ANALYSIS OF PROTEIN EXPRESSION IN Arabidopsis MUTANT Athspr LEAVES SUBJECTED TO HEAT SHOCK

Abstract:Total proteins from leaves of Arabidopsis wild type and mutant Athspr, subjected to heat shock for 0, 2 and 8 h, were isolated, using optimized methods of two-dimensional gel electrophoresis. The maps were analyzed with solftware ImageMaster, and more than 1000 spots were detected respectively, and exhibited obvious differences between mutant and wild type. The molecular weights of most proteins were distributed in 14.1~116.0 kDa, and their isoelectric points were around pH 5~7. Three abundant differentially expressed protein spots between mutant and wild type plants were cut off, digested in gel with Trypsin, identified with MALDI-TOF-TOF-MS. Full amino acid sequences of proteins were obtained and identified through searching database with GPS-MASCOT. One of them is Glycine-rich RNA-binding protein 7 (GRP 7) , the other is Cytochrome b6-f complex iron-sulfur subunit, and the third is Oxygen-evolving enhancer protein 1-2, (OEE1). All of them were related with plant resistance to stress, suggesting that T-DNA insertion could influence the functional activity of the gene in the mutant, alter the expressions and functions of those three proteins, and then lead to the alterations of phenotypes including decline of resistance to stress in the mutant.
