Abstract:Conjugated linoleic acid (CLA) is a collective term for the octadecadienoic acid isomers containing conjugated double bonds. This article reviewed CLA isomers from biological activities, biosynthesis mechanisms and analytical methods. The biological activities of CLA isomers in anti-obesity, cardiovascular protection, diabetes management and anti-cancer in vitro and in vivo were mainly reviewed. More attention has been paid to the production of the specific CLA isomer due to its biological activity. The biosynthesis methods of CLA isomers, such as dietary modification in ruminants and fermentation by microorganisms & enzymes, were systematically introduced. A rapid, accurate and economic analysis method will promote the research in both biological activities and biosynthesis mechanisms of CLA isomers. The merits of UV spectrometry, GC, HPLC, MS and CE used in the analysis of CLA isomers were also compared in detail. This paper aims to put into perspective the current status and future trends on CLA isomers.
Analysis of Conjugated Linolenic Acid from Plant Seeds

Abstract:The objective was to determine the lipid classes and conjugated linolenic acid(CLNA) contents in Trichosanthes kirilowii,pomegranate and bitter melon seed oils from China.Three pomegranate cultivars(Sanbaiyu,Qingpiruanzi and Tianluzi) and six bitter melon cultivars(Changbai,Manyouqing,Bingchengyihao,Kaihua-1,Kaihua-2 and Qingfeng) were chosen for analysis.The predominant lipid was triacylglycerol in the analyzed samples ranging from 92.6% to 97.0%.The major fatty acids were oleic acid(22.6%,18:1),linoleic acid(32.6%,18:2n-6) and punicic acid(32.6%,c9,t11,c13-18:3) in T.kirilowii seed oil.The total CLNA was 36.9% in T.kirilowii seed oil.Punicic acid was the most predominant fatty acid,ranging from 73.4% to 77.5% and the total CLNA ranged from 83.4% to 87.9% in the seed oils of pomegranate cultivars.The total CLNA ranged from 65.2% to 71.9%,and α-eleostearic acid(c9,t11,t13-18:3) was predominant one,account from 56.0% to 63.7% in bitter melon seed oils.T.kirilowii,pomegranate and bitter melon were rich in CLNA,however,main CLNA in T.kirilowii and pomegranate was punicic acid,and one in bitter melon was α-eleostearic acid.Natural resources containing CLNA,especially edible T.kirilowii seed,could be a potential dietary source of CLA,following CLNA metabolism.
Identification of enriched conjugated linoleic acid isomers in cultures of ruminal microorganisms after dosing with 1-(13)C-linoleic acid

Abstract:Most studies of linoleic acid biohydrogenation propose that it converts to stearic acid through the production of cis-9 trans-11 CLA and trans-11 C18:1. However, several other CLA have been identified in ruminai contents, suggesting additional pathways may exist. To explore this possibility, this research investigated the linoleic acid biohydrogenation pathway to identify CLA isomers in cultures of ruminai microorganisms after dosing with a (13)C stable isotope. The (13)C enrichment was calculated as [(M+1/M)×100] in labeled minus unlabeled cultures. After 48 h incubation, significant (13)C enrichment was observed in seven CLA isomers, indicating their formation from linoleic acid. All enriched CLA isomers had double bonds in either the 9,11 or 10,12 position except for trans-9 cis-11 CLA. The cis-9 trans-11 CLA exhibited the highest enrichment (30.65%), followed by enrichments from 21.06 to 23.08% for trans-10 cis-12, cis-10 trans-12, trans-9 trans-11, and trans-10 trans-12 CLA. The remaining two CLA (cis-9 cis-11 and cis-10 cis-12 CLA) exhibited enrichments of 18.38 and 19.29%, respectively. The results of this study verified the formation of cis-9 trans-11 and trans-10 cis-12 CLA isomers from linoleic acid biohydrogenation. An additional five CLA isomers also contained carbons originating from linoleic acid, indicating that pathways of linoleic acid biohydrogenation are more complex than previously described.
Re-characterization of three conjugated linolenic acid isomers by GC-MS and NMR.

