Exploration of Drug Resistance Mechanism with EZ-Tn5 Transposome Insertion in Bacterial DNA

Abstract:OBJECTIVE To investigate the genetic factors in the bacteria affecting the drug resistance.METHODS EZ-Tn5 transposome was used as a tool to create the genomic library by randomly inserting the transposomes into the genome of BL21.Disk diffusion susceptibility was used to test all the acquired strains to define their inhibition zones.The inverse-PCR method was applied and its products were subsequently sequenced to define the exact insert site.RESULTS The result of the optimum conditions for electroporation of E.coli bacteria was OD value of 0.8(600nm),10% glycerol buffer and 2.7v voltage electric shock.Tn5 was inserted into a total of 182 transposon clones,which composed of the E.coli strain BL21 random insertion mutation library.One strain in the library was picked out and showed a constant change in the resistance to penicillin,cephazolin,cefaclor,and cefprozil,among which the resistance,to penicillin was the most obvious.Its inhibition zone diameter of 12mm from the control strains expand to 17mm.cated at 1736,the reverse PCR products were sequenced and the result showed that the transposon insertion sites lo6 sites in the bacterial genome,which inactivated the expression for the glycerol-3-phosphate acyltransferase.CONCLUSION Inactivation of the gene coding Sn-glycerol-3-phosphase acyltransferase in the E.coil genome can change its resistance to hydrophilic antibiotics like penicillin.It possibly suggests that this gene can influence the resistance of E.coli to hydrophilic antibiotics.
