Protective Effects of Iex-1 on Ischemia/Reperfusion-Induced Cardiac Injury in the Rats

Abstract:IEX-1 (immediate early response gene X-1) is a stress-inducible gene and is involved in cell apoptosis and cell death. Previous studies have reported that cell apoptosis is the major cause of ischemia/reperfusion (I/R)-induced cardiac injury. The ischemia precondition (IPC) significantly reduces such injury. However, there is no study showing whether or not IEX-1 is involved in IPC or cardiac cell apoptosis. In the present study, we set up the cell IPC and I/R model based on neonatal rat cardiomyocytes, and constructed the adenoviruses expressed human IEX-1s (Ad-IEX-1s). The results showed that IPC could increase IEX-1 expression both in mRNA and protein levels. Its mRNA elevation reached the peak at 30 min after IPC. IEX-1 expression was obviously increased in IPC (10 min) plus I/R (2 h/3 h) compared with I/R alone. In addition, over expression of IEX-1 by Ad-IEX-1s in neonatal rat cardiomyocytes significantly reduced I/R-induced reactive oxygen species (ROS) formation, procaspase-3 activation and LDH release (67.2% decrease vs. Ad-GFP) in these cells. Furthermore, overexpression of IEX-1 in rat heart also diminished cardiac infarct size from 31.25% (Ad-GFP) to 11.25%(Ad-IEX-1) in rat I/R (0.5 h/24 h) model. Our study demonstrates for the first time that IPC can increase IEX-1 expression which might protect cardiac cell against I/R-induced injury by reducing intracellular ROS formation, cell apoptosis and cell death. *This work was supported by grants from NSFC (30500198) and “973” (2006CB503802).
