been used to extract and purify targets include solid-phase extraction, liquid-liquid extraction and various microextraction methods, such as solid-phase microextraction and liquid-phase microextraction, among others. Determination methods include liquid chromatography, gas chromatography, supercritical fluid chromatography and electrochemical methods, among others. In addition, we discuss and compare the advantages and disadvantages of various pretreatment and analytical methods and examine the prospects in this area.

Abstract:Herein, we develop a novel method using gas chromatography-ion mobility spectrometry (IMS) with an ammonia dopant for the determination of volatile N-nitrosamines in meat products. The IMS system was implemented with Fourier deconvolution multiplexing to simultaneously improve the resolving power and sensitivity. The ammonia dopant mitigated the formation of N-nitrosamine dimer ions, suppressing the ionization of interferents and shifting the reactant peak from hydrated protons to hydrated ammonium ions. The ammonium adduct product ions of N-nitrosamines were confirmed using time-of-flight mass spectrometry. The instrument limits of detection and quantitation for nitrosamines were 0.78-1.79 and 2.60-5.82 ng/mL, respectively, with recoveries between 71.39% and 110.59% using a simple water distillation method. The developed method showed satisfactory performance when applied to the detection of nitrosamines in various meat products.

Abstract:Increasing concerns have been raised about dangerous, yet nearly undetectable levels of nitrosamines in foods, medications, and drinking water. Their ubiquitous presence and carcinogenicity necessitates a method of sensitive and selective detection of these potent toxins. While the detection of two major nitrosamines─N-nitrosodimethylamine and N-nitrosodiethylamine─has seen success, low detection limits are scarcer for the other members of this class. One member, N-nitrosodiphenylamine (NDPhA), has had little progress not only in its detection in low quantities but also in its detection at all. NDPhA has unique difficulty in its identification due to its aromaticity, making it far more problematic to distinguish in the common GC-MS or LC-MS/MS methods used for nitrosamine sensing. Despite this detection barrier, it has been listed among the top 6 carcinogenic nitrosamines by the Food and Drug Administration as of 2023. Due to its evasive nature, a unique methodology must be applied to facilitate its sensitive identification. Herein, we describe the use of surface-enhanced Raman spectroscopy for the first account of liquid-phase detection of NDPhA using cysteamine-functionalized gold nanostars and a portable Raman spectrometer. Our methodology requires no chemical modification to the nitrosated structure as well as the usage of two well-understood biocompatible materials: cysteamine and gold nanoparticles.

Abstract:Nitrite is a common food additive, however, its reduction product, nitrosamine, is a strong carcinogen, and hence the ultra-sensitive detection of nitrite is an effective means to prevent related cancers. In this study, different sized gold nanoparticles (AuNPs) were modified with P-aminothiophenol (ATP) and naphthylethylenediamine (NED). In the presence of nitrite, satellite-like AuNPs aggregates formed via the diazotization coupling reaction and the color of the system was changed by the functionalized AuNPs aggregates. The carcinogenic nitrite content could be detected by colorimetry according to the change in the system color. The linear concentration range of sodium nitrite was 0-1.0 μg mL-1 and the detection limit was determined to be 3.0 ng mL-1. Compared with the traditional method, this method has the advantages of high sensitivity, low detection limit, good selectivity and can significantly lower the naked-eye detection limit to 3.0 ng mL-1. In addition, this method is suitable for the determination of nitrite in various foods. We think this novel designed highly sensitive nitrate nanosensor holds great market potential.
Derivatization Method for Determination of Nitrosamines by GC–MS

Abstract:Gas chromatography/low-resolution mass spectrometry with electron impact source was applied to detect eight nitrosamines (NAs). NAs were first denitrosated by a mixed solution of hydrobromic and acetic acids, followed by sulfonylation with p -toluenesulfonyl chloride. Variables affecting the denitrosation and sulfonylation, such as temperature, time, pH and reagent concentration, were optimized. Comparison of the determination of NAs with and without derivatization was performed. Results showed that better chromatographic behavior and larger mass response were obtained after derivatization, and instrumental detection limits ranged from 0.016 to 0.053 ng, a nearly 20-fold decrease of those without derivatization. The proposed method provided an alternative to performing accurate qualitative and quantitative analysis of NAs with expensive high-resolution mass spectrometry or thermal energy analyzer.

