Association of the Interaction between C-reactive Protein Gene Polymorphism and Environmental Factors with the Ischemic Stroke in Chinese Han Subjects

Abstract:Objective To investigate the association between two single nucleotide polymorphism(SNP) of CRP and ischemic stroke(IS) and its subtypes in order to further analyze the correlation between gene polymorphisms,interaction of environmental factors and IS.Methods Case-control study was adopted in this clinical study.159 IS patients admitted to the department of neurology of Aerospace Center Hospital from October 2010 to December 2011 were selected as IS group.175 healthy people with matched age and sex were selected as the control group.CRP gene polymorphisms(rs1800947 and rs3093059) were genotyped subsequently using polymerase chain reaction-restrictive fragment length polymorphism(PCR-RFLP) and DNA sequencing.Multiple Logistic regression analysis was used to explore the association of genotype rs1800947 and rs3093059 with IS and its subtypes,and interaction between environmental risk factors and gene polymorphism was explored by dichotomy analysis.Results The GG + GC genotype of rs1800947 was statistically different between patients in IS and control groups 〔OR = 3.456,95% CI(1.350,8.845),P = 0.010〕.Frequency of G and C allele of rs1800947 was statistically different between patients in IS and control groups 〔OR = 3.069,95% CI(1.237,7.612),P = 0.016 〕.In addition,the CC + GC genotypes of rs1800947 was associated with an increased risk of small-artery occlusion by subtype analysis of ischemic stroke 〔OR = 6.225,95% CI(1.739,22.291),P = 0.005〕.In gene-environmental interaction analysis,elevation of ischemic stroke risk was observed among GC genotype carriers who consume alcohol,smoke cigarette,with adjusted combined ORs of 6.667 〔95% CI(1.234,36.013) 〕and 11.110 〔95% CI(1.229,100.454) 〕respectively,compared with low-risk individuals for rs1800947 GG carriers who did not consume alcohol,smoke cigarette.Conclusion The data indicate that the rs1800947 SNP in the CRP is involved in the development of ischemic stroke and small-artery occlusion in the Chinese population.Synergism was also observed between genotype and smoking and drinking risk factors.