Abstract:Conjugated linolenic acids (CLN) refer to a group of octadecatrienoic acids with three conjugated double bonds. Minor positional and geometrical differences among CLN isomers make their separation and identification difficult. We have used GC-MS and NMR to study three common CLN isomers namely alpha-eleostearic acid, beta-eleostearic acid and punicic acid, finding that some signals of olefinic carbon atoms in NMR spectra were mistakenly assigned in the literature. The present study was therefore undertaken to re-characterize the location of CC double bonds and assign the chemical signals of proton and carbon atoms using (1)H NMR, (13)C NMR, (1)H-(1)H two-dimensional correlation spectra ((1)H-(1)H COSY) and (13)C-(1)H two-dimensional correlation spectra ((13)C-(1)H COSY). The geometrical structure of double bonds in these three CLN isomers was identified using homonuclear decoupling technique.
Bioconversion of conjugated linoleic acid by Lactobacillus plantarum CGMCC8198 supplemented with Acer truncatum bunge seeds oil

Abstract:Conjugated linoleic acid (CLA) isomers, c9, t11-CLA and t10, c12-CLA, have been proved to exhibit excellent biomedical properties for potential use in anti-cancer applications and in reducing obesity. Acer truncatum Bunge (ATB), which is rich in unsaturated fatty acids, including oleic acid, linoleic acid, and nervonic acid, is a new resource for edible oil. In the present study, we developed a new method for producing two CLA isomers from ATB-seed oil by fermentation using Lactobacillus plantarum CGMCC8198 (LP8198), a novel probiotics strain. Polymerase chain reaction results showed that there was a conserved linoleate isomerase (LIase) gene in LP8198, and its transcription could be induced by ATB-seed oil. Analyses by gas chromatography-mass spectrometry showed that the concentration of c9, t11-CLA and t10, c12-CLA in ATB-seed oil could be increased by about 9- and 2.25-fold, respectively, after being fermented by LP8198.

Abstract:Fatty acids containing conjugated carbon-carbon double bonds (C═Cs), such as conjugated linoleic acids (CLAs), attract growing research interest due to their bioactivities against diabetes, cancer, and atherosclerosis. Analysis of conjugated fatty acid (CFA) is challenging for existing analytical techniques because it requires determination of geometry (cis ( Z) vs trans ( E)) and location of individual C═C. In this study, we developed a method to achieve confident, fast, and quantitative analysis of CFA isomers from mixtures. This method combines the strength of trapped ion mobility spectrometry (TIMS) for fast isomer separation and the Paternò-Büchi (PB) reaction followed by tandem mass spectrometry (MS/MS) for C═C location determination. Notably, the PB reaction of CFA is regioselective to terminal C═Cs, thus forming diagnostic fragment ions unique to conjugated C═Cs from PB-MS/MS. These fragment ions facilitate identification and quantitation of individual CLA isomers differing in C═C locations, affording limit of identification of 1 nM. Given that PB-MS/MS alone cannot identify the geometry of C═C, TIMS has been employed for characterizing C═C geometry. TIMS is capable to separate various C═C geometric isomers of CLAs, allowing visualization of C═C isomerization during the PB reaction. By coupling the PB-MS/MS with TIMS, two CLA isomers, CLA 18:2(9 Z,11 E) (46.9 ± 1.1%) and CLA 18:2(10 E,12 Z) (53.1 ± 1.1%), are quantified in a commercial CLA supplement.

Abstract:Conjugated linoleic acids (CLA) are positional and geometric isomers of linoleic acid involved in a number of health aspects. In humans, CLA production is performed by gut microbiota, including some species of potential probiotic bifidobacteria. 128 strains of 31 Bifidobacterium species were screened with a spectrophotometric assay to identify novel CLA producers. Most species were nonproducers, while producers belonged to B. breve and B. pseudocatenulatum . GC-MS revealed that CLA producer strains yielded 9 cis ,11 trans -CLA and 9 trans ,11 trans -CLA, without any production of other isomers. Hydroxylated forms of LA were absent in producer strains, suggesting that the myosin-cross-reactive antigen (MCRA) protein that exerts hydratase activity is not involved in LA isomerization. Moreover, both CLA producer and nonproducer species bear a MCRA homologue. The strain B. breve WC 0421 was the best CLA producer, converting LA into 68.8% 9 cis ,11 trans -CLA and 25.1% 9 trans ,11 trans -CLA. Production occurred mostly during the lag and the exponential phase. For the first time, production and incorporation of CLA in biomass were assessed. B. breve WC 0421 stored CLA in the form of free fatty acids, without changing the composition of the esterified fatty acids, which mainly occurred in the plasmatic membrane.