Abstract:Halloysite nanotubes loaded with 4-aminothiophenol capped silver nanoparticles (mHNTs-AgNPs4-ATP) composite was synthesized as a sensitive surface-enhanced Raman scattering (SERS) probe for the determination of nitrite ions in sausage and pork luncheon meat. The as-prepared composite was characterized by transmission electron microscope, X-ray diffraction spectroscopy and Fourier transform infrared spectroscopy. The as-synthesized mHNTs-AgNPs4-ATP composite with evenly distribution of AgNPs can provide in-situ derivatization site for sensitive and selective SERS detection of nitrite ions. Under acid condition, the 4-ATP capped on the AgNPs can be transformed into p,p'-dimercaptoazobenzene (DMAB) through nitrite-triggered diazo reaction. This efficient nitrite-triggered reaction can be used to detect nitrite via the characteristic peaks of DMAB at 1143 cm-1, 1392 cm-1, and 1434 cm-1. This SERS method has a wide logarithmic range of 0.0069-6.9 mg L-1, with detection limit of 0.78, 3.4, 0.51 μg L-1 at the peak of 1143 cm-1, 1392 cm-1, and 1434 cm-1, respectively. Besides, this method can be applied to detect nitrite ions in sausage and pork luncheon meat with relative standard deviation less than 10.3%, and the results were consistent with that analyzed by UV-Vis method. This method has good potential in efficient detection of meat product in food safety.
Role of Nitrosamides in the High Risk for Gastric Cancer in China.

Abstract:Gastric cancer is the leading cause of death from cancer in China. Samples of fish sauce, a traditional seasoning, were collected in the high-risk area for gastric cancer in the Fuzhou area, Fujian Province. When fish sauce samples were nitrosated at pH 2.0, direct mutagenicity and high contents of N-nitrosamide were detected (30.9-78.0 microM); the N-nitrosamide content of three samples of fish sauce made in Guangdong and purchased from a market outside Fujian were low (2.1-6.0 microns). When the nitrosated fish sauce extract was given to newborn rats by gavage, dysplasia and adenocarcinoma were induced in the glandular stomach in the 4th and 16th experimental week, respectively. N-Nitrosamides were also found in fasting gastric juice from patients with chronic gastritis in the high-risk area of Putian. The mean concentration of total N-nitrosamides in the extracts correlated with the severity of gastritis in the stomach. These findings indicate that N-nitrosamides may play an important role in causing gastric cancer in China.

Abstract:The ability to effectively detect N-nitrosamine compounds by liquid chromatography–tandem mass spectrometry presents a challenge due to the problems of high detection limits and difficulty in simultaneous N-nitrosamine compound detection. In order to overcome these limitations, this study reduced the detection limit of N-nitrosamine compounds by applying n-hexane pre-treatment to remove non-polar impurities before the conventional process of column extraction. In addition, ammonium acetate was used as the mobile phase to enhance the retention of nitrosamine target substances on the chromatographic column, with formic acid added to the mobile phase to improve the ionization level of N-nitrosodiphenylamine, to achieve the simultaneous detection of multiple N-nitrosamine compounds. Applying these modifications to the established detection method allowed the rapid and accurate detection of N-nitrosamine in water within 12 min. The linear relationship, detection limit, quantification limit and sample spiked recovery rate of nine types of nitrosamine compound were investigated, showing that the correlation coefficient ranged from 0.9985–0.9999, while the detection limits of the instrument and the method were 0.280–0.928 μg·L−1 and 1.12–3.71 ng·L−1, respectively. The spiked sample recovery rate ranged from 64.2–83.0%, with a standard deviation of 2.07–8.52%, meeting the requirements for trace analysis. The method was applied to the detection of N-nitrosamine compounds in nine groundwater samples in Wuhan, China, and showed that the concentrations of N-nitrosodimethylamine and NDEA were relatively high, highlighting the need to monitor water bodies with very low levels of pollutants and identify those requiring treatment.

Abstract:To set up effective analysis method is important for environmental monitoring. In this work, the 4-aminophenol functionalized nitrogen and sulfur co-doped carbon quantum dots (AP-NSCDs) were synthesized based on the functionalization of NSCDs obtained by one-step hydrothermal method and used as a new fluorescent sensor for the determination of nitroalkene pesticide and medicine, such as nitenpyram, nizatidine and ranitidine. Nitroalkenes could quench the fluorescence of the functionalized carbon dots due to the fluorescence resonance energy transfer (FRET) principle. The sensor showed high sensitivity, selectivity, and stability, with a wide linear range of 0.05 ∼ 10 mg/L and a low detection limit of 0.02 mg/L. This sensor was successfully employed to detect nitenpyram in rice, orange and cabbage, and to determine nizatidine and ranitidine in serum, urine and river water as well. The developed method was rapid and sensitive, and had potential for monitoring the nitroalkene compounds in the environment and food.
