Abstract:AIM: To culture in vitro umbilical cord blood mesenchymal stem cells(UCB-MSCs) by two modified methods and to observe their osteogenic differentiation potential. METHODS: The experiment was conducted at the Cell Laboratory of Bone and Joint Center in Shanghai the Ninth People's Hospital Affiliated to Medical College of Shanghai Jiao Tong University from May to September 2006. ①The 28 cord blood samples were provided by Hospital of Gynaecology and Obstetrics Affiliated to Medical College, Fudan University. Ratified by Medical Ethics Committee, full-term healthy UCB samples were obtained with consent from mothers and their family members. ②The 28 cord blood samples were collected, 50-90 mL each, and anticoagulated with citric acid. Mononuclear cells (MNCs) were separated by density gradient centrifugation 12 hours after collection, seeded in 100 mm×20 mm culture dish at a concentration of 1×1010 L-1 and suspended in cell culture medium comprising α-minimum essential medium (α-MEM) supplemented with 0.1 volume fraction fetal bovine serum (FBS). The medium was half changed after 5-7 days' primary culture and then was totally changed at a 3-4 days' interval. ③When the adherence of MNCs happened, the subsequent management was taken according to modified methods. The suspension in Group 1 (n =10) was removed to a new plate when round megakaryocytes reached confluence and fibroblastoid cells detached. The medium in Group 2 (n =18) was changed into α-MEM supplemented with 0.15 volume fraction FBS when the round megakaryocytes were gradually prominated, then the medium was changed into α-MEM supplemented with 0.1 volume fraction FBS when most of the round megakaryocytes detached. Reaching approximately 80% to 90% confluence, the fibroblastoid cells were detached with trypsin and replated at 1∶2 or 1∶3 under the same culture conditions. ④The morphology was observed under microscope. Flow cytometry was used to examine the surface antigen phenotype of the fifth passage cells, and alkaline phosphatase method was applied to detect osteogenic differentiation ability. RESULTS: ①Adherent cells were obtained from 20 of 28 UCB samples (Group 1, 6 samples; Group 2, 14 samples), of which fibroblastoid cells from 13 samples were passaged steadily (Group 1, 4 samples; Group 2, 9 samples) with the success rate of 46.4%. ②There were two types of adherent cells after 5 to 7 days primary culture, fibroblast-like cells and round megakaryocyte. Fibroblastoid cell colonies were found after 5 weeks primary culture, showing homogeneous fusiform shape, morphologically similar to bone marrow MSCs, which could be passaged steadily to 22 passages without obvious differences. ③UCB-MSCs were all strongly positive for MSC-related antigens such as CD29 and CD105. Instead, they were all negative for neither hematopoietic lineage markers such as CD34 and CD45 nor endothelial markers such as CD106. ④Incubation of UCB-MSCs under the osteogenic condition for 1 week resulted in osteoplasts and was positive by alkaline phosphatase staining. CONCLUSION: MNCs in cord blood after modified culture can elevate the successful rate of UCB-MSCs culture. UCB-MSCs are similar with MSCs derived from other sources and can be amplified in vitro easily, passaged steadily and differentiated dramatically into osteoblast.