Abstract:Conjugated linoleic acid (CLA) is a class of naturally occurring octadecadienoic acid in humans and animals and is a general term for a group of conformational and positional isomers of linoleic acid. In order to obtain the development of excellent lactic acid strains with a high production of conjugated linoleic acid, 32 strains with a possible CLA conversion ability were obtained by initial screening using UV spectrophotometry, and then the strains were re-screened by gas chromatography, and finally, the strain with the highest CLA content was obtained. The strains were optimized for cultivation by changing the amount of substrate addition, inoculum amount, and fermentation time. The results showed that the yield of the experimentally optimized strain for the conversion of conjugated linoleic acid could reach 94.68 ± 3.57 μg/mL, which was 74.4% higher than the initial yield of 54.28 ± 2.12 μg/mL of the strain. The results of this study can provide some basis for the application of conjugated linoleic acid production by Lactobacillus paracasei in the fermentation of lactic acid bacteria.

Abstract:Conjugated linoleic acid is a mixture of positional and geometric isomers of linoleic acid with two conjugated unsaturated double bonds at various carbon positions in the fatty acid chain, mainly found in the milk and meat of ruminants. 2 of them (c-9,t-11 CLA and t-10,c-12 CLA) with physiological importance were reviewed in the pathways of biosynthesis. Understanding the mechanisms involved in the biosynthesis of CLA and their regulation pathways will direct us to design suitable strategies to get more CLA via synthesis biology methods.

Abstract:For the purpose of setting up a simple and effective analysis method to determine the quantity of c9,t11-CLA,applied GC-FID determination,used CP-Sil 88(60 m × 0.25 mm i.d ×0.20 μm)gas chromatography column,used normalization method which with adjusted factor to analyze.Results showed that c9,t11-CLA can be separated very well,average retrieve rate was 97.7% and RSD was 1.08%.
Screening and Identification of Lactic Acid Bacteria for Biosynthesis of Conjugated Linoleic Acid

Abstract:A strain ZS2058 was selected from the strains separated from fermented vegetables and raw milk for biosynthesis of conjugated linoleic acid. The strain was identified as Lactobacillus plantarum by API system. The strain could tolerate at 11.6%(1.024 mg/mL) LA and converte LA to CLA in MRS broth. The cis-9, trans-11 and trans-10, cis-12 isomers detected by Gas Chromatography were 75.9% and 24.1 % of the CLA produced.

Abstract:Two strains that can transform linoleic acid to conjugated linoleic acid were screened from soil.The morphology,growth curves and the optimum conditions for CLA production in MRS medium were studied in this paper.Though the two strains were different significantly in morphology,the optimum conditions for CLA production of them were similar: concentration of linoleic acid was 0.51%(v/v),and incubation time was 36～48 h.

Abstract:Conjugated linoleic acid (CLA) is a mixture of positional and geometric isomers, and numerous biological functions such as anticancer, reduction of body fat content, diabetes prevention, lower blood lipids and inhibition of arteriosclerosis are due to individual or synergistic action of most isomers including c9t11- and t10c12-CLA. Because of different positions of double bounds between c9t11- and t10c12-CLA isomers, high oxidization possibility of c9t11-CLA isomer was observed. Therefore, c9t11-CLA isomer exhibited a major responsibility for anticarcinogenic effect, whereas, t10c12-CLA isomer exhibited stronger reduction effect on body fat and blood lipids. Increasing studies suggested that the mechanisms of both isomers on cancer and fat metabolism were different.
Enzymatic fractionation of conjugated linoleic acid isomers to get functional monomers

Abstract:The two functional monomers of conjugated linoleic acid (CLA) was fractionated by Lipase AYS-catalyzed esterification of methanol and CLA.The process of esterification was optimized by single factor experiment as follows:Lipase AYS amount 180 U/g,reaction temperature 40 ℃,buffer pH 6.5,molar ratio of CLA to methanol 1 ∶ 1.After reaction for 8 h under the optimal conditions,the total esterification rate was 49.5％.After separation of methyl ester and fatty acid from reaction mixture,the fractionation efficiency of c9,t11-CLA in methyl ester phase and t10,c12-CLA in fatty acid phase were 86.2％ and 80.0％,and the recovery rates of the two monomers were 72.4％ and 54.2％,respectively.
Reasons for the differences in biotransformation of conjugated linoleic acid by Lactobacillus plantarum

