Abstract:A catalytic method was developed to synthesize primary arylamines from the corresponding aryl bromides and iodides under mild conditions (yields=80–99%). Crystalline 2,2,2-trifluoroacetamide was used as ammonia surrogate and CuI/N,N′-dimethyl ethylenediamine was used as catalyst to achieve the C–N cross-coupling.

Abstract:The coupling reaction of aryl halides with aqueous ammonia was catalyzed by copper/oxalohydrazide/ketone system in water to yield primary arylamines without inert atmosphere. This method was fast and facile. The coupling reaction proceeded at 90 °C for 20–80 min, or at room temperature for a prolonged reaction time. A variety of aryl bromides and iodides were found to be applicable to this three-component catalytic system, which afforded good to excellent isolated yields.

Abstract:An efficient approach for the synthesis of N -arylated amides was developed via copper(II) triflate-catalyzed direct oxidation of (aryl)methylamines to primary arylamides by air and subsequent N -arylation by diaryliodonium salts. Various substituted benzylamines could be applied in the reaction, providing a series of N -arylated amides in moderate to good yields. This method showed convenient, practical, and environment friendly advantages.

Abstract:A novel methodology for the primary amination of aromatic C(sp2)-H bond utilizing hydroxylamine derivative as ammonia surrogates has been developed. With a cost-effective and convenient copper oxide, a variety of aromatic primary amines were synthesized without external oxidant with moderate to good yields under the mild conditions. This amination exhibits excellent functional group tolerance, wide substrate scope, the use of readily available substrates, and a facile work-up procedure. Furthermore, the late-stage modification of drug-like small molecules with complex structures was also allowed.

Abstract:In this report, a highly efficient method for the room temperature installation of alkyl amino motifs onto the ortho position of anilines via Cu-catalyzed carboxamide-directed amination with alkylamines is described. This method offers a practical solution for the rapid synthesis of complex arylamines from simple starting materials and enables new planning strategies for the construction of arylamine-containing pharmacophores. A single electron transfer (SET)-mediated mechanism is proposed.

Abstract:Ullmann-type C-N coupling reactions represent an important alternative to well-established Pd-catalyzed approaches due to the differing reactivity and the lower cost of Cu. While the design of anionic Cu ligands, particularly those by Ma, has enabled the coupling of various classes of aryl halides and alkyl amines, most methods require conditions that can limit their utility on complex substrates. Herein, we disclose the development of anionic N1,N2-diarylbenzene-1,2-diamine ligands that promote the Cu-catalyzed amination of aryl bromides under mild conditions. Guided by DFT calculations, these ligands were designed to (1) increase the electron density on Cu, thereby increasing the rate of oxidative addition of aryl bromides, and (2) stabilize the active anionic CuI complex via a π-interaction. Under optimized conditions, structurally diverse aryl and heteroaryl bromides and a broad range of alkyl amine nucleophiles, including pharmaceuticals bearing multiple functional groups, were efficiently coupled at room temperature. Combined computational and experimental studies support a mechanism of C-N bond formation that follows a catalytic cycle akin to the well-explored Pd-catalyzed variants. Modification of the ligand structure to include a naphthyl residue resulted in a lower energy barrier to oxidative addition, providing a 30-fold rate increase relative to what is seen with other ligands. Collectively, these results establish a new class of anionic ligands for Cu-catalyzed C-N couplings, which we anticipate may be extended to other Cu-catalyzed C-heteroatom and C-C bond-forming reactions.

Abstract:Biaryl amines are essential structural motifs prevalent in agrochemicals, pharmaceuticals, and materials. Herein, we present a novel copper-catalyzed method for aniline cross-couplings promoted by a 6-hydroxy picolinhydrazide ligand. The method achieves room-temperature reactivity with aryl bromides, enabled by a methanol/ethanol solvent mixture and a mild, functional group-compatible base, with catalyst loadings as low as 0.5 mol%. The use of industrially preferred bases and solvents, as well as the high catalytic activity, offers a significant advancement in the practicality and scalability of industrial processes. Furthermore, the approach extends to the cross-coupling of aryl chlorides under elevated temperatures and demonstrates compatibility with additional nucleophile classes.

Abstract:Anilines are ubiquitous in bio‐active compounds and their synthesis can be achieved via metal‐catalyzed cross‐coupling reactions involving aryl halides. We describe an unusual, yet simple, CuII‐catalyzed system for the amination of aryl chlorides in pure aqueous ammonia with 2.5 mol% catalyst loading under non‐inert conditions. Different from previous systems, the reaction proceeds even without an additional organic solvent. Copper(II) sulfate in combination with 4,7‐dimethoxy‐1,10‐phenanthroline enabled the amination of several aryl chlorides containing electron‐neutral, ‐donating and ‐withdrawing groups to the corresponding anilines with good to excellent yields. The upscaling potential of the procedure has been shown by the synthesis at 50 mmol scale. The reaction proceeds as one of the rare cases of a CuII‐assisted coupling, in contrast to the typical CuI―CuIII intermediates postulated for most Ullmann‐type coupling reactions. The copper(II) center allows for a nucleophilic substitution pathway, enabled by the deprotonation of coordinated ammonia.

Abstract:A simple and efficient protocol was developed for the preparation of challenging α-aryl primary amides. This metal-free coupling process was triggered by TfOH-promoted electrophilic activation of α-silyl nitrile to generate keteniminium ion species, followed by reaction with aryl sulfoxide through [3,3]-sigmatrophic rearrangement to provide the target product. To the best of our knowledge, α-silyl nitrile has been rarely used as a pro-electrophilic reagent. Computational investigations confirmed the transient existence of a highly electrophilic keteniminium intermediate.

Abstract:We describe a new strategy for aminoarylation of alkenes by copper-catalyzed smiles rearrangement using <i>O</i>-benzoylhydroxylamines as the amine reagent. This method affords various β-amino amide derivatives possessing a quaternary carbon center with wide functional group tolerance and high regioselectivity. The mechanistic studies indicate that the transformation can involve aminyl radical intermediates under acid-free condition.
