Application of fluorescence in situ hybridization in analyzing endothelial chimerism in renal allograft

Abstract:Objective To find out whether endothelial chimerism is related to graft rejection.Methods Biopsy samples were studied from 34 renal transplants of female recipients who received the graft from a male donor for the detection of endothelial cells of recipient origin.Formalin-fixed,paraffin-embedded tissue sections of renal samples were examined by fluorescence in situ hybridization for the detection of endothelial cells containing two X-chromosomes using a biotinylated Y-chromosome probe and digoxigenin labeled X-chromosome probe.Results The FISH methods did identify endothelial cells of recipient origin.Endothelial chimerism was common irrespective of rejections(P>0.05).Its presence was focal and they coexisted closely with that of the recipient in the biopsy.Conclusion There was no correlation between the percentage of recipient endothelial cells in vascular endothelial cells and the type of graft rejection.