Abstract:Conjugated linoleic acid (CLA) has attracted a great deal of attention for its functions in weight loss, regulation of metabolism, and antioxidant capabilities. Many microorganisms, including rumen bacteria, propionic acid bacilli, and Lactobacillus, have CLA biotransformation ability. The CLA production capability of different species is different, as are those different strains of the same species. However, the reasons for this discrepancy remain unclear. In this study, 14 strains of Lactobacillus plantarum were found, through gas chromatography-mass spectrometry analysis, to be capable of converting linoleic acid to CLA. The transcriptional levels of CLA-related genes in the high- (AR195, WCFS1, and AR488) and low-yield strains (AR176, AR269, and AR611) were analyzed using real-time quantitative PCR. The transcriptional levels of cla-hy, cla-dh, and cla-dc in AR195 were the lowest in the exponential phase, but it had the highest CLA yield. Correlation analysis showed no correlation between CLA yield and the transcription level of these genes in the exponential phase. The results showed that a high transcriptional level in the exponential phase of cla-hy, cla-dh, and cla-dc did not necessarily lead to high CLA production. Investigation of the transcription level in different growth phases showed that the CLA biotransformation abilities of Lactobacillus plantarum strains significantly depended on the transcriptional maintenance of cla-hy, cla-dh, and cla-dc. We observed a correlation between CLA production and increased levels of cla-hy transcription, but a prerequisite is needed: the transcription of cla-dh and cla-dc should be upregulated and maintained a high transcriptional level during the platform period. This study provides a new strategy for screening high CLA-producing strains. It also lays a theoretical foundation for regulating CLA biotransformation and increasing the yield of CLA.
Bio-Molecular analysis of selected food derived Lactiplantibacillus strains for CLA production reveals possibly a complex mechanism

Abstract:Lactiplantibacillus is among the most extensively studied bacterial specie belonging to the genus Lactobacillus with proven probiotic and health promoting effects. These beneficial effects are generally strains specific but the underlying molecular mechanisms are still not fully understood. Dissecting the determinants behind probiotic topographies of this bacterium is of particular interest since it would help select strains that stand the best chance of success in clinical trials and potential industrial applications. In the current study, we have compared the oleate hydratase phylogeny of seven selected strains of L. plantarum on the basis of their sequence, physiochemical properties and 3D structures. All the strains were assessed on molecular level visualization to find out the active site residues which take part in binding with linoleic acid (LA) at the time of interactions. From the whole genome sequences, the genes responsible for conjugated linoleic acid (CLA) production were identified by Venn diagrams. Identified genes were further compared phylogenetically by MEGA X and physiochemical parameters were analyzed by utilizing ExPaSy-Protparam. The genes were further analyzed for the secondary structures using PSIPRED and tertiary structure was generated by trRosseta and SWISS-MODEL. For the analysis of molecular interactions, LA was used as a ligand and the docking was performed using AutoDock Vina. The phylogenetic analysis showed a close phylogeny of the strains with publicly available genomes. The best interaction energy with LA was observed as -6.7 kcal/mol. The bacteria perform an important role in the CLA production through LA metabolism. Oleate hydratase genes are involved in the complex mechanism of the saturated conversion of LA in to CLA. The current study provides further insights for CLA production by different strains of L. plantarum. There is an excellent opportunity for future studies to investigate different CLA production mechanisms in different bacterial strains.

