Abstract:The generation of thioesters through thioesterification of carboxylic acid has significant synthetic value given the easy availability of various carboxylic acids and the extensive application of thioesters. An economical and rapid thioester synthesis approach is still needed. Herein, we achieved thioesterification of carboxylic acids with valuable glycosyl thiol, aromatic and aliphatic thiols mediated by sulfonyl fluoride (SO2F2). The compatibility of numerous carboxylic acid substrates was tested, including (hetero)aromatic and aliphatic carboxylic acids and the reaction proceeded smoothly under mild conditions at good yields. Late‐stage modification of drug molecules rac‐Naproxen and Loxoprofen proved feasible. Besides, the gram‐scale reaction demonstrated the utility of this protocol.

Abstract:An efficient procedure was developed for direct preparation of aryl-substituted lactones from corresponding aryl carboxylic acids, which was promoted by ammonium iodide. In this protocol, aryl carboxylic acids were treated with ammonium iodide, potassium bromide, and oxidant Oxone in a mixture of acetonitrile and 2,2,2-trifluoroethanol (6:4) at room temperature for 12 h, resulting in corresponding aryl lactones in moderate to good yields.

Abstract:An efficient one-step synthesis of diarylacetic acids was successfully performed by electrochemical direct carboxylation of diarylmethanol compounds in DMSO. Constant-current electrolysis of diarylmethanol species in DMSO using a one-compartment cell equipped with a Pt cathode and a Mg anode in the presence of carbon dioxide induced reductive C(sp3)-O bond cleavage at the benzylic position in diarylmethanol compounds and subsequent fixation of carbon dioxide to produce diarylacetic acids in good yield. This protocol provides a novel and simple approach to diarylacetic acids from diarylmethanol species and carbon dioxide without transformation of the hydroxy group into appropriate leaving groups, such as halides and esters including carbonates.

Abstract:The novel efficient procedure has been developed for the oxathioacetalization of carbonyl compounds and 2-mercaptoethanol using the novel carbon-based sulfonic acid as catalyst under solvent-free condition at room temperature. The results showed that the novel catalyst was very efficient for the reactions with good to excellent yields in short time. The novel catalyst owned many advantages such as operational simplicity, without need of any solvent, small amount of usage, low cost of the catalyst used, high yields, applicability to large-scale reactions, reusability and chemoselectivity over the traditional catalysts, which made the catalyst one of the best choices for the reactions.

Abstract:Sulfur-containing compounds are commonly present in both natural and synthetic bioactive molecules. The ability to construct C−S bond using inexpensive and easily accessible raw materials under mild conditions is an area of significant interest in organic synthesis. In this study, we introduce a novel photo-induced iron-catalyzed strategy that facilitates the transformation of alkyl and heteroaryl carboxylic acids into their respective thioether products. Furthermore, our method also enables the site-selective synthesis of thioethers and sulfoxides from unreactive hydrocarbons using N2 or air reaction atmosphere. The procedure is operationally simple and readily scalable, and provides access to high-value sulfur-containing compounds from simple raw materials in one step.

Abstract:Odorless 2-(1,3-dithian-2-ylidene)-3-oxobutanoic acid (1c) was prepared and investigated in the thioacetalization of carbonyl compounds as a 1,3-propanedithiol equivalent. The results showed that the thioacetalization of various carbonyl compounds 2 with 1c proceeded smoothly and afforded the corresponding dithioacetals 3 in high yields (up to 99%) in the presence of acetyl chloride at room or reflux temperatures. Moreover, the thioacetalization exhibited high chemoselectivity between aldehydes and ketones. Abstract:This paper presents a novel direct conversion of cyclohexanone to caprolactam catalyzed by trifluoroacetic acid. High conversion of cyclohexanone (100%) and high selectivity to caprolactam (above 99%) have been observed using acetonitrile as the additive. In addition, the effects of solvent, temperature, and catalyst composition on the reaction rate and selectivity to caprolactam were investigated. It is found that water could greatly decrease the reaction rate, whereas HCl could increase the reaction rate. On the basis of the results, the possible reaction route has been provided. This is the first example of an organocatalyzed one-step transformation of ketoxime into the corresponding amide, which may lead to the synthesis of caprolactam via a shorter and more environmentally friendly route.

