Adaptation of Respiratory Metabolism to Sulfide Exposure in Urechis unicinctus

Abstract:In this paper,the changes of cytochrome c oxidase(CCO),succcinate dehydrogenase(SDH),and fumarate reductase(FRD) activities in tissues of Urechis unicinctus exposed to different concentrations of sulfide were investigated to understand its respiratory metabolism adaptation to sulfide exposure.Two sulfide groups containing sulfide concentrations of 50 and 150 μmol/L and one control group with no sulfide were respectively set and worm tissues(the body wall and the hindgut) were sampled at 0,2,12,24 and 48 h after sulfide exposure and 48 h after exposure removal.The results showed that the CCO activities increased in the sulfide groups at 2 h and those in the 150 μmol/L sulfide group were significantly lower than the control group 24 h after sulfide exposure.At 48 h,the CCO activities in the 150 μmol/L sulfide group nearly approximated 0 and the SDH activities were significantly lower,while the FRD activities were significantly higher compared to the control group.The above results indicate that exposed to sulfide for a short time,the worm increases its respiratory metabolism,which is probably due to sulfide oxidation.With the duration of sulfide exposure,the respiratory metabolism of the worm decreases.The anaerobic metabolism mode in which fumarate is reduced to succinate may exist when aerobic metabolism is inhibited completely by sulfide.In addition,U.unicinctus is capable of self-recovering after exposure removal.
