The Exploration of Green Tea Polyphenols on Vascular Cognitive Impairment and Its Mechanisms

Abstract:Objective To explore the effect of green tea polyphenols(GTP) on vascular cognitive impairment in rats with chronic cerebral hypoperfusion.Methods The male Wistar rats were randomly divided into four groups:Sham operation group,model group,various concentration GTP group administered with GTP 100 mg/(kg·d) or GTP 400 mg/(kg·d).The chronic cerebral hypoperfusion models were established by permanent bilateral occlusion of common carotid arteries.After model was established,drugs were administered from the fourth week.Spatial learning and memory abilities were evaluated by Morris water maze after the drug were given for 4 weeks.The levels of malondialdehyde(MDA),the activities of superoxide dismutase(SOD) and acetylcholine esterase(AchE) in the hippocampus were measured biochemically.The mRNA levels of NF-E2-related factor 2(Nrf2) in the hippocampus were examined by real-time quantitative polymerase chain reaction(RT-PCR).Results The escape latencies in GTP groups were shorten,and the time spent in the platform were longer than that of chronic cerebral hypoperfusion in the probe trials.The levels of MDA were decreased in GTP groups,the activity of SOD were enhanced,but had no significant effects on the activities of AchE.The mRNA levels of Nrf2 were up-regulated in 400 mg/(kg·d)GTP group.Conclusion GTP could lessen the lipid peroxidation,enhance the antioxidative effects of hippocampus after chronic cerebral hypoperfusion,especially by dosage of 400 mg/(kg·d).But GTP had no effects on cholinergic system.