Abstract:Abstract Conjugated Linoleic Acid (CLA) has attracted the attention of many researchers, especially that of microbial origin due to its biological importance to the consumer. The current study aims to extract LA Isomerase enzyme from Lactobacillus paracasei bacteria from milk and to use the enzyme in the production of CLA. Selective media, including MRS and MRS-Dagatose, were used in isolating local strains. The selected bacterial isolates were tested for their ability to produce LA-Isomerase enzyme. The isolate with high enzymatic activity was selected. After extraction and partial purification of the enzyme, the optimal conditions for the production of conjugated fatty acid were studied, and the reaction products were diagnosed using GC-MS technology. It was found that 11 isolates have the ability to produce CLA at different concentrations, H1 isolate showed the highest production of conjugated fatty acid at a concentration of 120.45 g.ml-1, this isolate was selected as the source for enzyme extraction. The enzymatic activity of the crude extract and partially purified with ammonium sulfate was estimated using color methods at wavelength of 233 nm. The effect of the optimum conditions (pH, temperature, linoleic acid concentration and enzyme concentration) on the CLA product was studied using the partially purified LA Isomerase enzyme, the optimum conditions for production were 6.5, 45 °C, 100 μg.ml-1 and 0.7 ml, respectively. The GC-MS technique showed the presence of a number of reaction products that are isomers of conjugated linoleic acid (C9T11, T9T12, T10C12) with different concentrations.

Abstract:This study was performed to characterize natural CLnA isomer production by Bifidobacterium breve LMC520 of human origin in comparison to conjugated linoleic acid (CLA) production. B. breve LMC520 was found to be highly active in terms of CLnA production, of which the major portion was identified as cis-9,trans-11,cis-15 CLnA isomer by GC-MS and NMR analysis. B. breve LMC520 was incubated for 48 h using MRS medium (containing 0.05% L-cysteine · HCl) under different environmental conditions such as atmosphere, pH, and substrate concentration. The high conversion rate of α-linolenic acid (α-LNA) to CLnA (99%) was retained up to 2 mM α-LNA, and the production was proportionally increased nearly 7-fold with 8 mM by the 6 h of incubation under anaerobic conditions at a wide range of pH values (between 5 and 9). When α-LNA was compared with linoleic acid (LA) as a substrate for isomerization by B. breve LMC520, the conversion of α-LNA was higher than that of LA. These results demonstrated that specific CLnA isomer could be produced through active bacterial conversion at an optimized condition. Because many conjugated octadecatrienoic acids in nature are shown to play many positive roles, the noble isomer found in this study has potential as a functional source.

Abstract:The term Conjugated Linoleic Acid (CLA) refers generically to a class of positional and geometric conjugated dienoic isomers of linoleic acid. Among the isomers of linoleic acid cis9, trans11-CLA (c9, t11-CLA) and trans10, cis12-CLA (t10, c12-CLA) are found to be biologically active isomers, and they occur naturally in milk, dairy products and meat from ruminants. In addition, some vegetables and some seafoods have also been reported to contain CLA. Although the CLA levels in these natural sources are insufficient to confer the essential health benefits, anti-carcinogenic or anti-cancer effects are of current interest. In the rumen, CLA is an intermediate of isomerization and the biohydrogenation process of linoleic acid to stearic acid conducted by ruminal microorganisms. In addition to rumen bacteria, some other bacteria, such as Propionibacterium, Bifidobacterium and some lactic acid bacteria (LAB) are also capable of producing CLA. In this regard, Lactiplantibacillus plantarum (formerly Lactobacillus plantarum) has demonstrated the ability to produce CLA isomers from linoleic acid by multiple enzymatic activities, including hydration, dehydration, and isomerization. L. plantarum is one of the most versatile species of LAB and the bacterium is widely used in the food industry as a microbial food culture. Thus, in this review we critically analyzed the literature produced in the last ten years with the aim to highlight the potentiality as well as the optimal conditions for CLA production by L. plantarum. Evidence was provided suggesting that the use of appropriate strains of L. plantarum, as a starter or additional culture in the production of some fermented foods, can be considered a critical factor in the design of new CLA-enriched functional foods.

Abstract:A lactic acid bacterial strain,lp15,having the ability to synthesize conjugated linoleic acid(CLA) was isolated from pickle juice samples.The strain lp15 was identified as Lactobacillus plantarum based on full-length 16S rRNA sequence analysis and API system.This strain was treated by ultraviolet and DES to obtain several mutant strains,which exhibited 29.3%-52.2% enhancement for CLA production.The strain lp15-2-1 exhibited the highest CLA productivity,which was up to 30.13 μg/mL after 48 h culture in the MRS medium added with LA at 0.2 mg/mL level.According to gas chromatographic analysis,cis9,trans11-CLA was the predominant compound in the fermentation product,with a relative content of up to 76.5%.trans10,cis12-CLA approximately accounted for 23.5% of the fermentation product.