Abstract:Thioesters are a common class of biologically active fragments, and synthetically useful building blocks. An attractive synthetic approach would be to use simple and bench-stable carboxylic acids as a coupling partner. Herein, we present a 4-bromo pyridine-borane complex as a catalyst for direct coupling of carboxylic acids with thiols. A wide range of thioesters with good functional group compatibility could be prepared via this metal-free approach. The merit of this strategy is exemplified by the modification of carboxylic acid-containing drugs.

Abstract:A novel and controllable synthesis of thioacetals/thioketals and β-sulfanyl ketones mediated by the reaction of aldehyde/acetone with thiols has been developed. In this protocol, β-sulfanyl ketones can be generated without the prior preparation of α, β-unsaturated carbonyl compounds. A variety of thiols reacted with aldehyde/acetone and provided the corresponding thioacetals/thioketals and β-sulfanyl ketones in good to excellent yields, respectively. This protocol is operationally simple, mild, and atom-economical, providing controllable access to thioacetals/thioketals and thia-Michael addition products under mild conditions.

Abstract:A nickel‐catalyzed reductive cross coupling reaction of carboxylic acids with disulfides is described. The readily available and inexpensive carboxylic acid is activated in situ by the formation of a mixed anhydride. The transformation occurs at room temperature with wide functional group tolerance to efficiently generate structurally diverse thioesters. Moreover, the reaction tolerated a broad substrate scope, including sterically hindered of disulfides. Ultimately, this powerful method could be applied to the late‐stage functionalization for the construction of diverse thioesters, affording a convenient and practical tactic to construct carbon‐sulfur bonds.

Abstract:2-[2-Chloro-1-(1-chlorovinyl)allylidene]-1,3-dithiane 1 was synthesized by the chlorination of 3-(1,3)-dithianylidenepentane-2,4-dione 2 using the Vilsmeier-Haack reagent in 99% yield. As a novel nonthiolic, odorless 1,3-propanedithiol equivalent, 1 was investigated in the thioacetalization reaction. Various types of aldehydes and ketones 3 were converted to the corresponding dithianes 4 in the presence of 1 in high yields (79-97%). Moreover, 1 exhibited obvious chemoselectivity between aldehyde and ketone in this thioacetalization reaction. A mechanism for this thioacetalization reaction is proposed.

Abstract:Thioesters are important in synthesis, materials science, and biology, and their preparation traditionally relies on the use of disagreeable thiols. Here, we report a thiol-free protocol that stitches together widespread carboxylic acids and aryl halides, producing a diverse array of thioesters. Crucial to this strategy is the discovery that tetramethylthiourea can serve as both a sulfur source and, upon direct excitation by purple light, as a strong reductant, suitable for activating aryl halides via single-electron transfer. Coupling of the resulting aryl radicals provides an isothiouronium ion intermediate, which can be attacked by carboxylic acids via a polar pathway, affording the thioester products under mild conditions.

Abstract:An electrophotochemical dual metal-catalyzed protocol for decarboxylative arylation of simple aliphatic carboxylic acids with aryl halides is reported. The relative stabilities of catalytically active metal complexes simultaneously generated at anode and cathode are the key design elements for the success of this convergent paired electrolysis. This new electrophotocatalytic method is mild, robust, and most importantly, capable of accommodating simple primary aliphatic acids including acetic acid - ubiquitous and variegated structural motifs yet remain oddly challenging substrates - directly as native functional groups for decarboxylative C(sp2 )-C(sp3 ) bond formation.

Abstract:An efficient three-step method has been developed for enantioselective synthesis of chiral alpha-aminoxy acids from aldehydes. In this method, chiral propargylic alcohols are obtained by asymmetric addition of terminal alkynes to aldehydes. The hydroxyl group then converted to aminoxy group via Mitsunobu reaction and oxidative cleavage of the internal carbon-carbon triple bond produce the carboxylic acid group. This represents a convenient approach to the general asymmetric synthesis of alpha-aminoxy acids with the advantages of substantial overall yields (37-73%) and high enantioselectivities (81-99% ee).
